Saxifraga fortunei sensu lato (Saxifragaceae) varies greatly in morphology and includes many infraspecific taxa. As many as five varieties have been recognized in western Japan, although their discrimination based on leaf size or leaf shape, such as cuneate base or degree of lobing is often difficult to determine due to continuous variation. To quantify the variation in leaf morphology, especially focusing on cuneate and parted leaves, 259 samples from 87 populations in western Japan were subject to principal component analysis. Additionally, developmental changes in plants from three selected populations were examined. The findings revealed that plants with parted leaved (var. suwoensis) were discriminated from plants with cuneate and normal leaves, while plants with cuneate leaves (var. obtusocuneata) were within the range of variation of var. fortunei. Leaf shape changes from cuneate to truncate or cordate during the transition from a vegetative state to the flowering state. Cuneate forms usually occurred in sterile plants, but also on flowering plants even within a population. This characteristic was less useful for recognizing var. obtusocuneata. Populations with parted (long lobed) leaves as well as those with cuneate leaves usually grow along streams, confirming that phenotypes are the result of adaptive differentiation toward becoming facultative rheophytes, as previously suggested.

A new color variant of Lecanorchis japonica Blume, L. japonica f. lutea (Orchidaceae), is described from Kagoshima and Chiba prefectures, Japan. Lecanorchis japonica f. lutea differs from L. japonica f. japonica only by its brilliant yellow coloration. Additionally, L. japonica f. lutea is superficially similar to L. hokurikuensis f. kiiensis but differs in having widely open flowers (vs. barely open flowers) and a nearly entire column wing (vs. apparent projections below column wing).

Cyperus pacificus (Ohwi) Ohwi var. margoinflatus S. Fujii, a new variety of C. pacificus (Cyperaceae) is described. It closely resembles C. pacificus var. pacificus in the oblong-ellipsoid achenes with winged margins, but the margins are well inflated with spongy parenchyma. It usually occurs in the dried bottoms of irrigation ponds, and often intermixed with other species of Cyperaceae such as C. extremiorientalis Ohwi, C. nipponicus Franch. et Sav. var. nipponicus and var. spiralis Ohwi, Fimbristylis dipsacea (Rottb.) C.B. Clarke, and F. squarrosa Vahl var. esquarrosa Makino. A key to the new variety and closely related taxa is also provided.

A new species, Chrysosplenium suzukaense, from the northern part of the Yoro Mountains and Suzuka Mountain Range, Central Honshu, Japan, is described. Chrysosplenium suzukaense belongs to ser. Macrostemon, and is most similar to C. fauriei in the C. fauriei group (C. fauriei, C. kiotense and C. nagasei) distributed on the Sea of Japan side of Japan. However, C. suzukaense differs from C. fauriei in always having four stamens opposite the calyx lobes (vs. eight stamens), and smaller stature and flowers. The distribution of C. suzukaense is nearer the Pacific Ocean side, but in an area of much snow in winter, showing climatic conditions similar to the Sea of Japan side. The rosette-like basal leaves retained until end of the flowering time in C. suzukaense and the C. fauriei group may be an adaptation to the Sea of Japan side environment, especially to a much snow. The chromosome number, representing the karyotype, of C. suzukaense was confirmed to be 2n=22, and similar to other members of the C. fauriei group.

A set of 8 polymorphic EST-SSR markers has been developed to evaluate the genetic diversity and genetic structure of the populations of Tricyrtis sect. Tricyrtis (Liliaceae) in Kyushu, which reproduces sexually and asexually by bulbils. The total number of alleles for each locus ranged from 2 to 11 with an average of 5.5, while the values of observed and expected heterozygosity ranged from 0.043 to 0.875 and 0.043 to 0.849, respectively. The combined probabilities of identity (PI = 6.0E-07 and PI-Sib = 2.7E-03) of these markers suggest that they are useful for not only evaluating genetic diversity but also for estimating clonal diversity within populations. Cross-species amplification was evaluated for three phylogenetically related species, T. macropoda, T. setouchiensis, and T. affinis, to show that all the EST-SSR markers are transferrable among these species.

Plants recognized as Liparis koreana (Nakai) Nakai ex W.T. Lee (Orchidaceae) in Japan and Korea is morphologically distinct from the type specimen of that species in the following points: flowers sparsely arranged (vs. closely arranged in the type); apex of anther cap mucronate (vs. beaked), lateral sepals twisting and enfolding the lip (vs. extending to the apex of the lip). Since these plants do not correspond with any known species, it is described as a new species, Liparis koreojaponica. The taxonomic status of L. koreana is reviewed.

A new species, Liparis purpureovittata (Orchidaceae), is described from central and northern Japan on the basis of both morphological and molecular characters. This species is distinguishable from the closely related Liparis kumokiri in the abruptly dilated labellum (vs. the gradually dilated labellum in L. kumokiri), the labellum recurved strongly in the middle (vs. the labellum strongly recurved in both the middle and distal part), and the column with rounded marginal wings (vs. angular marginal column wings).

A phylogeographic study of two species of Carpinus, C. japonica Blume and C. tschonoskii Maxim. (Betulaceae), based on the distribution patterns of their chloroplast DNA haplotypes, is reported. In Japan, these species are mainly distributed in Pacific-type deciduous broad-leaved forests. Using 439-440 and 627-629bp nucleotide sequences of noncoding regions of chloroplast DNA, we detected 5 and 6 haplotypes among 217 and 181 individuals sampled from 52 and 49 populations of C. japonica and C. tschonoskii, respectively. The geographic distribution patterns of the haplotypes were highly structured. We investigated the common phylogeographic patterns between the two species that would indicate the influence of common historical factors such as climate change since the last glacial maximum (LGM). Based on our results, we concluded that the Pacific-type Japanese deciduous broad-leaved forests were split into at least three refugia during the LGM. After the LGM, the species expanded to northern areas or moved to higher altitudes from each refugium, thus now occupying northeastern, central, and southwestern Japan.

Silene kiusiana (Caryophyllaceae) is an endangered perennial herb of semi-natural grasslands in Japan and Korea. To elucidate the genetic status of S. kiusiana in the Aso region, Kyushu, Japan, in which relatively large populations still remain, we analyzed the genetics of seven populations of S. kiusiana using five novel microsatellite markers. Each population of S, kiusiana has maintained relatively high genetic diversity (mean H_E = 0.791 and mean N_A =12.0). In addition, the values of pairwise F_<ST> indicating the degree of genetic differentiation among populations were significantly larger than zero between all pairs of populations, although the degrees of difference were small. The semi-natural grasslands in the Aso region have decreased in size over the past several decades, and thus habitat fragmentation may have caused genetic differentiation among populations of S. kiusiana. Although the populations of S. kiusiana have maintained relatively high genetic diversity, the genetic dynamics of S. kiusiana populations should be continuously monitored.

Polyploidization is an important evolutionary force in plant speciation, and elucidating the origin and evolutionary history of a particular polyploid is important for understanding the evolution and distribution of plants. We investigated the origin of the endangered tetraploid Adonis ramosa Franch., which is endemic to Japan. To clarify the origin and phylogenetic relationships among A. ramosa and diploid species, we conducted a phylogenetic analysis of the species of Adonis in Japan and Korea using chloroplast trnL-trnF spacer and nuclear ribosomal internal transcribed spacer (ITS) sequences. The sequences of the ITS region of showed that the sequences of A. ramosa were completely consistent with or quite similar to those of A. amurensis Regel & Radde of Hokkaido, Japan, and clearly differed from those of A. amurensis in Korea and three diploid species from Japan and Korea. The results suggest the possibility that A. ramosa is an autotetraploid, and the diploid progenitor is A. amurensis from Hokkaido. The distribution pattern of the ITS haplotype of A. ramosa also indicates range expansion of A. ramosa from northern to southern Japan Ecological divergence and adaptation to new habitats after polyploidization are likely to increase the survival of A. ramosa and enable it to widely colonize in new environments in Japan.

Veronicastrum sibiricum var. zuccarinii (Plantaginaceae) is an endangered perennial herb native to semi-natural grasslands in southern Korea and Kyushu, Japan. To elucidate the genetic status of V. sibiricum var. zuccarinii in the Aso region of Kyushu, in which relatively large populations still remain, we analyzed seven populations using five novel microsatellite markers. Each population of V. sibiricum var. zuccarinii has a maintained moderate level of genetic diversity (mean H E = 0.596, mean N A = 5.6, and mean AR = 4.7). Pairwise F ST indicated significant genetic differentiation between all population pairs (0.065−0.309). The semi-natural grasslands in the Aso region have decreased in area over the past decades. Thus, habitat fragmentation may be causing genetic differentiation among populations of V. sibiricum var. zuccarinii. Although the populations of this variety have maintained a moderate degree of genetic diversity, the dynamics of gene flow among them should be continuously monitored.

We examined morphological differentiation and allozyme diversity in nine populations of Vincetoxicum katoi and its close relative V. yamanakae. Although, V. yamanakae has been distinguished from V. katoi in having a larger corolla and gynostegium, the principal component analysis based on 12 floral and three leaf characters did not support the distinction of the two taxa. The six allozyme loci examined revealed that population of V. katoi and V. yamanakae maintained high genetic diversity (P=72.1, A=2.40, h=0.313). The nine populations were clustered into three regions (Kanto-Tokai; middle Kiniki; and Kii-Shikoku) based on genetic differences, contradicting the current taxonomic treatment. Considering the evidence, it is appropriate to treat V. yamanakae as a synonym of V. katoi. The high genetic differentiation among regions suggested that disjunct distribution of the V. katoi-V. yamanakae complex might reflect the persistence of refugia since the last glacial period. In particular, the middle Kinki (Hyogo) population is located outside of known evergreen forest refugia, suggesting that it might have survived during the glacial period within cooler vegetative zones, such as in temperate forests, whereas the populations on the Pacific-side retreated to warm-temperature coastal refugia.

Phylogenetic relationships among the members of the epiphytic genus Yakushimabryum H. Akiyama et al. and related genera (Pylaisiadelphaceae) were studied using cpDNA (rbcL, rps4, and trnL-F) gene sequences as well as morphological features. Present analyses (1) suggested the presence of three clades in the Pylaisiadelphaceae and Sematophyllaceae (Pylaisiadelphaceae s. str., Sematophyllaceae s. str. + Heterophyllioideae, and Clastobryum), and (2) confirmed the distinctive generic status of Yakushimabryum from other members of the family. One new genus Orientobryum H. Akiyama and two new species O. ovalifolium H. Akiyama and Y. brevigemmium H. Akiyama were recognized and described. In addition, a close inter-relationship of Pseudotrismegistia undulata (Broth. & Yasuda) H. Akiyama & Tsubota and Heterophyllium amblystegum (Mitt.) J. Yu et al. (= Mastopoma subfiliferum Horik. & Ando) was confirmed. Four new combinations, O. oligonema (Cardot & P. de la Varde) H. Akiyama (≡ Clastobryum oligonema Cardot & P. de la Varde), Y. subintegrum (P. Tixier) H. Akiyama (≡ Aptychella subintegra P. Tixier), Y. tonkinense (Broth.) H. Akiyama (≡ Clastobryum tonkinense Broth. & Paris), and Pseudotrismegistia amblystega (Mitt.) H. Akiyama (≡ Stereodon amblystegus Mitt.) were proposed.

Potamogeton ×tosaensis Kadono, Horii & Yamanouchi (Potamogetonaceae) is described here as a new hybrid. Although similar to P. octandrus, it is morphologically distinguishable in several traits. Most conspicuous of these are the connate stipules and fusiform turion bearing one acerose leaf. The plants were found only in and around Kochi City, Shikoku. Potamogeton octandrus and P. pusillus are the putative parents.
