Familial aggregation and twin studies have suggested that genetic factors play a significant role in susceptibility to eating disorders such as anorexia nervosa (AN) and bulimia nervosa(BN). A sib-pair linkage study suggested a linkage region for restricting AN in chromosome 1 and that for BN in chromosome 10. Candidate-gene association studies indicated associations of SLC6A4, HTR2A and BDNF genes with AN in multiple studies. We found that ghrelin precursor gene SNPs and their haplotype were associated with purging type BN. In addition, the same gene SNPs were significantly associated with increased body weight, body mass index, fat mass, waist circumference, thickness of skinfolds, elevated fasting acylated ghrelin concentration, decreased HDL-cholesterol concentration and elevated scores in the Drive for Thinness-Body Dissatisfaction in non-clinical young female. A genome-wide association study using microsatellite markers is in progress in Japan. Studying susceptibility genes for eating disorders are expected to lead preventions and development of new treatments.
