Pholiota microspora (“nameko” in Japanese) is one of the most common edible mushrooms, especially in Japan, where sawdust-based cultivation is the most dominant method accounting for 99% of the production. The current strains for sawdust cultivation in Japan are considered to have been derived from a single wild strain collected from Fukushima, Japan, implying that commercial nameko mushrooms are derived from a severe genetic bottleneck. We tested this single founder hypothesis by developing 14 microsatellite markers for P. microspora to evaluate the genetic diversity of 50 cultivars and 73 wild strains isolated from across Japan. Microsatellite analysis demonstrated that sawdust-cultivated strains from Japan were significantly less genetically diverse than the wild strains, and the former displayed a significant bottleneck signature. Analyzing the genetic relationships among all genotypes also revealed that the sawdust-cultivated samples clustered into one monophyletic subgroup. Moreover, the sawdust-cultivated samples in Japan were more closely related than full-sibs. These results were consistent with the single founder hypothesis that suggests that all commercial nameko mushrooms produced in Japan are descendants of a single ancestor. Therefore, we conclude that cultivated P. microspora originated from a single domestication event that substantially reduced the diversity of commercial nameko mushrooms in Japan.

Microstoma longipilum sp. nov. collected from two localities in Japan is described. It is characterized by long apothecial hairs and salmon pink discs. Molecular phylogenetic analyses supported the novelty of the fungus. We additionally reported the overlooked morphology of hyphal mats, conidiogenous cells produced directly from ascospores, and conidia. With the addition of M. longipilum, now six species of Microstoma are documented in Japan.

Gentiana zollingeri (Gentianaceae) is an initial mycoheterotrophic plant that depends on a specific group of arbuscular mycorrhizal (AM) fungi for carbon source during underground growth after seed germination. In this study, a mycorrhizal fungus dominant in mycoheterotrophic seedlings of G. zollingeri was successfully isolated from a soil core collected from a point close to a flowering G. zollingeri. The AM fungal isolate was identified as conspecific or closely related to Dominikia aurea (Glomeraceae) by spore morphology and molecular phylogeny. Basic Local Alignment Search Tool (BLAST) searches against the MaarjAM database showed that the nuclear small subunit ribosomal DNA sequences of the isolate matched the AM fungal sequences obtained from a wide range of plants in various ecosystems, including several mycoheterotrophs. Thus, it is suggested that the AM fungal isolate is one of the cheating susceptible AM fungi. Furthermore, the sequences corresponded to those of a group of AM fungi dominantly detected in Japanese temperate forests. Accordingly, there is a possibility that mycoheterotrophic plants, including seedlings of G. zollingeri, may target AM fungi with a wide host range and ubiquitous distribution.

A previously unknown morel species apparently endemic to Japan is here described. Morchella nipponensis is proposed for this species. This new taxon displays archaic features recalling section Rufobrunnea (pileus lanceolate, few primary alveoli), and a yellow pileus similar to subsection Sceptriformis of section Morchella. The phylogenetic analysis of ITS rDNA, as well as RPB1, RPB2 and TEF1 genes from up to four collections suggests that this morel species represents a basal branch of section Morchella (yellow morels), for which the new subsection Japonicae is proposed.

Fungi in the genus Termitomyces are external symbionts of fungus-growing termites. The three rhizogenic Termitomyces species T. eurrhizus, T. clypeatus, and T. intermedius, and one species similar to T. microcarpus that lacks pseudorrhiza, have been reported from Ryukyu Archipelago, Japan. In contrast, only two genetic groups (types A and B) of Termitomyces vegetative mycelia have been detected in nests of the fungus-growing termite Odontotermes formosanus. In this study, we investigated the relationships between the mycelial genetic groups and the basidiomata of Termitomyces samples from the Ryukyu Archipelago. We found that all the basidioma specimens and the type B mycelia formed one clade that we identified as T. intermedius. Another clade consisted of the type A mycelia, which showed similarity to T. microcarpus, was identified as T. fragilis. Our results indicate that the Japanese T. eurrhizus and T. clypeatus specimens should re-named as T. intermedius.

Myconymphaea yatsukahoi is a fungus that has only been isolated once from a forest in the Sugadaira Research Station, Nagano, Japan. Over 20 y have passed since its first discovery but since then it has not been rediscovered. Here, we re-isolated M. yatsukahoi from the type locality and another location, Tambara Moor, Gunma, Japan. Sporophores of this species were detected by direct field observation in Sugadaira and by induction from soil from Tambara. We attempted to narrow down isolation sources of this species by investigating the excrements of Lithobiomorpha and Scolopendromorpha centipedes, which are frequently found in the two locations where the species is distributed. In both locations, we found M. yatsukahoi in the excrements of Lithobiomorpha but not Scolopendromorpha. Myconymphaea yatsukahoi appears to be a coprophilous fungus and the excrements of the predators living in soil may be promising isolation sources for understanding the hidden diversity of kickxellalean fungi.

長野県の亜高山帯針広混交林で得た標本について,形態比較ならびに分子系統解析によって,アメリカを基準産地とするRussula granulataと認め,日本新産種として報告する.子実体は中型,傘は灰色を帯びた橙色から赤味を帯びた褐色,表面に強い粘性を有し,中央部は黄色を帯びた赤色から赤味を帯びた褐色,柄は触れると中・下部が淡褐色に変色する.核rDNAの分子系統解析によって,日本産と北アメリカ大陸およびユーラシア大陸産R. granulataは単系統群をなした.本種の和名をヌメリクサハツモドキとする.

“Kakishimeji” identified as Tricholoma ustale and belonging to Tricholoma sect. Genuina is a common poisonous mushroom in Japan. Kakishimeji contains the toxic compound ustalic acid and causes digestive trouble. However, this fungus is consumed in some regions of Japan without any digestive issues. We clarified the probable species complex of Kakishimeji based on a phylogenetic analysis. We collected 89 basidioma specimens of Kakishimeji and related species from various forest sites in Japan and conducted phylogenetic analyses using 7 nuclear and mitochondrial gene sequences. Kakishimeji was found to consist of four distinct phylogenetic clades based on all DNA regions tested. Of these, two clades included European T. stans and T. albobrunneum type specimens. Another two clades consisted of sister clades to T. pessundatum and T. ustaloides. In addition, all four phylogenetic clades of Kakishimeji had different spore and basidium sizes. Therefore, we regarded the latter two clades as two new Tricholoma species: T. kakishimeji and T. kakishimejioides.

We describe a new species of Gerhardtia from Japan based on basidiomata morphology, live culture characteristics, and molecular phylogenetic analyses. Gerhardtia venosolamellata is found on broad-leaf litter, and is characterized by tricholomatoid to marasmioid basidiomata, an off-white to pale salmon-pink pileus surface with faint marginal striae, subdistant lamellae with lateral veins, a tomentose to strigose stipe base with hyphal strands generating arthroconidia measuring 4–7 × 2–3 μm, cyanophilic, elongate-ellipsoid to cylindrical, slightly verrucose or undulate basidiospores measuring 4.5–6 × 2.5–3 μm, and cyanophilic basidia measuring 25–35 × 5–6 μm and containing siderophilous granules. Phylogenetic analyses based on the internal transcribed spacer and large subunit regions of the fungal nrDNA indicates that G. venosolamellata is related to G. sinensis and G. highlandensis, but differs from the former with respect to basidiomata color, basidiospore shape, and habitat. An isotype specimen of G. highlandensis exhibited relatively close lamellae without veins, and slightly larger basidiospores (4.5–6.5 × 2.5–3 μm). Cultured mycelia of G. venosolamellata produced arthroconidia measuring 4.5–8.5 × 2.5–3 μm with both schizolytic and rhexolytic secession on MA and PDA media, and chlamydospores occasionally covered with crystals on MA and MYG media.

The genus Aciculosporium (Clavicipitaceae, Hypocreales, Ascomycota)was established in 1908 for A. take, which is the causal fungus of witches’ broom of bamboo. Although the original description was valid at that time, a type specimen for A. take has not been designated. To standardize the use of this genus and species name, a neotypification and reference specimen of A. take are proposed. Multilocus phylogenetic analyses based on DNA sequences from 28S rDNA, TEF, Tub2, Mcm7, and RPB2 revealed that A.sasicola is from a different lineage to A. take, and other specimens from wavyleaf basket grass (Oplismenus undulatifolius) represent a distinct species proposed here as Aciculosporium oplismeni sp. nov. Chemical analysis using mass spectrometry and nuclear magnetic resonance spectroscopy showed that A. take produces four proline-containing cyclic dipeptides, which are moieties of ergot alkaloids. However, ergot alkaloids, lolines, peramine, indole-diterpenes, and lolitrem were not detected in the culture solvent. This study offers clarification of the lineage and morphology of this genus.

Typhula ishikariensis and the related fungi were separated into three biological species by morphological and physiological characteristics, as well as DNA sequences and mating reactions. We propose that the T. ishikariensis complex should be divided into three species (T. ishikariensis, T. canadensis and T. hyperborea) and two varieties (T. ishikariensis var. ishikariensis and var. idahoensis). Typhula hyperborea was reappraised to be recognized also as a separate species of the T. ishikariensis complex.

In this work, we characterize naturally occurring mycorrhizae formed by Amanita viscidolutea on Guapira opposita in the Atlantic Forest in Brazil. We sequenced the rDNA ITS region from the mycorrhizae and basidiomata to identify both symbionts. Amanita viscidolutea mycorrhizae were up to 43 mm long, mostly simple, and unbranched to irregularly pinnate. The fungal mantle surface was velvety to slightly cottony and white to yellowish with silver patches. Hyphal strands were infrequently present. Although the fungal mantle consisted of clampless hyphae, emanating hyphae and hyphal strands had sparsely distributed clamp connections. A unique character of the mycorrhizae was the absence of a Hartig net.

Corticioid fungi form effused basidiomata with various hymenophore configurations, mostly on woody substrates, and they are presently classified into 15 orders in Agaricomycotina in the phylum Basidiomycota. In this review, the corticioid fungi of Japan are treated on the basis of the current classification system, and all currently known species are listed and classified by order. Japanese corticioid fungi number 442 species in 160 genera belonging to 14 orders. Analysis of the distribution of Japanese corticioid species reveals that 375 species (85% of the total) have wide distributions, and that the Japanese corticioid fungal flora is composed of species with diverse patterns of distribution. In the subtropical regions, 146 corticioid species belonging to 77 genera are listed, including 100 species from the Nansei Islands and 76 from the Ogasawara Islands. Although these two island groups are located at similar latitudes, they share only 30 corticioid species—21% of the total reported from the subtropical regions. Along with this summary of the current status, this study presents future directions for the taxonomic research of the Japanese corticioid fungi.

Fungi-algae interactions, such as lichen-forming fungi and parasitic chytrids on phytoplankton, are common in ecosystems. In contrast, interactions between filamentous fungi and soil algae that can be observed with the naked eye have been given little attention and remain unexplored. Here, we report a fungus that was associated with a visible symptom of dead algae on a soil surface in Sugadaira-kogen, Nagano, central Japan. Acremonium-like conidiophores were growing on vesicles and dead bodies of a yellow-green alga, Botrydium granulatum. The fungus was identified as Emericellopsis mirabilis based on its morphology by microscopic observation, phylogenetic analysis, and the similarity of the isolation substrate with the first description of the species. Co-culture experiments showed a filamentous cell differentiation of the alga by the fungus, but no harmful or beneficial effects on algal growth. Therefore, we speculate that E. mirabilis is a facultative parasite of B. granulatum under natural conditions.

Hydropodia silvae-nipponicae, a new agaric species was described based on the specimens collected from evergreen broad-leaved forests in Japan. The present new species is distinct from H. subalpina, the type species of the genus by morphological characteristics and nuclear rDNA sequences. Hydropodia silvae-nipponicae is characterized by its nodulose cystidium-like terminal elements of pileipellis, cylindrical to clavate pileocystidia with irregular excrescences, and dimorphic pleurocystidia. It is the second species belonging to the genus Hydropodia.

Fungi exhibit saprophytic, parasitic, and symbiotic lifestyles, and flexibly switching between them by the environmental changes and host conditions. However, only a few studies have elucidated the detailed changes in fungal DNA or morphology, including the formation of reproductive structures along with lifestyle switching. We hypothesized that Pyrenopeziza protrusa, which occurs abundantly and specifically on Magnolia obovata as a saprophyte, is also associated with living hosts and switches its lifestyles as part of its lifecycle. To elucidate this hypothesis, we periodically sampled the fresh/fallen leaves of M. obovata to observe the seasonal occurrence of reproductive structures for the isolation and detection/quantification of P. protrusa DNA with newly developed species-specific primers. The isolation frequency and amount of P. protrusa DNA drastically increased in the fresh leaves just before defoliation in autumn, but remained high in fallen leaves from autumn to spring. Abundant production of conidiomata and apothecia was also observed in the fallen leaves with increasing DNA content. These results clarified a large part of the lifecycle of P. protrusa, suggesting that the lifestyle is switched from symbiotic to saprophytic stage by significantly increasing the amount of DNA in response to host conditions according to the seasonal variations.

In the search for new strains of edible mushrooms in the Brazilian Amazon Forest, we found Lentinula specimens different from Lentinula raphanica. These were described morphologically and evaluated phylogenetically within the Lentinula clade. The mating system was determined, and interbreeding compatibility with L. raphanica was verified. The basidiomata have a cinnamon or deep orange to fulvous brown, moist to dry pileus, occasionally with whitish scales; crowded whitish cream lamellae; and an eccentric to lateral stipe. The typical, predominant basidiospores are 4.4-7.2 µm in length. Endogenous, elongate (7.8-14 µm) basidiospores were also found in some specimens. The long spores seem rare and occasional, but nonetheless a novelty for the group. Basidia are homogeneous in size, cheilocystidia are pyriform or bulboid, and caulocystidia are long and spheropedunculate. The hyphae of the pileipellis are pigment-encrusted. The characteristics match those of Agaricus ixodes originally described from Guyana, currently a synonym of Lentinula boryana. In the phylogenetic trees, such taxon appears distinct from L. boryana and sister to L. raphanica with strong support. This unique lineage was confirmed to be reproductively isolated from sympatric L. raphanica strains. Lentinula ixodes comb. nov. is the second species of the genus reported in the Amazon Forest.
