We investigated bluish-purple Platycodon grandiflorus flowers for yet unidentified flavonoid-related compounds and their flower coloration mechanisms. We identified a new polyacylated anthocyanin, delphinidin 3-O-[6-O-(α-rhamnopyranosyl)-β-glucopyranoside]-7-O-[6-O-(4-O-(6-O-(4-O-(6-O-(4-O-(β-glucopyranosyl)-trans-caffeoyl)-β-glucopyranosyl)-trans-caffeoyl)-β-glucopyranosyl)-trans-caffeoyl)-β-glucopyranoside] (3) as the minor anthocyanin component along with four known anthocyanins, four known flavones, and chlorogenic acid. The major anthocyanin of the bluish-purple P. grandiflorus flowers is platyconin. While platyconin has two caffeic acids linked in series via glucose molecules at the 7-position of delphinidin, anthocyanin 3 has three caffeic acids linked in series via glucose molecules at the 7-position of delphinidin. To investigate the effects of the number of aromatic acyl groups in polyacylated anthocyanin on bluing and stability, the color and stability of anthocyanin 3 in diluted aqueous solution (5 × 10−5 M) at a weak acidic condition (pH 5.7, the same pH as the petal sap) were compared with those of platyconin. Results showed that even if the three caffeic acids were bound in series at the 7-position of delphinidin, there was no further bluing or stabilizing effects of anthocyanin 3 compared to platyconin, but a tendency toward weakening color intensity was observed. Investigation of the change in anthocyanin content at the flower developmental stages showed that delphinidin 3-rutinoside-7-glucoside, which is the deacylated form of platyconin, first accumulated up to sufficient amounts at the bud stage, followed by an increase in platyconin content. At the bud stage, the petals were not purple or violet, except for veins, despite the presence of a good amount of delphinidin 3-rutinoside-7-glucoside, indicating the delphinidin 3-rutinoside-7-glucoside was present in some colorless state.

This paper presents simulation based design (SBD), which is an indispensable technique, particularly for emerging advanced machine developments. This makes the support of SBD a requirement for Computer-Aided Engineering (CAE) tools.While experience plays a crucial role in conventional CAE, in SBD, the design is created solely via simulations to achieve a highly optimal design with high reliability. SBD primarily relies on large scale optimizations with a vast design space and on solving large scale problems for virtual prototyping. Both require high computation power, however large computations result in better designs. Therefore, an SBD tool is expected to motivate and inspire users to aggressively use the latest computational power, instead of minimizing simulations.Unlike conventional CAE, SBD workflow is not a simple waterfall type, instead, it adopts a multi-perspective iterative process to effectively improve the design. The workflow in practice is primarily dictated by the tool. If the necessary functions are divided into multiple tools, the workflow will also be divided, which negatively affects iterations. The tool is expected to integrate the functions into one environment and encourage users to perform an adequate number of iterations in multi-perspective studies.The implementation of SBD is demonstrated via practical examples, and the expected functionality of an SBD tool is discussed.

Background: Our aim was to investigate the baseline clinical and echocardiographic parameters for predicting left ventricular (LV) dysfunction after anthracycline chemotherapy and heart failure (HF) hospitalization in a single cancer disease. Methods and Results: We studied 73 patients with malignant lymphoma and preserved LV ejection fraction (LVEF). Echocardiography was performed before and after anthracycline chemotherapy. Global longitudinal strain (GLS) was determined from 3 standard apical views. LV dysfunction after anthracycline chemotherapy was defined according to the current definition of cancer therapeutics-related cardiac dysfunction. Long-term (50-month) unfavorable outcome was prespecified as hospitalization for HF. A total of 10 patients had LV dysfunction after anthracycline chemotherapy. Multivariate logistic regression analysis showed that baseline GLS was the only independent predictor of this dysfunction. Receiver-operating characteristic curve analysis identified the optimal GLS cutoff for predicting LV dysfunction after anthracycline chemotherapy as ≤19% (P=0.008). Furthermore, the Kaplan-Meier curve indicated that fewer patients with GLS >19% were hospitalized for HF than among those with GLS ≤19% (log-rank P=0.02). For sequential logistic models, a model based on baseline clinical variables (χ2=2.9) was improved by the addition of baseline LVEF (χ2=9.0; P=0.01), and further improved by the addition of baseline GLS (χ2=13.1, P=0.04). Conclusions: Watchful observation or early therapeutic intervention with established cardioprotective medications may be necessary for patients with malignant lymphoma and preserved LVEF but with abnormal GLS.
