One anthocyanin and three flavones were isolated from the flowers of Tripora divaricata (Lamiaceae). They were identified as delphinidin 3-O-[(6″′-E-p-coumaroylglucosyl)-(1→2)-(6″-E-p-coumaroylglucoside)]-5-O-(6″′-malonylglucoside), apigenin 7-O-glucuronide, scutellarein 4′-methyl ether 7-O-glucuronide, and acacetin 7-O-glucuronide. In addition, we measured absorption spectra of intact petal and the buffer solutions (pH 5.6) containing isolated compounds for elucidation of the blue violet color development in T. divaricata. The absorption spectral curve of the buffer solution containing isolated anthocyanin showed the blue violet. Its color was identical to that of the intact flowers. These results indicate that the intramolecular copigmentation of anthocyanin mainly occurred in the blue violet flowers of T. divaricata.

Three anthocyanins, delphinidin 3-O-glucoside, petunidin 3,5-di-O-glucoside, and petunidin 3-O-glucoside, and rosmarinic acid were isolated and identified from the flowers of three lungwort (Pulmonaria) cultivars. Composition of these compounds were quantitatively compared during two developmental stages, i.e. violet color in early flowering stage and blue color in later flowering stage. In addition, pH values of petal juices and flower color value were measured in both stages. Qualitative differences among three cultivars were observed in anthocyanins. However, qualitative and quantitative changes during development was not observed. Increase of rosmarinic acid was observed in one cultivar during development of the flowers. Notable alkalization was not revealed in vacuoles. These results mean that the flower bluing in these lungwort cultivars is not mediated by intramolecular copigmentation and alkalization of vacuole. Furthermore, we performed in vitro examination using the isolated anthocyanin, delphinidin 3-O-glucoside or petunidin 3-O-glucoside, and rosmarinic acid. As the results, it was shown that the absorption curves of the mixture solutions and intact petal did not match. Thus, it was suggested that the petal bluing after flowering may be due to intermolecular interaction which was influenced by metal ions.

Double-flowered Eustoma, a popular cut flower, sometimes shows nonuniform coloration of its petals when harvested at the flower bud stage. At the tips of the petals, pale greenish areas remain after the flowers open. This considerably reduces the value of cut Eustoma. In this study, to identify appropriate postharvest treatments leading to normal coloration of these cut flower buds, we investigated the influence of harvest stage, temperature, sugar, and methyl jasmonate (MeJA) on petal coloration using the Eustoma ‘Voyage (Type II) Blue’. Investigation of the harvest stage, which was compared at three flower bud stages (STG2, 3, and 4), revealed that detached pale greenish flower buds (STG2 and 3) had a high probability of becoming nonuniformly colored flowers, so these stages were used for subsequent investigations. Temperature, which was investigated at 20, 25, 30, and 35°C, did not show any influence on petal coloring. Investigation of sugar treatments, such as 1% or 3% glucose or sucrose, also showed no influence. However, MeJA treatment led to a significant difference in petal coloration. The nonuniform coloration of petals was reduced by continuous exposure to MeJA vapor. Furthermore, we performed shorter MeJA treatments, such as for 1 or 2 days after harvesting, which reduced the nonuniform coloration as well as continuous MeJA exposure. To evaluate the effect of MeJA treatment on petal coloration, we measured the rate of pale green areas of petals using digital images. The analysis revealed that the greenish areas in petals exposed to MeJA vapor were significantly smaller than in petals not exposed to MeJA. In addition, there were fewer days to flowering after MeJA treatment. We concluded that postharvest treatment with MeJA is effective for reducing nonuniform coloration of early harvested flower buds of the double-flowered Eustoma ‘Voyage (Type II) Blue’.

A new tetraglycosyl flavonol, 3-O-[2-O-xylosyl-6-O-(3-O-glucosyl-rhamnosyl) glucosyl] kaempferol was isolated from pale purplish-pink petals of Wabisuke camellia cv. Tarokaja with three known flavonols. It was named urakunoside after the species name of Tarokaja, Camellia uraku. Urakunoside was a major flavonol component in the Tarokaja petals, but was not detected in petals of Tarokaja’s presumed ancestor species.
