Purpose: Mucinous adenocarcinoma (MA) is associated with worse clinicopathological characteristics and a poorer prognosis than non-MA. Moreover, MA is related to worse tumor regression grade and tumor downstaging than non-MA. This study investigated whether lesions in MA and non-MA can be quantitatively assessed by T2 mapping technique and compared with the diffusion-weighted imaging (DWI).Methods: High-resolution MRI, DWI, and T2 mapping were performed on 81 patients diagnosed with rectal cancer via biopsy. Afterward, T2 and apparent diffusion coefficient (ADC) values were manually measured by a senior and a junior radiologist independently. By examining surgical specimens, the patients with MA and non-MA were identified. Inter-observer reproducibility was tested, and T2 and ADC values were compared using Mann–Whitney U test. Finally, receiver operating characteristic (ROC) curves were drawn to determine the cut-off value.Results: Of the 81 patients, 11 patients with MA were confirmed by pathology. The inter-observer reproducibility of T2 and ADC values showed an excellent intraclass correlation coefficient (ICC) of 0.993 and 0.913, respectively. MA had higher T2 (87.9 ± 5.11 ms) (P = 0.000) and ADC (2.03 × 10−3 mm2/s) (P = 0.000) values than non-MA (66.6 ± 6.86 ms and 1.17 × 10−3 mm2/s, respectively). The area under the ROC curves (AUC) of the T2 and ADC values were 0.999 (95% confidence interval [CI]: 0.953–1) and 0.979 (95% CI: 0.920–0.998), respectively. When the cutoff value in T2 mapping was 80 ms, the Youden index was the largest, sensitivity was 100%, and specificity was 97%.Conclusion: As a stable quantitative sequence, T2 mapping of MRI is useful in differentiating MA from non-MA. Compared to ADC values, T2 values are also diagnostically effective and non-inferior to ADC values.

Calorie restriction (CR) by 30-40% decreases morbidity of age-related diseases and prolongs the lifespan of various laboratory animal species. Taurine (2-aminoethanesulfonic acid) is an important nutrient for lipid metabolism as it conjugates bile acids. Here, we investigated how taurine supplementation induces effects similar to the CR beneficial effects. Sprague Dawley rats were fed a diet containing different taurine concentrations (0, 0.5, 1.0, 3.0, 5.0%) to analyze the effects on growth, blood, and hepatic parameters. Rats fed a 5% taurine-supplemented diet showed a significant decrease in visceral fat weight, compared with control rats. Moreover, there were significant decreases in the serum total cholesterol, hepatic cholesterol and triglyceride concentrations in the taurine-supplemented groups compared with the control group in a dose-dependent manner. These results were associated with decreased mRNA expression of fatty acid synthase, and increased mRNA expression of carnitine palmitoyltransferase 1α. C57BL/6 mice were fed a 5.0% taurine-supplemented diet, and their response to 3-nitropropionic acid-induced oxidative stress was analyzed. The rate of weight loss due to oxidative stress decreased and the survival rate significantly increased in the taurine-supplemented groups compared with the control group. Finally, cells were treated with 100 μM taurine and their resistance to UV-induced oxidative stress was analyzed. We found that the p53-Chk1 pathway was less activated in taurine-treated cells compared with control cells. Furthermore, damage to cells evaluated by oxidative stress indicators revealed a reduction in oxidative damage with taurine treatment. These findings suggest that taurine partially acts as a CR mimetic.

Integrins, transmembrane molecules that facilitate cell-to-cell and cell-to-extracellular matrix interactions, are heterodimers that consist of an α- and β-subunit. The integrin α4 gene (itgα4) is expressed in various type of cells and tissues. Its biochemical functions and physiological roles have been revealed using cultured cell assays. In contrast, the primary effect caused by itgα4 deletion on vertebrate development is poorly understood, because knockout mice exhibit multiple defects that can lead to embryonic lethality in the uterus. Zebrafish are a convenient vertebrate model to investigate morphogenesis during embryogenesis, because of their external fertilization and subsequent development outside the female's body. Here, we generated a zebrafish mutant line named itgα4 ko108 using the CRISPR/Cas9 genome editing system; the mutant genome harbored an approximately 2.0-kb deletion in the itgα4 locus. A truncated transcript was detected in itgα4 (+/-) or (-/-) fish but not in (+/+) fish. The mutant transcript was hypothesized to encode a truncated Itgα4 protein due to a premature stop codon. itgα4 (-/-) embryos obtained from the mating of heterozygous parents exhibited no apparent phenotype during development at 24 hours post-fertilization (hpf). However, approximately half of them exhibited cephalic hemorrhage at 48 hpf. The incidence ratio was significantly higher than that in (+/+) or (+/-) embryos. Embryonic hemorrhage has also been reported previously in Itgα4 knockout mice. In contrast, embryonic lethality with the other defects reported in the knockout mice was not observed in our zebrafish model. Therefore, the mutant line itgα4 ko108 should be a useful model to investigate a physiological function for Itgα4 in the blood circulation system.

More than 80 years ago, McCay and colleagues first reported that limiting the amount of food provided to experimental animals (i.e. calorie restriction or CR) prolongs their lifespan and suppresses the onset and progression of various age-related diseases. Today, CR remains the most reliable method of delaying aging in experimental animals, and research into its underlying molecular mechanisms is ongoing. CR has been reported to have anti-aging and life-extension effects on primates, with progress being made toward applications for humans. Studies on mechanisms underlying the onset and prevention of lifestyle-related diseases such as diabetes have elucidated the cellular signaling pathways that regulate energy metabolism, and commonalities have been discovered between the targets of existing diabetes drugs and the signaling pathways affected by CR. This finding has led to research into the discovery of drugs that have the anti-aging effects of CR in the absence of food intake limitations, namely CR mimetics (CRM). Several drugs have been reported to extend the lifespan of experimental organisms, which may thus have the potential to also extend human lifespan. In this article, we outline and compare those drugs that have been reported to date and discuss the possibility of taurine as a CRM, which is a topic of our ongoing research.
