Here, we report the recovery of cell nuclei from 14,000-15,000 years old mammoth tissues and the injection of those nuclei into mouse enucleated matured oocytes by somatic cell nuclear transfer (SCNT). From both skin and muscle tissues, cell nucleus-like structures were successfully recovered. Those nuclei were then injected into enucleated oocytes and more than half of the oocytes were able to survive. Injected nuclei were not taken apart and remained its nuclear structure. Those oocytes did not show disappearance of nuclear membrane or premature chromosome condensation (PCC) at 1 hour after injection and did not form pronuclear-like structures at 7 hours after injection. As half of the oocytes injected with nuclei derived from frozen-thawed mouse bone marrow cells were able to form pronuclear-like structures, it might be possible to promote the cell cycle of nuclei from ancient animal tissues by suitable pre-treatment in SCNT. A 25-year-old female California sea lion (Zalophus californianus) reared in an aquarium died following a history of anorexia, lethargy, abnormal protrusion of the skin, and oral respiration. At necropsy, multiple yellowish-white nodules with diameters of 0.1–0.5 cm were disseminated in the thoracic cavity and lungs. Histopathologically, the nodules were continuous with normal mesothelium and were characterized by the proliferation of spindle-shaped to polygonal neoplastic cells with prominent atypia. The neoplastic cells exhibited diffuse, strong staining for vimentin and partial, weak to moderate staining for cytokeratin AE1/AE3. Based on these findings, the lesions were diagnosed as pleural mesothelioma. This study reports the first case of pleural mesothelioma in California sea lion.

This paper describes a novel statistical bandwidth extension (BWE) technique based on a Gaussian mixture model (GMM) and a sub-band basis spectrum model (SBM), in which each dimensional component represents a specific acoustic space in the frequency domain. The proposed method can achieve the BWE from speech data with an arbitrary frequency bandwidth whereas the conventional methods perform the conversion from fixed narrow-band data. In the proposed method, we train a GMM with SBM parameters extracted from full-band spectra in advance. According to the bandwidth of input signal, the trained GMM is reconstructed to the GMM of the joint probability density between low-band SBM and high-band SBM components. Then high-band SBM components are estimated from low-band SBM components of the input signal based on the reconstructed GMM. Finally, BWE is achieved by adding the spectra decoded from estimated high-band SBM components to the ones of the input signal. To construct the full-band signal from the narrow-band one, we apply this method to log-amplitude spectra and aperiodic components. Objective and subjective evaluation results show that the proposed method extends the bandwidth of speech data robustly for the log-amplitude spectra. Experimental results also indicate that the aperiodic component extracted from the upsampled narrow-band signal realizes the same performance as the restored and the full-band aperiodic components in the proposed method.
