As people age, geriatric syndromes characterized by frailty significantly impact both clinical practice and public health. Aging weakens people's immune functions, leading to chronic low-grade inflammation that ultimately contributes to the development of frailty. Effectively managing geriatric syndromes and frailty can help alleviate the economic burden of an aging population. This review delves into the intricate relationship among aging, infection-induced inflammation, chronic inflammation, and frailty. In addition, it analyzes various approaches and interventions to address frailty, such as smart rehabilitation programs and stem-cell treatments, offering promising solutions in this new era. Given the importance of this topic, further research into the mechanisms of frailty is crucial. Equally essential is the devising of relevant measures to delay its onset and the formulation of comprehensive clinical, research, and public health strategies to enhance the quality of life for elderly individuals.

The charge structure evolution of a mesoscale convective system with an anomalous or inverted charge structure, observed in the Severe Thunderstorm Electrification and Precipitation Study, a field project on the Colorado–Kansas border in summer 2000, is simulated using the Weather Research and Forecasting (WRF) model coupled with electrification and discharge processes. Two noninductive electrification schemes are used, based on the liquid water content (LWC) and the graupel rime accretion rate (RAR). The simulation with the LWC-based electrification scheme cannot reproduce the inverted charge structure with a positive charge region sandwiched by two negative charge layers, while the RAR-based electrification scheme produces the evolution process of a normal–inverted–normal charge structure in the convective region, which is consistent with the observations. In the low RAR (< 2 g m−2 s−1) region, graupel is mainly negatively charged when it bounces off ice crystals, while the ice crystals take up positive charge. However, in the zone where the inverted charge structure forms, a strong updraft (> 16 m s−1), high LWC (> 2 g m−3), and high RAR (> 4.5 g m−2 s−1) region appears above the height of the −20°C layer, so that a positive graupel charging region is generated above the −20°C layer of the convective region, resulting in a negative dipole charge structure with negatively charged ice crystals above the positively charged graupel. The negative dipole is superposed on the positive dipole (positive above negative) charge structure at the lower position to form an inverted tripole charge structure.

This study aimed to investigate the therapeutic effect of human umbilical cord mesenchymal stem cells (hUCMSCs) on experimental autoimmune thyroiditis (EAT) and the underlying mechanisms by utilizing a porcine thyroglobulin-induced EAT rat model. The rats received four tail vein injections of vehicle or hUCMSCs at an interval of 7 days and were sacrificed on day 28 after the first injection. Hematoxylin and eosin staining and enzyme-linked immunosorbent assays (ELISAs) were used to assess the therapeutic effects of hUCMSCs on EAT. Splenic lymphocytes were isolated from rats, and the proportions of CD4+ T cell subsets were analyzed by flow cytometry. Splenic CD4+ T cells from EAT rats were cocultured with hUCMSCs. A loss-of-function assay for protein tyrosine phosphatase non-receptor type 2 (PTPN2) was performed to explore the involvement of PTPN2/signal transducer and activator of transcription 3 (STAT3) signaling on the therapeutic benefit of hUCMSCs in EAT. hUCMSC treatment significantly alleviated inflammation, reduced serum thyroid antibody levels, and decreased the ratios of IL-17α+/CD25+FOXP3+ cells and serum IFN-γ/IL-4 in EAT rats. Furthermore, hUCMSC treatment upregulated PTPN2 protein expression in splenic lymphocytes of EAT rats as well as enhanced the PTPN2 protein level and attenuated phosphorylation of STAT3 in CD4+ T cells in vitro. Importantly, knockdown of Ptpn2 significantly reversed hUCMSC-mediated suppression of cell proliferation and hUCMSC-induced alterations in the expression of inflammatory cytokines in CD4+ T cells. Thus, hUCMSC treatment alleviates thyroid inflammation and the CD4+ T cell imbalance in EAT via PTPN2/STAT3 signaling, serving as a promising therapeutic approach for autoimmune thyroiditis.
