Flavonoid 3'-hydroxylase (F3'H) has an important role in determining anthocyanin patterns in plants. Here, we analyzed root coloration characteristics in the radish by investigating anthocyanidin and expression of F3'H genes in cultivars with purple, red, or white roots. Cyanidin was detected in the roots of the purple radish, while pelargonidin was found in the red radish; however, neither of these anthocyanidins was detected in white radish. We isolated the RsF3'H gene of the purple root radish and found that it showed a relatively higher level of expression compared to the RsF3'H of the red radish. Moreover, we identified a retrotransposon, gypsy-Ty3, in the first exon of the F3'H homolog in the red radish. These results suggested that the F3'H enzyme may determine cyanidin-based anthocyanin in the purple radish, and that the lack of F3'H function due to the retrotransposon insertion, contributed to pelargonidin-based anthocyanin accumulation in the red radish.

Wide-ranging varieties and/or strains of bulb onions (Allium cepa Common onion group) and shallots (A. cepa Aggregatum group) were utilized to understand the variation in chemical compounds responsible for their taste. The bulb samples of 10 F1 commercial onion varieties (seven short-day and three long-day varieties) from Japan and 12 shallot landraces from abroad (Vietnam: three landraces; Indonesia: nine landraces) were collected as plant materials once a year in 2014 and 2015. The contents of S-alk(en)yl-L-cysteine sulfoxides, total flavonoids, and soluble sugars—including fructose, glucose, sucrose, and fructans—were determined to find differences between bulb onions and shallots, as well as to detect variations among varieties and/or landraces. While a principal component analysis (PCA) based on the results from both 2014 and 2015 could clearly discriminate shallots from bulb onions from a phytochemical perspective, bulb onions mainly had higher monosaccharides than shallots. By contrast, shallots produced more disaccharides than bulb onions. In most cases, regression analyses using the numerical data of the chemical compounds found in bulb onions and shallots suggested year-year correlations between 2014 and 2015. The flavonoid and PeCSO (S-(1-propenyl)-L-cysteine sulfoxide: isoalliin) contents in shallots were higher than those detected in bulb onions, which indicated the stronger pungent and bitter taste could be attributable to excess amounts of these compounds in this tropical plant.

Tree peony (Paeonia spp.) is a highly ornamental shrub originally from China. Its cultivars are classified into four groups: Chinese, Japanese, French, and American, based on morphological characteristics and geographic location. The French and American cultivars were developed by crossing the wild species P. lutea Delavay ex Franch. and P. delavayi Franch. with Japanese and Chinese cultivars. In this study, we evaluated phenetic relationships among major cultivar groups in the genus Paeonia L., using a dendrogram reconstructed from band pattern data for random amplified polymorphic DNA (RAPD) markers developed in a previous study. We also developed two sequence tagged site (STS) markers using RAPD markers specific to the French and American tree peony cultivar groups and an intersubgeneric hybrid. The former is an LuDeB marker able to detect the genetic relationship with P. lutea and P. delavayi, and the latter is an HPB marker able to identify the cpDNA of herbaceous peony in intersubgeneric hybrid cultivars between herbaceous and tree peony. The possession of bands obtained in this study also supported the breeding history of cultivar development in the genus Paeonia.

Purple and red colored root characteristics were introduced to a local pungent radish “Izumo orochi daikon” ‘Susanoo’, and several characteristics including food functional components were evaluated. The root and leaf phenotype, the pigment composition, the isothiocyanate and soluble solids contents, and the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity in the root were investigated. These characteristics were compared between the mass selection breeding lines of purple, red roots and their parent cultivars. Although one-third of individuals in the M6 and M7 were non colored, the colored root characteristics was introduced into ‘Susanoo’. The main anthocyanins in the deep reddish purple colored purple root lines were acylated cyanidin 3-sophoroside-5-glucosides, and those in the vivid purplish red colored red root lines were acylated pelargonidin 3-sophoroside-5-glucosides, which corresponded to the main anthocyanins in their respective colored root parental cultivars. The isothiocyanate contents in the purple and red root lines were almost the same as that in ‘Susanoo’. The DPPH radical scavenging activity of the purple and red root lines was almost two times higher than that of ‘Susanoo’. These results showed that the food functionality was enhanced by introduction of a colored root characteristic in ‘Susanoo’.

Double flower form is an important trait of ornamental flower plants such as Japanese azalea because of its high visual appeal. We investigated floral phenotypes and a MADS-box C class gene to characterize floral morphology and this gene in single- and double-flowered cultivars of Japanese azaleas. Normal floral organs (sepals, petals, stamens, and carpels) were observed from each whorl of single flowers, whereas in double flowers, petaloid stamens or petals were observed from the third and fourth whorls, and there were no normal stamens or carpels in any flowers. Molecular analysis revealed that expression of the azalea AGAMOUS/PLENA (AG/PLE) homolog was lower in the inner two whorls of double flowers than in inner two whorls of single flowers, and expression of the AG/PLE homolog was higher in the double-flowered cultivars ‘Surugaman-yo’ and ‘Yodogawa’ than in other double-flowered cultivars. Moreover, sequence analysis of AG/PLE mRNA revealed deletion mutations in the coding regions of the AG/PLE homolog in ‘Surugaman-yo’ and ‘Yodogawa’. These results suggest that the double flowers of Japanese azalea cultivars are formed as the down-regulated expression and deleted sequence mutation of the azalea AG/PLE homolog.
