Blockchain systems rely on peer-to-peer (P2P) overlay networks to propagate transactions and blocks. The node management of P2P networks affects the overall performance and reliability of the system. The traditional structure is based on random connectivity, which is known to be an inefficient operation. Therefore, we propose MSLT, a multiscale blockchain P2P network node management method to improve transaction performance. This approach involves configuring the network to operate at multiple scales, where blockchain nodes are grouped into different ranges at each scale. To minimize redundancy and manage traffic efficiently, neighboring nodes are selected from each range based on a predetermined set of rules. Additionally, a node updating method is implemented to improve the reliability of the network. Compared with existing transmission models in efficiency, utilization, and maximum transaction throughput, the MSLT node management model improves the data transmission performance.

To identify the spectrum and prevalence of thirteen causative genes mutations in congenital hypothyroidism (CH) patients, we collected blood samples and extracted genomic DNA of 106 CH patients, and designed a customized targeted next-generation sequencing panel containing 13 CH-causing genes to detect mutations. A total of 132 mutations were identified in 65.09% of patients (69/106) on the following nine genes: DUOX2, TG, TPO, TSHR, TTF1, TTF2, NKX2-5, PAX8 and GNAS. 69.70% (92/132) mutations related to thyroid dyshormonogenesis genes, including DUOX2 (n = 49), TG (n = 35), and TPO (n = 8). 21.21% (28/132) mutations related to thyroid dysgenesis genes, including TSHR (n = 19), TTF1 (n = 5), TTF2 (n = 1), PAX8 (n = 2), and NKX2-5 (n = 1). 9.09% (12/132) mutations related to GNAS, which was associated with thyrotropin resistance. No mutation of THRA, TSHB, IYD or SLC5A5 was detected. Among 69 mutations detected patients, 41 (59.42%) patients were two or more mutations detected, and mutations of 30 (43.48%) patients related to two or three genes. According to the pathomechanism of the mutant genes, 57.97% CH patients were classified as thyroid dyshormonogenesis. Overall, DUOX2, TG and TSHR mutations were the most common genetic defects in Chinese CH patients, and thyroid dyshormonogenesis could be the first genetic etiology of CH in Chinese. Besides, multiple mutations accounts for a part of genetic pathogenesis.

Using the Hilbert Singular Value Decomposition (HSVD) and the Nonlinear Canonical Correlation Analysis (NLCCA), we analyzed the statistical relationship of MJO (Madden-Julian Oscillation) and ENSO (El Niño and Southern Oscillation). It was found that while a linear analysis produced no significant relationship between MJO and ENSO, a low-order nonlinear analysis based on the quadratic function of HSVD led to statistically significant lagged correlations. When their nonlinear relationship was further extracted by the NLCCA, stronger correlation was obtained, with the maximum correlation coefficient appearing while the MJO signals preceding the ENSO signals by around 2 months and 5.6 months respectively. The time lags producing the maximum correlation are respectively consistent with the characteristic time of MJO influencing ENSO dominated by two physical processes: the equatorial Kelvin waves and air-sea feedback. Corresponding to the two different physical processes, ENSO shows different features of development. In the former scenario, the westerly winds in the western Pacific excite the equatorial Kelvin waves which propagate eastward and deepen the thermocline in the eastern Pacific, resulting in the sea surface warming at the far eastern Pacific near the coast. In the latter scenario, the westerly anomalies in the western Pacific precede the development of El Niño through bringing surface warm water into the central and eastern Pacific, thus the sea surface warming occurs across the whole eastern Pacific ocean.
