Agrobacterium-mediated transformation is a key innovation for plant breeding, and routinely used in basic researches and applied biology. However, the transformation efficiency is often the limiting factor of this technique. In this study, we discovered that oxicam-type nonsteroidal anti-inflammatory drugs, including tenoxicam (TNX), increase the efficiency of Agrobacterium-mediated transient transformation. TNX treatment increased the transformation efficiency of Agrobacterium-mediated transformation of Arabidopsis thaliana mature leaves by agroinfiltration. The increase of efficiency by TNX treatment was not observed in dde2/ein2/pad4/sid2 quadruple mutant, indicating that TNX inhibits the immune system mediated by jasmonic acid, ethylene, and salicylic acid against to Agrobacterium. We also found that TNX-treatment is applicable for the transient expression and subcellular localization analysis of fluorescent-tagged proteins in Arabidopsis leaf cells. In addition, we found that TNX increases the efficiency of Agrobacterium-mediated transient transformation of Jatropha. Given that treatment with oxicam compounds is a simple and cost effective method, our findings will provide a new option to overcome limitations associated with Agrobacterium-mediated transformation of various plant species.

This paper discusses the role played by enjoyment in class of "Nihon jijyo" ("Studies of Japanese culture and society" for international students) at a university. First, the significance of incorporating experiential learning and audio-visual aids into the class to facilitate active learning is discussed. Next, active learning activities — namely, watching "Dahtsu no Tabi" ( a Japanese TV show: "Journey by darts"), and going to a Japanese festival — were conducted, and ways to enhance students' enjoyment through the activities were suggested. Finally, the role of enjoyment in enhancing students' learning motivation are discussed.The results shows that students' enjoyment was enhanced, and they have become more active learners after participating in the activities. And then, after experiential learning, they have wished to get many new experiences.

In plant cells, the trans-Golgi network (TGN) is known to act as the early endocytic compartment, whereas RAB5-localizing multivesicular endosomes (MVEs) act as the later compartment. Land plants and certain green algal species possess plant-unique RAB5 homologs (ARA6/RABF1 in Arabidopsis thaliana) in addition to the orthologs of animal RAB5 (RHA1/RABF2a and ARA7/RABF2b in A. thaliana), and these two RAB5 members reside in substantially overlapping but different subpopulations of MVEs. Several studies indicate that the TGN and MVEs are closely related; however, the distribution of the two RAB5 groups in relation to the TGN remains elusive. Here, we quantitatively showed that ARA6 and ARA7 are closely associated with the TGN, and the subpopulation of ARA6 and ARA7 overlaps with the TGN in the root epidermal cells of A. thaliana.
