Due to the growing demand of n-3 polyunsaturated fatty acids (PUFA) as supplements and pharmaceutical products worldwide, there are concerns about the exhaustion of n-3 PUFA supply sources. We have successfully prepared high-quality scallop oil (SCO), containing high eicosapentaenoic acid and phospholipids contents, from the internal organs of the Japanese giant scallop (Patinopecten yessoensis), which is the largest unutilized marine resource in Japan. This study compared the cholesterol-lowering effect of SCO with fish oil (menhaden oil, MO) and krill oil (KO) in obese type II diabetic KK-A y mice. Four-week-old male KK-A y mice were divided into four groups; the control group was fed the AIN93G-modified high-fat (3 wt% soybean oil + 17 wt% lard) diet, and the other three groups (SCO, MO, and KO groups) were fed a high-fat diet, in which 7 wt% of the lard in the control diet was replaced with SCO, MO, or KO, respectively. After the mice were fed the experimental diet for 42 days, their serum, liver, and fecal lipid contents as well as their liver mRNA expression levels were evaluated. The SCO group had significantly decreased cholesterol levels in the serum and liver; this decrease was not observed in the MO and KO groups. The cholesterol-lowering effect of SCO was partly mediated by the enhancement of fecal total sterol excretion and expression of liver cholesterol 7α-hydroxylase, a rate-limiting enzyme for bile acid synthesis. These results indicate that dietary SCO exhibits serum and liver cholesterol-lowering effects that are not found in dietary MO and KO and can help prevent lifestyle-related diseases.

Although high dietary iodine intake due to consumption of edible kelp (kombu) is observed, there are few health problems associated with iodine in Japan. Since several kinds of isoflavones contained in soybeans act as goitrogens, the consumption of soybean may modify the adverse effects of excess iodine derived from kombu. In the present study, to clarify the effects of eating soybean meal on thyroid function and tissue distribution of iodine, we measured iodine content of serum and several organs in rats fed casein diet or soybean meal diet with or without kombu powder. Four types of experimental diet (casein diet, casein diet with 5% kombu, soybean meal diet, soybean meal diet with 5% kombu) were prepared using dried kombu powder (Saccharina diabolica; iodine content, 7.18 mg/g). Twenty-four male 4-week-old Wistar rats were divided into four groups and fed these four diets for four weeks each. No significant differences were observed in animal growth, thyroid weights, serum total thyroxine, triiodothyronine and thyroid stimulating hormone concentrations among the four groups. Irrespective of dietary protein (casein or soybean), the iodine content in thyroid gland of all four groups was significantly increased to a similar level by the administration of kombu. Iodine concentrations in serum, liver and kidney were also increased by the kombu, but the increases of iodine in these tissues were significantly lower in rats fed soybean than in those fed casein indicating that soybean meal reduced the content of iodine in serum, liver and kidney. These results suggest that the consumption of soybean products could prevent the manifestation of adverse effects due to an excess iodine intake from kombu.

For determination of selenium (Se) in biological materials, an improved method based on high performance liquid chromatographic determination of the fluo-rophore formed by reaction of selenite with 2, 3-diaminonapththalene was developed. The concentration detection limits were 0.5 ng/g in dried materials and 0.03 ng/mL in fluid ma-terials. In quadruplicate assays of 11 biological reference materials using the proposed method, measured Se concentrations were not significantly different from their certified val-ues, Thus, the proposed method is reliable and suitable for the determination of trace levels of Se in foods. Using the proposed method, Se concentrations in various kinds of tea were de-termined to assess the contribution of tea to daily Se intake in the Japanese population. Se concentration in the leaves of general black, green and oolong tea obtained in local retail stores was 33±19 ng/g (n=40). The leaves of a particular Chinese green tea sold under the name “high Se tea” were found to contain 455±184 ng/g (n=14) of Se. While the percent-age of Se extractable by infusion was less than 5% for the general teas, that in the high Se tea was more than 20%. These results indicated that intake of tea does not contribute to daily Se intake in the Japanese population. However, since infusions from high Se tea con-tained over 5 ng/mL of Se, consumption of over 1 L/d of tea derived from such high Se teas may increase the daily Se intake by close to 10%.
