[Author abstract]A simple method for separation and determination of synthetic water-soluble colors was investigated by means of agar column chromatography. (1) Qualitative examination: An agar powder was swollen with a Mcllvaine's buffer solution (pH 5.0), and poured into a chromatographic glass tube (400×7mm.) up to a height of about 250mm.. After the column was thoroughly washed with the Mcllvaine's buffer solution, a solution of 0.2 mg. of a color in water was applied to the top of hte column bed. The color was then developed with a Mcllvaine's buffer solution (pH 5.0), and the R-value of the color was determined which was expressed as the ratio of two distances, namely, the traveled distance of the color band and that of eluant surface. It proved possible to qualitatively analyze synthetic water-soluble colors by means of determining their R-values only if their protein and sodium chloride contents were not more than 0.1%. (2) Quantitative examination: A mixture of 12 colors, containing 0.2 or 0.5mg. of each color, was applied to the top of the column bed, and the colors were developed with McIlvaine's buffer solutions (pH 4.4 and pH 5.0). The column was cut to fractions including each color, when the fastest one reached the bottom of the column,,and the recovery percentages were determined by measuring the absorbances of the extracted colors from each fraction. As the results, the recovery percentages of not less than 90% were obtained for all colors except for Blue No. 2 and Red No. 106. (3) Assay of officially permitted synthetic food colors in the presence or absence of sucrose, citric acid and sodium chloride was studied, and these substances proved inert on the assay results.[著者抄録]寒天カラムクロマトグラフィーにより、水溶性タール色素の分離分析を試みた。1.定性試験は0.7×40cmのガラス管カラムへ、マクイルパインクエン酸緩衝液に膨潤させた寒天を充塡し、同緩衝液で洗浄後、カラム頂部に色素を吸着させ、同緩衝液で各色素を展開する。展開液が約10cm移動したときのR値を測定して定性を行なった結果、色素液中、タンパク質、食塩濃度が0.1%以内であれば、本法での定性が可能であった。2.定量試験は、カラムベッド70cmのカラムに、タール色素1種を試料として吸着させた後、定性試験の場合と同様に操作し、色素を抽出後、吸光度測定より、回収率を求めた結果、青色2号、赤色106号を除いては、90%以上の回収率を示した。3.共存物質の影響を見るために、各色素液にショ糖とクエン酸の混合溶液および5%食塩水を添加したもののクロマトグラフィーを行ない、吸光度より回収率の検討を行なったが、なんらの影響も認められなかった。
