Mitochondrial dysfunction due to oxygen deficiency was studied in the systems of in vivo and in vitro using rat livers, and the following results were obtained The decrease in oxidative phosphorylation in mitochondria exposed to oxygen deficiency in vitro was improved by treating them with membrane stabilizing agents such as chlorpromazine and cepharanthine. The oxygen deficiency induced membrane deterioration involved in barrier function and the sensitivity to lipid peroxidation of the mernbranes. The agents protected the membranes from the above lesions without giving any effects on adenine nucleotide degradation in the absence of oxygen. Previous intravenous injection of cepharanthine (0.4mg/100g body weight) protected the mitochondria from the dysfunctions due to hepatic ischemia. From these results, in addition to our previous results, the mechanism of mitochondrial dysfunction due to oxygen deficiency was discussed.
