The toxic catabolic intermediates of branched chain amino acids can cause insulin resistance, and are involved in different mechanisms in different metabolic tissues. In skeletal muscle, 3-hydroxy-isobutyrate produced by valine promotes skeletal muscle fatty acid uptake, resulting in the accumulation of incompletely oxidized lipids in skeletal muscle, causing skeletal muscle insulin resistance. In the liver, branched-chain α-keto acids decompose in large amounts, promote hepatic gluconeogenesis, and lead to the accumulation of multiple acylcarnitines, which damages the mitochondrial tricarboxylic acid cycle, resulting in the accumulation of incomplete oxidation products, oxidative stress in mitochondria, and hepatic insulin resistance. In adipose tissue, the expression of branched-chain amino acid catabolic enzymes (branched-chain amino acid transaminase, branched-chain α-keto acid dehydrogenase) is reduced, resulting in an increased level of plasma branched-chain amino acids, thereby causing massive decomposition of branched-chain amino acids in tissues such as skeletal muscle and liver, and inducing insulin resistance. However, branched-chain amino acids, as a common nutritional supplement for athletes, do not induce insulin resistance. A possible explanation for this phenomenon is that exercise can enhance the mitochondrial oxidative potential of branched-chain amino acids, alleviate or even eliminate the accumulation of branched-chain amino acid catabolic intermediates, and promotes branched-chain amino acids catabolism into beta-aminoisobutyric acid, increasing plasma beta-aminoisobutyric acid concentration, improving insulin resistance. This article reveals the mechanism of BCAA-induced insulin resistance and the relationship between exercise and BCAAs metabolism, adds a guarantee for the use of BCAAs, and provides a new explanation for the occurrence of diabetes and how exercise improves diabetes.

The Yangtze River Delta is the best-known homeland of wet-rice agriculture. From the Middle Neolithic, rice farming expanded from the Yangtze region to both the north and the south. However, poor preservation of ancient human skeletal remains in the region has meant that the population history of these expansions has not been fully understood. In order to clarify the ancestry of early wet-rice farmers in East Asia, we conducted a cranial morphometric analysis and comparison of a Middle Neolithic skeletal assemblage excavated from the Guangfulin site, Shanghai. The results of bivariate and multivariate analyses showed that: (1) Neolithic wet-rice farmers from the lower Yangtze retained local morphological characteristics, but were nevertheless morphologically more similar to Neolithic and later populations in northern China, which was home to early millet farmers, than to Neolithic populations in south China; and (2) Neolithic and later agricultural populations in East Asia were morphologically homogeneous compared to pre-Neolithic hunter-gatherer groups even though the area occupied by both was equally vast. These results suggest, respectively, that: (1) Middle Neolithic wet-rice farmers in the Yangtze Delta experienced significant gene flow from regions of northern China such as the Central Plains and Shandong even though there is currently no evidence that millet cultivation itself had yet reached the delta region; and (2) Neolithic populations resulting from interaction between the Yangtze Delta and northern China dispersed widely across much of East Asia including the Japanese archipelago together with the spread of wet-rice agricultural technologies. These two proposals are paralleled by recent stable isotope analyses using tooth enamel and bone collagen, as well as archaeological evidence from Shandong. Finally, a facial approximation was conducted using a cranium (M252) excavated from Guangfulin for the purpose of visually expressing the results of this study.

Maternal folate and vitamin B12 deficiency predict poor pregnancy outcome. To improve pregnancy outcomes in rural area of China, we investigate rural women’s folic acid supplementation (FAS) status and the associations between maternal vitamin B status during the first trimester and subsequent adverse pregnancy outcomes. We collected the questionnaire information and drew 5 ml of blood from 309 early pregnant rural women. The birth outcomes were retrieved from medical records after delivery. Out of the total, 257 had taken FAS, including 50 before conception (group A) and 207 during the first trimester (group B). The concentration of plasma folate and the RBC folate supplementation groups were obviously higher than that of no-supplementation group (group N, p<0.01). The mean vitamin B12 levels in FAS group were significantly higher than those in groups N and B (p<0.05). Women who delivered SGA or premature infants had reduced plasma folate levels (p<0.05) compared with controls. The multiple linear regression models revealed that RBC folate levels affected the infant birth weight (p<0.01) and birth length (p<0.05). In conclusion, FAS can significantly improve plasma folate and RBC folate levels in childbearing-age women and reduce the risk of subsequent adverse pregnancy outcomes.
