Ticks are known to transmit various pathogens, radically threatening humans and animals. Despite the close contact between ticks and viruses, our understanding on their interaction and biology is still lacking. The aim of this study was to experimentally assess the interaction between canine parvovirus (CPV) and a widely distributed hard tick, Haemaphysalis longicornis, in laboratory condition. After inoculation of CPV into the hemocoel of the ticks, polymerase chain reaction (PCR) assay revealed that CPV persisted in inoculated unfed adult female ticks for 28 days. Canine parvovirus was recovered from the inoculated ticks using a cell culture, indicating that the virus retained intact in the ticks after inoculation, but significant positive reaction indicating virus infection was not detected in the tick organs by immunofluorescence antibody test using a monoclonal antibody. In the case of ticks inoculated with feline leukemia virus, the virus had shorter persistence in the ticks compared to CPV. These findings provide significant important information on the characteristic interaction of tick with non-tick-borne virus.

Canine distemper virus (CDV) causes a lethal disease among members of the Carnivora. To clarify the distribution of CDV in wild animals, we examined 106 raccoon sera collected from two prefectures in Japan, Hyogo and Osaka, from 2005 to 2007. Among them, 34 raccoons (32.1%) possessed a virus-neutralizing (VN) antibody to KDK-1 strain (genotype Asia-1). There was no significant difference in seroprevalence of CDV regardless of places, gender, and body weights. In Hyogo, a geometric mean of VN titers to KDK-1 was significantly higher than that to Onderstepoort (vaccine strain), indicating that KDK-1-like CDV different from vaccine strain might have spread among raccoon population in Hyogo. In conclusion, CDV is epidemic among feral raccoons in Japan, suggesting that CDV might have been spreading among Japanese wild animals.

Canine parvovirus type 2 (CPV) is a virulent pathogen that emerged in the late 1970s, probably originating from feline panleukopenia virus (FPLV) or a closely related carnivore parvovirus belonging to the feline parvovirus (FPV) subspecies. In contrast to FPLV, CPV has evolved rapidly since its emergence. The original antigenic type of CPV disappeared more than two decades ago and several new antigenic as well as genetic CPV variants have appeared and spread in the field. Both high mutation rate and positive selection of mutations in the capsid gene appear to be the driving force for such rapid evolution. In addition, genetic recombination has been assessed as a factor in parvovirus evolution. Recently, we provided the first evidence of inter-antigenic type recombination of CPV in nature. Here, an inter-FPV subspecies recombinant was revealed by analyzing the genetic data deposited in databases with several recombination detection programs, and by phylogeny. FPLV strain XJ-1, submitted by Su et al., Harbin, China in 2007 (GenBank accession no. EF988660), was most likely generated by recombination between CPV and FPLV. Its genome was generally composed of the NS1 gene of CPV origin and the VP1 gene of FPLV origin. This is the first demonstration of recombination between different FPV subspecies in nature. Consequently, recombination should be considered as an element in the generation and evolution of parvoviruses of the FPV subspecies.

Reovirus infection is common in mammals. However, seroepidemiological data of reovirus neutralizing antibodies are limited in dogs. In this study, sera of 65 healthy dogs from six prefectures across Japan were tested for neutralizing antibodies against reovirus serotype 1 strain Lang (T1L), serotype 2 strain Amy (T2A) and serotype 3 strain Dearing (T3D) using plaque reduction neutralization test (PRNT). Seropositivity against reovirus T1L, T2A and T3D was 53.85%, 33.85% and 46.15%, respectively. Distribution of reovirus seropositive samples displayed no distinguishable geographical pattern. However, reovirus seropositivity increases with age and in dogs housed outdoor. Co-infection of multiple reovirus serotypes in dogs is also possible. These data will provide valuable insights towards the usage of reovirus in oncolytic virotherapy in canine cancers.
