1. A new assay method of chemotherapeutic agents, a vertical diffusion assay method at low temperature (15°C) using Serratia marcescens as the test organism, was developed. S. marcescens No. 33 strain, has suitable biological characteristics for this new method. The microbiological activity of bacteriostatic agents such as sulfa drugs is successfully evaluated by this method. The method can also be applied widely for the assay of antibiotics which are active against Gram-negative bacilli.2. S. marcescens No. 33 strain was found to grow anaerobically under certain culture conditions using a new synthetic medium and protein-free medium modified from the experimental results of QUASTIL and STEPHENSON. This method is very useful for the assay of chemotherapeutic agents which are adsorbed onto protein.3. In this method, amount of the samples to be assayed can be minimized by using a gauze, sterilized and impregnated with the sample solution. The amount of the sample solution required is less than 0.1 ml.
