This study investigated size at sexual maturity of the male sergestid shrimp Lucensosergia lucens (“Sakura-ebi” in Japanese). The shrimps were captured in the inner part of Suruga Bay, Japan in August 2013 using experimental trawls. Carapace length (CL), body length (BL), development of petasma, and presence of spermatophores in the terminal ampoule were documented. Probabilities of maturity as a function of CL were modeled using general linear models with a binomial and logit link function and predicted the CL at which 50% of individuals were mature (CL50). Morphologically mature males had a petasma connected with the pars astringens, and the basal part of the pars media was developed. In physiologically mature males, chambers in terminal ampoules were filled with spermatophores, including spermatozoa and appendages. CL50 (mm) was estimated to be 8.87 (=27.86 mm BL) and 7.84 (=24.65 mm BL) based on the morphological and physiological criteria, respectively. It is believed that most of the age-0 males attain sexual maturity within the first year and mature males are exploited in the autumn fishing season (October–December).

Cryphonectria carpinicola is an ascomycetous fungus that has been regularly found in its asexual form on European hornbeam (Carpinus betulus) in Europe over the past two decades. Here we describe the discovery of C. carpinicola in Japan and report for the first time its sexual state on Carpinus species. No symptomatic trees were observed, but stromata were found saprotrophically on broken branches of Carpinus species on the forest floor. The sexual structures of C. carpinicola resembled that of other Cryphonectria species and strongly resembled those of the closely related species C. radicalis. A phylogenetic tree based on the internal transcribed spacer sequences showed monophyly for the Japanese and European isolates of C. carpinicola. Further studies on the distribution and host range of C. carpinicola in Japan and on the life history strategies of this fungus are needed.

Agrobacterium tumefaciens (Rhizobium radiobacter) is used for the transient expression of foreign genes by the agroinfiltration method, but the introduction of foreign genes often induces transcriptional and/or post-transcriptional gene silencing (TGS and/or PTGS). In this study, we characterized the structural features of T-DNA that induce TGS during agroinfiltration. When A. tumefaciens cells harboring an empty T-DNA plasmid containing the cauliflower mosaic virus (CaMV) 35S promoter were infiltrated into the leaves of Nicotiana benthamiana line 16c with a GFP gene over-expressed under the control of the same promoter, no small interfering RNAs (siRNAs) were derived from the GFP sequence. However, siRNAs derived from the CaMV 35S promoter were detected, indicating that TGS against the GFP gene was induced. When the GFP gene was inserted into the T-DNA plasmid, PTGS against the GFP gene was induced whereas TGS against the CaMV 35S promoter was suppressed. We also showed the importance of terminator sequences in T-DNA for gene silencing. Therefore, depending on the combination of promoter, terminator and coding sequences on T-DNA and the host nuclear genome, either or both TGS and/or PTGS could be induced by agroinfiltration. Furthermore, we showed the possible involvement of three siRNA-producing Dicers (DCL2, DCL3 and DCL4) in the induction of TGS by the co-agroinfiltration method. Especially, DCL2 was probably the most important among them in the initial step of TGS induction. These results are valuable for controlling gene expression by agroinfiltration.
