Simple and rapid tools for antibody detection are beneficial for therapeutic monoclonal antibody development. Using a synthetic cationic antibody-recognizing peptide, a label-free colorimetric assay for antibody detection was established, in which a solution containing silver nanoparticles (AgNPs) changes color depending on particle aggregation/dispersion. Among the peptide probes we previously screened as IgG binding, one (NKFRGKYK) has four cationic amino acids and a pI of 10.46. Hence, we hypothesized that the peptide would both bind IgG and induce anionic AgNP aggregation via electrostatic interactions. This dual functionality of the IgG binding peptide could be useful for colorimetric detection. The detection of IgG in a solution containing culture media was investigated, and IgG was successfully detected at 100 to 500 nM within 2 min. This method is promising for high-throughput selection of IgG-producing cells since it is fast, readily observable, and does not involve complicated nanoparticle functionalization.

In the present study, biomolecule-stabilized Au nanoclusters were demonstrated as a novel fluorescence probe for sensitive and selective detection of pazufloxacin mesilate (PZFX) for the first time. The linear decrease in the fluorescence intensity of Au nanoclusters induced by PZFX allowed for the quantitative detection of PZFX in the range of 0.15 μg/mL to 1 mg/mL, and the detection limit for PZFX was 0.2 μg/mL. Circular dichroism spectroscopy and fluorescence decay studies were then performed to discuss the quenching mechanism. In addition, practical application of the present approach was also demonstrated for real samples, which suggested its great potential for accurate analysis of similar drugs.

<p>This paper describes a novel method for label-free mercury(II) ion detection based on exonuclease III-induced target signal recycling amplification using double-stranded DNA templated copper nanoclusters. The synthesized DNA-Cu nanoclusters were used with exonuclease III loop amplification technology for ultra-high sensitivity detection of mercury(II) ions, which were detected by significantly decreased fluorescence intensity. Under the optimal experimental conditions, there was a clear linear relationship between Hg<sup>2+</sup> concentration in the range of 0.04 to 8 nM and fluorescence intensity. The detection limit for Hg<sup>2+</sup> was 4 pM. In addition, the interference of other metal ions on the mercury(II) ion detection was also studied. To confirm the application of the fluorescent sensor, it was applied to determine the concentrations of mercury(II) ions in tap water, and the results showed that the method can be used to detect mercury(II) ions in water samples successfully.</p>

Nanogold particles of size 15 nm were used to label goat anti-human ceruloplasmin (GCP) to obtain an immunonanogold probe (AuGCP) for ceruloplasmin (CP). In a pH 7.8 citric acid-Na<sub>2</sub>HPO<sub>4</sub> buffer solution and in the presence of polyethylene glycol (PEG), an immunoreaction between AuGCP and CP took place, and the released nanogold particles aggregated to bigger clusters, which caused the resonance scattering (RS) intensity at 552 nm (<i>I</i><sub>552nm</sub>) to be enhanced greatly. The enhanced intensity &Delta;<i>I</i> was proportional to the CP concentration (<i>C</i><sub>CP</sub>) in the range from 0.0030 to 1.26 &micro;g/mL, with a regress equation of &Delta;<i>I</i> = 101.0<i>C</i><sub>CP</sub> + 1.8, a correlation coefficient of 0.9970, and a detection limit of 1.1 ng/mL CP. This simple and sensitive RS immunoassay was applied to the determination of CP in human plasma, with satisfactory results.

Di(phenyl)-(2,4,6-trinitrophenyl)iminoazanium [DPPH] is widely used as a standard for measuring the number of free radicals. Here, we evaluated the number of free radicals of “DPPH” reagents from three manufacturers by effective magnetic moment method. Interestingly, the reagents from different manufacturers had varying temperature dependencies for both magnetic moment and g-value at low temperatures. As a result, the maximum relative difference among the three reagents on the number of free radicals per unit mass was 20%. Carbon hydrogen nitrogen (CHN) analyses, high-resolution EPR measurements, FT-IR measurement, and NMR measurement confirmed that a major component of only one among the three reagents was “pure” DPPH. The evaluated purity based on free radical content was 0.998 kg kg−1 with expanded uncertainty of 0.036 kg kg−1. The other two reagents were found to be contaminated by several % of benzene in the DPPH crystal structure.

The inorganic arsenic (i-As) in grape products, in particular juice, wine and raisins, collected from the Japanese market was investigated. The concentrations of total As in nine grape concentrated juices ranged from 3 to 20 ng g−1, and more than 80% of the As was inorganic according to the results of speciation by HPLC-ICP-MS. Among them, four samples contained more than 10 ng g−1 of i-As, although 10 ng g−1 of i-As is the limit for apple juice recommended by the U.S. Food and Drug Administration. Moreover, more than 10 ng g−1 of i-As was found in some of the wine and raisin samples, with the total As concentrations ranging from 17 to 37 ng g−1. When fresh grapes were analyzed, As was mainly concentrated in the pericarp and a small amount was found in the fruit, although no As was observed in the branches and the juice. When grapes, including the pericarp, are processed, the As concentration in the products may increase during production processes such as drying, concentration, and ripening.

Double-viewing-position single-particle inductively coupled plasma–atomic emission spectrometry (DVP-SP-ICP-AES) measures emission intensity at two ICP vertical positions simultaneously using a single photomultiplier tube. A particle travelling up the ICP gives two consecutive temporal emission peaks. The Yb II 328.937-nm emission intensity of the two peaks for single Yb2O3 particles of diameter of 200 – 2000 nm are plotted against each other in a correlation plot. The correlation is poor when the gas temperature at the lower observation position is approximately the boiling point of the particles. Poor particle vaporization at the center of the central channel occurs because the gas temperature is 400 K lower than the temperature at the rim. The correlation is improved by shifting the observation positions up or using helium–argon mixed carrier gas to increase the gas temperature. Gas temperature is an important parameter for precise single particle-ICP measurements. DVP-SP-ICP-AES can be used to identify poor particle vaporization without the need of temperature measurement.

The increase in the use of therapeutic monoclonal antibodies (mAbs) and antibody-drug conjugates (ADCs) has made the detailed bioanalysis of these drugs essential not only for planning optimal therapeutic programs for clinical practice, but also for evaluating the biological equivalencies in the development of other biosimilars. The ligand binding assays that are widely in use now are being replaced rapidly by the highly accurate, sensitive, and selective analytical method using a mass spectrometer. This review will discuss the progress in and challenges observed during the development of a mass spectrometry-based bioanalytical method for therapeutic mAbs and ADCs.

We performed a comprehensive quantification of 20 amino acids in RPMI 1640 medium-cultured human colorectal adenocarcinoma cells to evaluate the efficacy of 5-fluorouracil treatment under hypoxic and hypoglycemic conditions, which mimic the tumor microenvironment. In this study, we developed a simple and comprehensive analytical method by using LC-MS/MS connected to the Intrada amino acid column, which eluted amino acids within 9 min. The present method covered a linearity range of 3.6 – 1818 μM, except for Gly (227 – 1818 μM), Ala, Asp, His (7.1 – 1818 μM each), and Trp (3.6 – 909 μM). The limits of detection were in the range of 0.02 – 38.0 pmol per injection in a standard solution. Amino acid concentration data were analyzed using principal-component analysis to represent samples on two-dimensional graphs. Linear discriminant analysis was used to classify samples on the score plots. Using this approach, the effect of 5-fluorouracil treatment could be successfully discriminated at high discrimination rates. Moreover, several amino acids were extracted from corresponding loading plots as candidate markers for distinguishing the effects of the 5-fluorouracil treatment or tumor microenvironmental conditions. These results suggest that our proposed method might be a useful tool for evaluating the efficacy of anticancer drugs in the tumor microenvironment.

Herbal medicine (HM) formulae are the combinations of two or more types of constituting herbs. This study has proposed a novel approach to efficiently develop HPLC methods for HM formulae, which take advantage of the mutual retention rules between HM formulae and their constituting herbs. An HM formula composed of two herbs, Radix Salviae Miltiorrhizae and Rhizoma Chuanxiong, was taken as a case study. Based on design of experiments and stepwise multiple linear regression, models relating the analytical parameters to the chromatographic parameters were built (correlation coefficients >0.9870) for chemical compounds in the two herbs. These models representing the retention rules were utilized to predict the elution profile of the formula. The analytical parameters were numerically optimized to ensure adequate separation of the analytes. In validation experiments, satisfactory separations were achieved without any pre-experiments on the formula. The approach can significantly increase the HPLC method development efficiency for HM formulae.

In this study, a rapid and reliable high-performance liquid chromatography–tandem mass spectrometry (HPLC-MS/MS) method for the determination of ambroxol in human plasma was developed and validated using palmatine as an internal standard (IS). Ambroxol and IS were extracted from 200 μL of human plasma via a simple protein precipitation preparation. Chromatographic separation was achieved on a Platisil C18 column (150 × 4.6 mm, 5 μm) using methanol–0.01% formic acid (70:30, v/v) as the mobile phase at a flow rate of 0.6 mL/min under an isocratic condition. The MS acquisition m/z 379 → 264 for ambroxol and 352 → 336 for IS was performed by atmospheric-pressure chemical ionization (APCI) mass spectrometry in selected reaction monitoring mode. The calibration curve for ambroxol was linear over the concentration range of 2.500 – 180.0 ng/mL. The matrix effects of ambroxol ranged from 104.6 to 112.7%. This fully validated method was successfully applied to a pharmacokinetic study of ambroxol in humans after oral administration of ambroxol at a single dose of 75 mg.
