SDS-PAGE is essential for protein experiment in many laboratories. SDS-PAGE gel is constituted by separating and stacking gels. For the preparation of SDS-PAGE gel, the reagent mixture of separating gel is added between glass plates and then, usually, water is overlaid. After the polymerization of the separating gel, the water was removed and the reagent mixture of stacking gel is added. It was suggested from this study that the overlaid water was not essential for the preparation of SDS-PAGE gel.
