We demonstrated that LPA is synthesized in endometrium and there is higher LPA concentration and mRNA expression for LPA1 in bovine endometrium during pregnancy than estrous cycle. Thus LPA may contribute to early pregnancy establishment. In <I>in vivo</I> study we investigated LPA effect on conception rate, P4 and PG secretion in heifers. Animals were inseminated 72 h after second PGF2α injection. From Day 15 to 18 after insemination, heifers were treated intravaginally with: saline (control), LPA (1 mg) or VPC32183 (1 mg; n = 8 for each group). Blood samples were collected on the following days after insemination: 0, 6, 12, 15, 16, 17, 18, 21 and 30. Pregnancy was confirmed by USG and <I>per rectum</I> examination on days 30 and 49. LPA administration resulted in increase in P4 and PGE2 concentrations compared to saline and VPC32183 - treated heifers (P<0.05). In saline and LPA-treated groups 6 out of 8 heifers were pregnant, whereas conception rate in VPC32183 – treated heifers was 37% (P<0.05). In <I>in vitro</I> study we examined the effects of LPA on PG secretion and <I>PGES</I> and <I>PGFS</I> mRNA expressions in stromal and epithelial cells of bovine endometrium on Days 16-18 of pregnancy and estrous cycle. LPA increased PGE2 production and <I>PGES</I> mRNA expression in stromal cells at estrous cycle and pregnancy (P<0.05). The overall results indicate that LPA serves as luteotropic factor during early pregnancy stimulating P4 and PGE2 secretion through activation of <I>PGES</I> mRNA transcript expression in stromal cells. Moreover, at pregnancy establishment, endogenous LPA protects CL and embryo development.
