Alpha-fetoprotein (AFP) is an important disease biomarker, relating to cancers such as hepatocarcinomas and gastric cancer. However, traditional methods are time-consuming, relied on bulky instruments and trained professionals, cannot satisfy the demand for low cost and point-of-care testing (POCT). In this study, a power-free POCT device was developed for the rapid and low-cost detection of AFP via one-sampling. Based on the principle of sandwich immunofluorescence, the chip is capable of automatically accomplishing on-chip mixing, labeling and capturing procedures, which only require that operator add 40 μL sample into the chip one time. The proposed device is capable of sensitively detecting human AFP in FBS with a dynamic range of 10 – 1000 ng/mL and LOD (1.88 ng/mL) within a short time of 3 min. Predictably, our method holds a great potential to be applied in the POC diagnostics of proteins, especially for some regions that are resource-limited.

Enterohemorrhagic Escherichia coli are a dangerous bacterium known to be harmful to the human body, with some infections even resulting in death. Given this danger, food factories are required to perform a quick bacterial test to confirm the absence of this pathogen prior to shipping. We have developed a novel molecular imprinting polymer (MIP) particle that has encapsulated gold nanoparticles (AuNPs) and which can function as both a receptor and an optical signal transmitter in biological systems. This MIP particle is artificially synthesized and can be engineered to specifically recognize and capture antigens on the bacterial cell membrane. In addition, MIP particles containing AuNPs generate strong scattered light signals, and binding of the MIP particles improves the optical intensity of the target bacterial cells. This enables clear visualization under a dark-field microscope and quantification of the target bacteria using the scattering light intensity. Here we describe the successful quantification of Escherichia coli O157 cells in real meat samples using this technology in conjunction with a simple labelling step.

The creaming behavior of an oil-in-water (O/W) emulsion was quantitatively evaluated via resonance-enhanced multiphoton ionization time-of-flight mass spectrometry. Styrene O/W emulsions were prepared with initial styrene concentrations of 1 and 4 g/L, and the height at the center of the sample was monitored. A peak area of the molecular ion of styrene was set as the signal intensity, for which a time profile was constructed from a series of mass spectra. As a result, the averaged time profiles showed that the signal intensities increased once and then decreased with the onset of creaming. In addition, in order to fit an experimentally obtained time profile, a modified fit function was proposed. Based on the fit results, the ratios of the increases and decreases in signal intensities were different between the two emulsions—higher in the case of an O/W emulsion with a higher initial oil concentration. On the other hand, the duration of the enhancement of the signal intensity with the onset of creaming was independent of the initial oil concentration. The present method offers the possibility to quantitatively evaluate the creaming behavior of an emulsion without pretreatment, and, therefore, would be useful for confirming the stability and quality assurance of emulsions.

Chemical modification of proteins is important for creating a myriad of engineered proteins and for elucidating the function and dynamics of proteins in live cells. A wide variety of chemical protein modification methods have been developed and can be categorized into three classes: (i) modification of proteins using the reactivity of naturally occurring amino acids; (ii) modification by bioorthogonal reactions using unnatural amino acids, most of which can be site-selectively incorporated into proteins-of-interest using genetic codon expansion techniques; and (iii) recognition driven chemical modification, which is the only approach that allows modification of endogenous proteins without any genetic manipulation even under heavily crowded and multi-molecular conditions, as in live cells and organisms. All of these approaches have merits and limitations. In this review, we summarize these approaches and discuss their characteristics with respect to specificity, reaction rate and versatility.

In this study, nanocomposites of ligand-free gold nanoparticles that are anchored onto the graphene surface (Graphene/AuNPs) were synthesized by a sonochemical method in a single reaction step. A highly sensitive amperometric sensor using Graphene/AuNPs is proposed for the determination of trace hexavalent chromium Cr(VI) in environmental water samples. Compared with a gold electrode, a glassy carbon electrode and a AuNPs modified glassy carbon electrode, the Graphene/AuNPs modified glassy carbon electrode exhibits the highest electrocatalytic activity and stability towards the reduction of Cr(VI), based on the results by cyclic voltammetry and electrochemical AC impedance studies. This study shows that the Graphene/AuNPs-based sensor can detect Cr(VI) with a low detection limit of 10 nM (∼0.5 μg/L), a wide dynamic range of 0 to 20 μM (R = 0.999) and very good selectivity and reproducibility. The electrode is applied to the determination of Cr(VI) in river samples with satisfactory recovery values.

A near-infrared (NIR) and water-soluble probe was synthesized and studied for the detection of Cys/Hcy in aqueous solution, living cells and mice. The probe was composed of cyanine derivative as the NIR fluorescent reporting unit and pyrimidiny-thioether moiety as the Cys/Hcy responsive unit. Treatment with Cys/Hcy induced the formation of sulfur-substituted products, then intramolecular rearrangement reaction would occur to produce amino-substituted products and resulting in enhanced red fluorescence emissions. It could be applied to sense Cys/Hcy both in solution with the detection limit of 0.17 μM (or 0.32 μM) and in living cells. Cell imaging experiments proved that such a probe exhibited good cell penetration. In addition, the probe could detect Cys/Hcy in live mice with strong turn-on fluorescent response.

Capillary electrophoretic methods have been developed for the analysis of Monascus yellow pigments for food additives. Monascus yellow pigments were successfully separated by micellar electrokinetic chromatography (MEKC) with a 20mM sodium dodecyl sulfate (SDS) solution containing 50mM phosphate buffer, pH 7.0, and 20% acetonitorile or a 2.0% butyl acrylate/butyl methacrylate/methacrylic acid copolymer sodium salts (BBMA) solution containing 20mM ammonium formate buffer, pH 7.0, and 10% methanol. The xanthomonasin A was identified with the molecular mass measured by FABMS. Monascus yellow pigments were extracted from candies with solid phase extraction and analyzed by the developed technique.

A novel organic-inorganic hybrid optical sensor (NUS) was designed and prepared in two steps: the grafting of 3-(isocyanatopropyl)trimethoxysilane onto the surface of SBA-15, and then the attachment of naphthalene-1-amine. The obtained materials were characterized using low-angle XRD, N2 adsorption-desorption, TGA, FT-IR, and TEM techniques. A fluorescence study revealed that the NUS was a highly selective optical sensor for the detection of Cr2O72− among various anions, including F−, I−, Cl−, Br−, CO32−, HCO3−, NO3−, H2PO4−, CH3COO−, and NO2− with a good linearity between the fluorescence intensity and the concentration of Cr2O72− as well as a detection limit of 1.2 × 10−7 M in a 100% aqueous medium. Moreover, the applicability of real samples of the sensor is also discussed.

Microfluidic paper-based analytical devices (μ-PADs) fabricated in Japan were employed for the determination of total chromium (Cr) in water, soil, and lettuce irrigated with wastewater in Ethiopia. The μ-PADs, which were printed by wax printing in Japan, were transported to Ethiopia and prepared for the determination of total Cr by adding appropriate reagents to the pretreatment and detection zones. Soil and lettuce samples were determined by the μ-PADs and a UV-Vis spectrophotometer in Ethiopia. A paired t-test showed that the mean total Cr concentrations determined in the soil and lettuce samples were not significantly different between μ-PADs and UV-Vis spectrophotometric analysis at the 5% level of significance. This implies that the μ-PADs have good accuracy and reliability, and could be employed to monitor Cr in environmental samples. We found that the total Cr concentrations in all soil and lettuce samples were above the permissible limit. Moreover, evaluating Cr contamination level using the geo-accumulation index indicated that the soils were contaminated with Cr moderately to heavily. Thus, the present work successfully demonstrated the potential of remote investigations of pollution in a less-equipped laboratory by transporting the μ-PADs fabricated in another laboratory.

Immunochromatography assay is an easy and rapid on-site detection method. However, conventional sandwich immunochromatographies using two antibodies can only detect target molecules above a threshold size. Small molecules below 1000 in molecular weight are usually detected using competitive immunoassay. However, competitive immunoassay is not suitable for visual detection of low concentration samples. Based on the principles of open sandwich immunoassay, which detects small molecules via interchain interaction of separated variable region fragments (VH and VL) from a single antibody, we developed non-competitive open sandwich immunochromatography. Bone Gla protein (BGP)-C7, a peptide containing the seven C-terminal amino acids of human osteocalcin, was selected as the target. By using VL fragments fixed on a nitrocellulose membrane, and colored cellulose bead-labeled VH fragments, we specifically detected 10 ng/mL of BGP-C7. This is the first report of open sandwich immunochromatography, which is an easy and rapid method for on-site, signal-on detection of small molecules.
