Cells are exposed to several kinds of stimulation and some types of mechanical stimulations are known to affect the corresponding cell, but the transduction mechanism has long been a black box. Though there are results that second messengers, like Ca2+, are mechanosensitive, it is unclear which receptor reacts to mechanical stimulation. We used ATDC5, chondrocytes derived from mice, to survey the mechanosensitive receptors against hydrostatic pressure (HP). Fura2 AM was used for imaging the intracellular Ca2+ intensity. It was observed by EM-CCD camera (HAMAMATSU), under the HP ranged from 0 to 40MPa. We found that some cells under high ordered HP, raised their Ca2+ intensity slightly in the period of less than 3 minutes. From this experiment, we showed the possibility that chondrocytes could be mechanosensitive against high HP. For further study, we could use inhibitors for each possible Ca2+ receptors and strive to find the gate of mechanotransduction.
