In this paper we describe the development of a fully-automated solution-phase synthesizer, ‘ChemKonzert’ that can be used to prepare a wide variety of organic compounds. The automated synthesizer is ingeniously designed to perform most of the chemical reactions currently used by synthetic organic chemists and utilizes a centrifugal separator to efficiently achieve liquid-liquid extraction. The design of the hardware and software will be described in this paper, and several examples of organic reactions will also be presented as applications of the apparatus.

We report a case of non-necrotizing soft tissue infection and streptococcal toxic shock syndrome (STSS) caused by a novel emm subtype (emm76.10) of group A Streptococcus pyogenes (GAS). A 54-year-old Japanese woman suffering from fever, fatigue, and lower abdominal pain along with erythema for 3 days was admitted to our hospital. Additionally, she presented with hypotension and multiple organ failure. Exploratory incision was performed due to the presence of STSS and for an examination of the necrotizing soft tissue infection from her lower abdomen to the left thigh. Tissue cultures from the exudates and fascia yielded positive results for GAS growth, although blood cultures returned as negative. After 15 days of antimicrobial therapy, she recovered fully without any complications. Genotyping of the isolate indicated a novel emm subtype (emm76.10), with 5 amino acid substitutions in the emm76.0 subtype sequence and the full-length sequence of 780 bp. This isolate was resistant to tetracyclines, macrolides-lincosamides, and fluoroquinolones, owing to the presence of antibiotic resistance determinants, tet(M) and erm(B), and point mutations, Ser79Phe/Ser81Phe, in the quinolone resistance-determining regions of parC/gyrA. In conclusion, our observations suggest the importance of early-stage exploratory incision and drainage from the infected region for the isolation and characterization of causative bacteria to facilitate selection of appropriate antibiotics for treatment.

Adults of Oithona atlantica, Oithona plumifera and Oithona similis occurred throughout the year in Toyama Bay (0-500 m), but in the surface layers (0-50 m), the adults of O. atlantica and O. plumifera were observed in Toyama Bay during different seasons. Based on the differences in adult distribution, O. atlantica nauplii were found in samples collected during spring (April to May) while O. plumifera were found in samples collected during autumn (September to October). Identification of O. atlantica and O. plumifera nauplii and comparisons of their morphological differences were based on this temporal separation. Nauplii of O. atlantica, O. plumifera and O. similis differed in their ratio of body height (maximum length of dorsal-ventral axis) to length (H/L). This characteristic ratio was common to all naupliar developmental stages and is therefore useful as a basis for distinguishing between naupliar stages of the three species. The population and community structure and densities of these species of Oithona were investigated in the surface water layer (0-50 m depth) of Toyama Bay from 1997 to 1999. Major population structural changes occurred in different sampling seasons, and may have been influenced by changes in hydrographic conditions, including temperature and water current. In winter and spring (from February to June), many O. similis and O. atlantica nauplii appeared, and the former nauplii were more dominant. Oithona similis nauplii are suggested to may be the most important species as prey for fish larvae.

We aimed to clarify antimicrobial susceptibility patterns of anaerobes from diseased companion animals. Bacterial identification was based on the Japanese 2012 guidelines for the testing of anaerobic bacteria. AST was performed using the broth microdilution method. The anaerobe-containing samples collected from 2014 to 2018 included blood (anaerobe recovery rate, 5.0%), bile (9.4%), joint fluids (0.6%), pleural effusions (42.6%), ascites (64.1%), cerebrospinal fluids (3.0%), and punctures (75.0%). The anaerobes identified included Bacteroides spp. (33.2%), Peptostreptococcus spp. (19.6%), Prevotella spp. (13.6%), Propionibacterium spp. (10.3%), Clostridium spp. (9.3%), and Fusobacterium spp. (7.5%). Bacteroides fragilis group isolates were resistant to penicillin G (100%), ampicillin (100%), cefmetazole (63.6%), ceftizoxime (90.0%), and clindamycin (40.0%). Our observations demonstrated antimicrobial susceptibility in anaerobes isolated from Japanese companion animals.
