In the present study, Streptomyces sp. CW1 was isolated from a constructed wetland system mesocosm and identified as such based on 16S rDNA analysis, and additional biochemical properties were evaluated. This isolate was found to be halotolerant (up to 11% NaCl) and able to grow and utilize biopolymers such as: agarose, agar, gellan gum (a bacterial polysaccharide), polypectate and chitin as the sole carbon and energy source. Since the primary isolation was performed based on its capability to degrade phenol, the high cell yield coefficient showed rapid growth on phenol (0.82–0.98 mg dry biomass/mg phenol). Its halotolerance and the capability to biodegrade biopolymers found mainly in the marine environments, suggest on its primordial oceanic origin. In the present study some of its characteristics are described and discussed along with its beneficial use for wastewater purification processes in constructed wetland systems.

(Using colony direct PCR we established, kan gene (GenBank accession No. AB028210) coding for an aminoglycoside acetyltransferase, AAC(3), was surveyed among strains of Streptomyces griseus and its related taxa (S. anulatus group). The regulatory and ORF regions of kan gene were targeted by the use of original primers. With the regulatory region primers, the target fragment (518 bp) amplification was observed in all the streptomycin (SM)-producing strains of S. griseus and in several strains of S. anulatus group. The other target fragment (930 bp) amplification using the ORF primers was observed only in the SM-producing strains of S. griseus including ‘S. ornatus’ that has substantially identical properties to those of S. griseus. It was thus conclusive that kan gene is highly conserved in SM-producing strains of S. griseus and distributed in taxa related to S. griseus. There were numbers of strains that provided no amplification upon PCR but hybridization bands upon Southern hybridization, suggesting the lack or alteration of kan gene. Characterization by colony direct PCR using appropriate primers will be a big help for taxonomic as well as ecological characterization of actinomycetes.

In this study, we investigated the taxonomic diversity of actinomycetes isolated from swine manure compost that had undergone self-heating. Pretreatment with sodium dodecyl sulfate and subsequent cultivation of the compost sample on humic acid-vitamin agar at 30 and 50°C resulted in the selective isolation of 125 actinomycete strains. These isolates were classified using a combination of morphological characterization and 16S rDNA sequence analysis, with sequence similarities of ≥97% and ≥99% used to identify isolates at genus and species level, respectively. The isolated actinomycete assemblage consisted of a diverse array of species, with the 125 isolates falling into 10 families, 15 genera, and 22 species. The most dominant thermophilic actinomycetes were Saccharomonospora (Sam.) viridis and Thermobifida fusca, followed by Actinomadura spp. Other species that were frequently isolated at 30°C included Glycomyces spp., Nocardiopsis (Nop.) alba, Nop. composta, Rhodococcus rhodochrous, and Sam. azurea.

pSN22, an 11 kb multicopy conjugative plasmid from Streptomyces nigrifaciens, promotes chromosome recombination in Streptomyces lividans. Five genes have been identified to be involved in plasmid transfer and pock formation: traB is essential for plasmid transfer; traA for pock formation; spdA and spdB are concerned with pock size; and traR, which corresponds to a kor gene in a kil-kor system, encodes a repressor of traR itself and the traA-traB-spdB (tra) operon. Studies on the interaction of TraR with promoter regions suggest that the negative regulation of transfer-related genes by TraR is achieved by two mechanisms, i.e. promoter hiding and roadblock. The predicted ATPase activity and the membrane localization of TraB suggest that the protein plays a direct role in ATP-driven DNA translocation. TraB is also thought to be involved in intra- and intermycelial transfers of pSN22.
