Herein, we applied hypericin (Hyp) for the detection of DNA on streptavidin-functionalized magnetic beads (SA-MB) through non-covalent π-π stacking interaction between 1-pyrenebutanoic acid (PBA) and Hyp. Briefly, the target DNA (tDNA) was hybridized to morpholino oligonucleotide (MO) that was immobilized on the surface of SA-MB. PBA was incorporated to the backbone of tDNA by means of phosphate-zirconium-carboxylate coordination reaction. Then, fluorescent Hyp was linked to PBA via π-π stacking interaction. Under the optimal conditions, this biosensor displayed a good linear relationship between the fluorescence intensity and the tDNA concentrations in the range from 1 to 100 nM with a low detection limit of 2.9 nM. The mismatch type in a specific DNA sequence can also be efficiently discriminated. Compared with some other reports, this method is more convenient, and has great potential for practical applications.

Cysteine (Cys), as one of the important amino acids, plays a vital role in various physiological and pathological processes. Hence, it is meaningful to develop a convenient and sensitive detection method. Herein, a novel BODIPY-based fluorescent probe (BDP-DM) was developed, which had a higher selectivity for Cys than other amino acids, including homocysteine (Hcy) and glutathione (GSH). Ultimately, we concluded that the BDP-DM probe could be used to successfully detected intracellular Cys in living HeLa cells.

Aim: Inflammation plays a significant role in the pathogenesis of human abdominal aortic aneurysm (AAA). AEBP1 can promote activation of the NF-κB pathway, subsequently affecting the expression of NF-κB target genes, including inflammatory cytokines and matrix metalloproteinases (MMPs). Our objective was to examine the role of AEBP1 in the development of AAA and characterize the underlying mechanism. Methods: ITRAQ, RT-PCR, western blot, immunohistochemistry, and ELISA were used to compare different experimental groups with the controls and to determine the differentially expressed genes. We generated an AAA model using porcine pancreatic elastase in Sprague–Dawley rats and silenced their AEBP1 in vivo by adenoviruses injected intra-adventitially. We also silenced or overexpressed AEBP1 in human vascular smooth muscle cells in vitro in the presence and in the absence of NF-κB inhibitor BAY 11-7082. Results: Proteome iTRAQ revealed a high expression of AEBP1 in AAA patients, which was verified by qRT-PCR, western blot, immunohistochemistry, and ELISA. The mean expression level of AEBP1 in AAA patients was higher than that in controls. Along with AEBP1 upregulation, we also verified mis-activation of NF-κB in human AAA samples. The in vivo studies indicated that AEBP1 knockdown suppressed AAA progression. Finally, the in vitro studies illustrated that AEBP1 promotes activation of the NF-κB pathway, subsequently upregulating pro-inflammatory factors and MMPs. Conclusions: Our results indicate a role of AEBP1 in the pathogenesis of AAA and provide a novel insight into how AEBP1 causes the development of AAA by activating the NF-κB pathway.

Colla corii asini (E’jiao), donkey-hide gelatin prepared by stewing and concentrating from Equus asinus L. donkey hide, is a traditional Chinese medicine preparation widely used in clinical hematic antanemic therapy in China. The aim of the present study was to investigate potential anti-aging effect of Colla corii asini and explore related mechanisms in D-galactose (gal) induced aging model mice. The mice were artificially induced aging by subcutaneously injection with D-gal at the dose of 100 mg/kg·d for 8 weeks. Colla corii asini was simultaneously treated to them once daily by intragastric gavage. Appetite, mental condition, body weight, and organ index were observed. Activities of superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GSH-Px), as well as levels of malondialdehyde (MDA) in serum, brain, and liver were determined by according assay kits. Western blotting analysis was used to detect p16 and p21 expression. Results indicated that Colla corii asini could improve appetite, mental condition, body weight, and organ condition of model mice, improve SOD, CAT, and GSH-Px activities, reduce MDA levels, and modulate age-related genes expression in D-gal induced mice. Therefore, Colla corii asini may have effect to suppress the aging process through enhancing antioxidant activity, scavenging free radicals, and modulating aging-related gene expression.
