Point-of-care testing (POCT) of biomarkers, such as proteins and nucleic acids, is a hot topic in modern medical engineering toward the early diagnosis of various diseases including cancer. Although microfluidic chips show great promise as a new platform for POCT, external pumps and valves for driving those chips have hindered the realization of POCT on the chips. To eliminate the need for pumps and valves, a power-free microfluidic pumping method utilizing degassed poly(dimethylsiloxane) (PDMS) was invented in 2004. In this article, the working principle of the degas-driven power-free microfluidic chip is first described, and then applications of those chips to biomarker analysis are reviewed. The biomarker analysis on the chip was typically achieved with a small sample volume of ∼1 μL and a short analysis time of ∼20 min. For protein analysis, the sandwich immunoassay format was adopted. The limit of detection (LOD) was improved by three orders of magnitude by using laminar flow-assisted dendritic amplification (LFDA), which was a newly devised amplification method specialized for microfluidic chips. For analysis of nucleic acids such as DNA and microRNA, the sandwich hybridization format was adopted, and the LFDA was also effective to reduce the LOD. With the LFDA, typical LOD values for proteins and nucleic acids were both around 1 pM.

The elucidation of the mechanism for steel corrosion under a coating layer has been attracting research attention. Herein, we utilized a confocal micro-X-ray fluorescence (XRF) analytical instrument to conduct non-destructive elemental analysis near the surface of a steel sheet. Using this method, elemental map images of steel sheet cross sections were obtained without sample destruction. To confirm corrosion suppression in the presence of Mg ions, we observed the corrosion behavior of hot-dip galvanized steel sheets immersed in an aqueous NaCl solution to which Mg ions were added. By using the confocal micro XRF system, the elution of the coating components and the precipitation process of the corrosion products were confirmed.

Cell analysis is of great significance for the exploration of human diseases and health. However, there are not many techniques for high-throughput cell analysis in the simulated cell microenvironment. The high designability of the microfluidic chip enables multiple kinds of cells to be co-cultured on the chip, with other functions such as sample preprocessing and cell manipulation. Mass spectrometry (MS) can detect a large number of biomolecules without labelling. Therefore, the application of the microfluidic chip coupled with MS has represented a major branch of cell analysis over the past decades. Here, we concisely introduce various microfluidic devices coupled with MS used for cell analysis. The main functions of microfluidic devices are described first, followed by introductions of different interfaces with different types of MS. Then, their various applications in cell analysis are highlighted, with an emphasis on cell metabolism, drug screening, and signal transduction. Current limitations and prospective trends of microfluidics coupled with MS are discussed at the end.

The electrorotation (ROT) rates of K562 cells accompanied by erythroid differentiation were estimated to identify the differentiation status by using a novel electrorotation device with a microwell arranged on polynomial electrodes. Successive estimations of individual cells were achieved by sequential manipulations which involve trapping of the cell by positive dielectrophoresis (DEP), rotating by ROT, and removing by negative DEP. The ROT rate increased with the differentiation of K562 cells, because the cytoplasm conductivity would increase with an increase of the concentration of iron ions to produce hemoglobin. The ROT rate could be utilized to estimate the stage of cell differentiation without labeling.

A sensitive, useful and preliminary screening method was proposed to quantitate the containable cannabinoids most commonly included in mineral water and gummi candy products, specifically cannabidiol (CBD), cannabinol (CBN), 11-nor-9-carboxy-Δ9-tetrahydrocannabinol (THCA), cannabigerol (CBG), and cannabidiolic acid (CBDA), using liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS) for quality evaluations. Based on the electrospray positive ion mode, the limit of detection and the limit of quantification values were 0.2 to 0.5 ng/mL and 0.8 and 2.0 ng/mL. Samples (0.5 g) were diluted by water/methanol (50/50), to which stable isotope internal standards were added; the recovery results appeared in range from 91.3 to 101.2%. This method was applied to evaluate CBD products (6 kinds) from the Japanese market. Our survey found obvious discrepancies between the labeling and the results were overserved in products. In addition, CBN, THCA, CBG, and CBDA were not detected in full-spectrum products that contained various cannabinoids that naturally occur in the cannabis plant. Thus, it is necessary to be able to verify the accurate concentration and impurity in various CBD products from the Japanese market as quickly as possible.

We propose a novel concept for detecting color transition by the inner product (IP) of RGB unit vectors. A digital microscope-based detector and a Visual Basic program were developed in-house. The concept is applied to indicator-based flow titration. The IP is 1 or < 1 if the vector’s direction is the same or different, respectively. The IP’s change can be used as a criterion for the indicator’s color transition. The present IP-based approach is simple, economical, and versatile because it is applicable to any color transition without selecting an analytical wavelength.

We report on a simple, cost-effective, instrument-free, and portable distance-based paper device coupled with NFs for the determination of glucose. The analysis reaction is based upon the oxidative etching reaction of silver nanoparticles (AgNPs) in the presence of H2O2 that is produced from glucose after a glucose oxidase (GOx) catalytic reaction leading to a morphological transformation of AgNPs. A color band length of AgNPs is coated on to a detection channel and then etched by H2O2, and these were changed from a purple color to colorless as a correlate of the glucose concentration. To improve the performance of the enzyme immobilization, NFs, which are biocompatible without compromising their structure and biological activity, were then placed onto the sample zone. The naked-eye detection limit was 0.1 mM for 40 min of analysis time. The recoveries of glucose spiked in the artificial urine samples and control urine samples were then verified by our device and were in the acceptable range of 96 – 100%.

Enzyme-linked immunosorbent assay (ELISA) is a widespread analytical biochemistry assay. In this work, a direct ELISA method using a metallic nanoparticle (NP)-immobilized 96-well plate was developed for high-throughput, highly sensitive fluorescence analysis. Immobilization of metallic NPs on a 96-well plate effectively amplified fluorescence signals of the assay. The silver (Ag) NP-immobilized plate showed the best fluorescence enhancement effect of all the metal-immobilized plates tested. We used the Ag NP-immobilized plate to detect biomolecules and bacteria and found that both the fluorescence intensity and the limit of detection (LOD) were strongly enhanced by more than 100 times compared with those of the unmodified 96-well plates. Quantitative and qualitative considerations for target bacteria regarding the impact of autofluorescence on detection were successfully obtained for several strains. Our results demonstrate the potential of applying Ag NPs for enhancing the efficiency of direct and indirect ELISA assays.

Renal cell carcinoma is a very aggressive and often fatal disease for which there are no specific biomarkers found to date. The purpose of this work was to find features that differentiate urine metabolic profiles of healthy people and cancer patients. Laser desorption/ionization mass spectrometry on gold nanostructures-based techniques were used for the metabolic analysis of urine of 50 patients with kidney cancer. Comparison with data from 50 healthy volunteers led to the discovery of several compounds that may be considered potential renal cell carcinoma (RCC) biomarkers. Statistical analysis of data allowed for the discovery of m/z values that had the greatest impact on group differentiation. A database search enabled the assignment of signals for the most promising 15 features among them: serine, heptanol, 3-methylene-indolenine, 2-methyl-3-hydroxy-5-formylpyridine-4-carboxylate, phosphodimethylethanolamine, 4-methoxyphenylacetic acid, N-acetylglutamine, 3,5-dihydroxyphenylvaleric acid, hydroxyhexanoylglycine, valyl-leucine, leucyl-histidine, oleamide, 9,12,13-trihydroxyoctadecenoic acid, stearidonyl carnitine and squalene. Differences of metabolite profiles of human urine could be identified by gold nanoparticle-enhanced target (AuNPET) LDI MS method and used for the detection of renal cancer.

An acriflavine-graphene oxide (GAF) supramolecular assembly has been prepared from water-soluble graphene oxide (GO) and a fluorescent dye, acriflavine (AF). Upon binding this non-covalently to the GO, the fluorescence of acriflavine has been “turned off” effectively, competitive binding potential of the sensor substrates such as ATP, ADP, AMP and the pyrophosphate weakens the supramolecular assembly of GAF, which allows the release of acriflavine quantitatively, which also “turns-on” the fluorescence of the dye under UV irradiation. Interestingly, GAF displayed the highest sensitivity towards ATP within the family of adenosine phosphates. We have developed a naked eye detection method for the adenosine phosphates biomolecules. For the first time, acriflavine has been utilized for the sensing of adenosine phosphates in combination with GO, which can be useful for the detection of other biomolecules.

Cysteine (Cys), as one of the important amino acids, plays a vital role in various physiological and pathological processes. Hence, it is meaningful to develop a convenient and sensitive detection method. Herein, a novel BODIPY-based fluorescent probe (BDP-DM) was developed, which had a higher selectivity for Cys than other amino acids, including homocysteine (Hcy) and glutathione (GSH). Ultimately, we concluded that the BDP-DM probe could be used to successfully detected intracellular Cys in living HeLa cells.

Concentrations of minor (Mg and Sr) and trace (Ba and U) elements in four natural calcium carbonate samples were first analyzed by inductively coupled plasma mass spectrometry (ICP-MS) after chemical dissolution and calibrated against a standard dolomite. Their homogeneities were checked by in situ laser ablation (LA) ICP-MS with 10 - 20 spots. The carbonate samples were measured by using a high lateral resolution secondary ion mass spectrometer (Nano-SIMS NS50). A ∼4 nA O- primary beam was used to sputter a 5 - 6-µm diameter crater on the sample surface, and secondary positive ions were extracted for mass analysis using an accelerating voltage of 8 kV and a Mattauch-Herzog geometry. A multi-collector system was adjusted to detect 26Mg+, 43Ca+, 88Sr+, 138Ba+, 238U16O+ and 238U16O2+ ions at the same time. A resolving power of 2500 - 5000 at 10% peak height was attained by an entrance slit set at 40 µm, and each exit slit at 50 µm with adequate flat-topped peaks. The observed 26Mg/43Ca, 88Sr/43Ca, 138Ba/43Ca and 238U16O2/43Ca ratios agreed well with those measured by LA-ICP-MS. Foraminifera shells were analyzed at 5 - 6 µm scale by Nano-SIMS. There was a large variation of the Mg/Ca ratios, up to ±38%, even in a single fragment of the shell, suggesting that although the ratios provide a useful paleoceanographic proxy at bulk scale, they may reflect a more complex pattern at <10 µm scale.

In the early stage of tumor development, tumor-associated macrophages (TAM) works to suppress tumor growth by secreting soluble factors including nitric oxide (NO). L-Arginine (Arg) is a substrate of nitric oxide synthase (NOS) expressed in TAM. Here we examined whether NO produced from Arg by macrophages works to enhance the effect of the anti-cancer drug, doxorubicin (Dox) by using a co-culture system of cancer cells with macrophages. By employing colorimetric analyses methods (Griess Reagent and Cell Counting kit-8), we found that NO produced from Arg by co-cultured macrophages could enhance the cytotoxic effect of Dox to cancer cells. Moreover, we found that augmentation is affected by the order of the addition of Arg and Dox. A prior addition of Arg to Dox and simultaneous addition showed the same enhancement effect, but a prior addition of Dox to Arg abolished the augmentation. This suggests that the co-administration of Arg with Dox would be an effective treatment to improve chemo-therapies.

This paper describes a novel method for fabricating paper-based microfluidic devices using a laser beam scanning technique. Cellulose chromatography papers were treated with octadecyltrichlorosilane (OTS) to make them entirely hydrophobic. A photoacid generator (CPI-410S) was soaked into the paper, and irradiated with a 405-nm laser beam to induce acid generating reactions. Since the silyl ether bond between cellulose and OTS was cleaved by the hydrolysis reaction, the photo-irradiated area changed to hydrophilic. By scanning the laser beam using a Galvo mirror system, arbitrary shaped hydrophilic patterns were successfully created on the paper in 50 μm resolution. To the best of our knowledge, this is the first report on the fabrication of hydrophilic channels on the OTS-treated paper using photo-induced acid generation processes coupled with the laser beam scanning technique. Quantification of nitrite was demonstrated with the paper device made by this method.

Liquid chromatography/electrospray ionization–tandem mass spectrometry (LC/ESI-MS/MS) enables the accurate and precise quantification of various analytes at very low concentrations, but it has room for improvement in analysis throughput. Multiplexing of samples in the same injection could be a promising procedure for enhancing the analysis throughput. This could be achieved by derivatization of the multiple samples with multiple isotopologous reagents. In this study, a sample-multiplexed LC/ESI-MS/MS assay using the 1-[(4-dimethylaminophenyl)carbonyl]piperazine (DAPPZ) isotopologues (2H0-, 2H3-, and 2H6-forms) was developed and validated for the simultaneous determination of primary bile acids in three different plasma samples in a single run. The developed method had satisfactory intra- and inter-assay precisions (≤ 2.3 and ≤ 4.2%, respectively) and accuracy (99.0 – 100.3%), and could reduce the total LC/ESI-MS/MS run time by more than 60% for 42 samples compared to the conventional method. Thus, the derivatization with the DAPPZ isotopologues worked well for enhancing the throughput of the LC/ESI-MS/MS assay of the bile acids.
