Fluoride is a natural component of the biosphere, the 13th most abundant element in the earth’s crust. Fluoride has been known to play an important role in mineralization of bone and teeth, and can be therapeutically used at low doses for dental care and prevention or at high doses for the treatment of osteoporosis. Particularly in cases of the development of a fluoride compound as a proprietary drug, liberated fluoride ions may bring the risk of causing dental or skeletal fluorosis in animals treated with high doses of fluoride drugs in toxicity studies. However, there are limited data on changes in fluoride levels in hard tissues of the body over animal life spans. In the present work, we obtained plasma, teeth (incisors and molars), bones (alveolar, femur and tibia) and nails from SD rats at 8, 11, 20, 33, 46, 59 and 72 weeks of age and determined fluoride levels individually. Fluoride accumulated time-dependently in bones, nails and molars in a similar manner, with fluoride levels increasing 2-3 folds from 8 to 72 weeks of age. In contrast, fluoride levels in plasma and incisors, which grow continuously in living rats, showed almost constant values. These data can be used not only as a historical database for the effective evaluation of data from toxicology studies, but also as a contribution to biological characterization of SD rats.
