Typhula ishikariensis and the related fungi were separated into three biological species by morphological and physiological characteristics, as well as DNA sequences and mating reactions. We propose that the T. ishikariensis complex should be divided into three species (T. ishikariensis, T. canadensis and T. hyperborea) and two varieties (T. ishikariensis var. ishikariensis and var. idahoensis). Typhula hyperborea was reappraised to be recognized also as a separate species of the T. ishikariensis complex.

This review summarizes recent findings regarding the status of physical activity and introduces effective methods of intervention. Data from our serial cross-sectional study between 2003 and 2012 suggested that the decline in step counts over the last decade is mainly related to a reduction in non-exercise activity associated with the increased use of cell phones or computers and with playing video games. We then examined the effects of lifestyle interventions using an activity monitor with computerized game functions or an activity monitor and Twitter on physical activity and body composition. These findings suggested that lifestyle interventions using both of these strategies increases daily physical activity and reduces body fat more effectively than using an activity monitor alone. In addition, changes in physical activity and in body fat were significantly correlated. We also applied a randomized intervention to examine an effective method of increasing physical activity levels among college physical education students using a pedometer. We found that using a pedometer and inducing friendly competition or encouragement from peers increased step counts more effectively during soccer classes. These findings therefore have important implications for ensuring compliance with the Physical Activity Reference for Health Promotion 2013 and Active Guide.

DNA extraction has been difficult from some types of soil. Seven soil samples from agricultural fields and a forest, which were mainly volcanic ash soils, were used. Soil DNA could be extracted from only two of them using a commercially available kit exploiting bead-beating. When skim milk was added to the extraction buffer at 40 mg g-1 soil, DNA could be detected by electrophoresis from all the samples, indicating that the DNA from lysed cells was adsorbed by soil colloids. The addition of skim milk did not affect PCR-DGGE profiles. The improved method is applicable to the analysis of molecular communities in soils which strongly adsorb DNA.

Senescence-associated secretory phenotype (SASPs) secreted from senescent cells often cause the deleterious damages to the surrounding tissues. Although dedifferentiated fat (DFAT) cells prepared are considered a promising cell source for regenerative therapies, SASPs from DFAT cells undergoing long-term cell culture, which latently induce replicative senescence, have barely been explored. The present study was designed to investigate senescent behaviors in rat-derived DFAT cells at high passage numbers and to analyze the possible types of SASPs. Our data show that DFAT cells undergo senescence during replicative passaging, as determined by multiple senescent hallmarks including morphological changes in cell shape and nucleus. Moreover, RT2 PCR array analysis indicated that senescent DFAT cells expressed higher levels of 16 inflammatory cytokines (Ccl11, Ccl12, Ccl21, Ccl5, Csf2, Cxcl1, Cxcl12, Ifna2, IL11, IL12a, IL13, IL1a, IL1rn, IL6, Mif, and Tnf) associated with SASPs than non-senescent cells. This study implicates that rat DFAT cells undergo cellular senescence after long-term cell culture; cautious consideration should be paid to treat SASP secretion when senescent DFAT cells are used in regenerative medicine.

There is an increasing interest in the extraction of DNA from soil, since molecular techniques facilitate the analysis of the microbial community including unculturable microorganisms. Though various methods of direct DNA extraction have been devised, it still remains difficult to extract DNA from some soils. We developed an improved method to extract DNA from such “recalcitrant soils”. Andisol, volcanic ash soils are recalcitrant in this context. Skim milk increased the efficacy of DNA extraction from such soils, when added to the extraction buffer as an adsorption competitor to soil particles. This method is applicable to molecular community analysis of soils which strongly adsorb DNA.

The occurrence of ameloblastic fibro-odontoma (AFO) in the oral region is unusual and accounts for 1-3% of all odontogenic tumors. AFO presents mixed radiopaque patterns within the lesion with diverse findings; therefore, it is important to compare this tumor with other odontogenic tumors that radiographically present with calcified bodies. Herein, we observed the calcification patterns within the lesion in seven AFO cases (five males, two females; mean age, 8.3 years; age range, 4-13 years). Periapical and panoramic radiographs were obtained from all seven cases. Five cases underwent conventional computed tomography (CT) scanning, and one underwent cone beam CT. Classification of the calcifications primarily involved the following two characteristics on the X-rays: appearance and location of the lesions. All seven cases were located in the molar regions of the mandible in association with impacted teeth. The calcification patterns of these AFOs were mixed or inhomogeneous within the lesion with various findings, including complex odontoma-like calcifications. However, the patterns differed between panoramic radiography and CT in some cases. The radiolucent lesions in AFO demonstrated varying calcification patterns and were associated with impacted teeth on the CT images.(J Oral Sci 58, 533-537, 2016)
