Biochemical and histochemical studies on the phosphatase activities of the mucous membrane of the frog tongue were carried out. The biochemical study strongly suggested that the mucous membrane of the tongue contains both ATPase and alkaline phosphatase activities. But, since the ATPase activity was dependent on the pH of the incubating solution and the maximal activity was obtained around pH 9.0, the ATPase activity may be partly due to the alkaline phosphatase activity. The histochemical study revealed that ATPase activity was localized on the superficial layer of the tongue. Great activity was exhibited in the sensory discs of the fungiform papillae and in the ciliated cell layer of the back of the tongue. Alkaline phosphatase activity could not be demonstrated histochemically in the present study.
