For efficient use of sunlight, the light-harvesting core antenna (LH1) and the reaction center (RC) form a supramolecular complex (LH1-RC) in photosynthetic bacteria. We determined the crystal structure of a LH1-RC complex from a purple bacterium, Thermochromatium tepidum. The structure reveals that the RC is completely surrounded by 16 αβ-heterodimers. Calcium ions are bound at the periplasmic side of LH1. 32 bacteriochlorophyll and 16 spirilloxanthin molecules form an elliptical assembly in LH1. The pigment geometry involved in the absorption characteristics and excitation energy transfer is determined. In addition, ubiquinone channels and lipid molecules are observed in the complex.

The final goal of this research project is the understanding of all fundamental biological phenomena in terms of physical chemistry. As a model organism for the structural and functional studies, an extremely thermophilic bacterium, Thermus thermophilus HB8, is very promising because of the small genome size, the availability of genetic tools for functional analysis, and the thermostability of its proteins (http: //www.thermus.org/) . In this report, we summarize the recent progress of this research project toward the systems biology.

The crystal structure of 8-HDF type photolyase from Anacystis nidulans showed the similarity of the backbone structure with MTHF type E. coli photolyase but completely different binding site of the light-harvesting cofactor. This is a first example that homologous primary and tertiary structures in closely related proteins recognize two different types of cofactors at different binding-sites. The structure and function of photolyase and its cofactor recoginition are reviewed and discussed.
