Previous workers have reported that Rubus chromosomes are too small for karyotype analyses at mitosis, and also that meiotic studies indicate no major structural differences between Rubus genomes. However, it was found possible to identify the seven chromosomes of Glen Clova, a cultivar of R. idaeus, and of R. coreanus. The chromosomes of R. idaeus differed from those of R. coreanus, four of them appreciably, and it was possible to identify each of the 14 chromosomes of four F1hybrids between the two species. In spite of these differences, meiosis and pollen formation in the hybrids appeared normal. It is suggested that genome differentiation in the genus is greater than realised, and that further karyotype analyses may aid studies of species relationships.

Meiotic chromosome configuration frequencies at diakinesis and metaphase I were tabulated in diploid and induced tetraploid <I>Rosa laevigata</I> Michx., diploid <I>R. roxburghii</I> Tratt., and tetraploid amphidiploids of <I>R. laevigata</I> with <I>R. roxburghii</I> and <I>R. banksiae</I> Ait. Numerical analysis of configuration frequencies suggests up to two-fold length variation between short and long chromosome arms in the diploids, and incomplete independence of chromosome arms during pairing and chiasma formation in the tetraploids. Since the sampled genomes pair homeologously in intersubgeneric amphidiploids, they are relatively closely related. The meiotic data collectively suggest that <I>R. roxburghii</I> should be classified in subgenus <I>Rosa</I>.

Labeling of specific DNA sequences in living organisms is important for understanding the mechanism of gene regulation during development, differentiation, and environmental adaptation. Here we describe a CRISPR/Cas9-based chromatin visualization system in plants. We adapted the mammalian nuclease-dead Cas9-fluorescent protein (dCas9-FP) system to a plant expression vector. Transient expression of dCas9-FP and single guide RNA (sgRNA) for telomeric sequences efficiently labeled telomere repeats in tobacco.

Antimitotic effect of holothurin, a water soluble saponin isolated from the sea cucumber Holothuria vagabunda, was assessed by Allium test supplemented by autoradiography. Endoreduplication, chromosome clumping, positive and/or negative pycnosis of nuclei, nuclear polymorphism and chromatin extrusion through cytoplasmic buds were the common cytological abnormalities induced by holothurin. Inhibition of DNA and RNA synthesis was observed and this may be the molecular mechanism behind the above abnormalities. In all these features holothurin appears to be similar to Adriamycin, currently used in Cancer Chemotherapy.

Cyanophora paradoxa is an enigmatic biflagellate that may represent the first photosynthetic eukaryote morphologically. This alga has been widely studied as a model organism of primitive phototrophs. However, surface ornamentations of the vegetative cells have not been examined using ultra-high resolution field emission (FE) scanning electron microscopy (SEM). In the present study, C. paradoxa NIES-547 vegetative cells were examined using FE-SEM and compared with the data using conventional SEM. Our FE-SEM images demonstrated that the cell surface was ornamented with angular fenestrations framed by ridges. In contrast, conventional SEM did not reveal similar surface ornamentation. Transmission electron microscopy showed the ridge was formed by the edges of overlapping or attaching outermost plate vesicles at the cell periphery.
