Here, we report details of a new infectious disease in wild-caught Japanese fire-bellied newts (Cynops pyrrhogaster), a Near Threatened species. Skin lesions consisting of numerous masses were found in the animals near Lake Biwa, Shiga Prefecture, Japan. The gross appearance of the skin lesions showed blister-, cyst-, and/or tumor-like morphology. Various sizes of skin lesions were observed on their entire body surface. Histologically, spherical basophilic cysts, including numerous spores, were observed in the dermis layer. Ultrastructural analysis indicated the presence of main bodies of flagellated zoospores within the spores. While 18s rRNA gene sequencing indicated that the skin lesions were due to dermocystid infection. To our knowledge, this is the first report of dermocystid infection in this amphibian in Japan. Further studies are needed to prevent epidemics and to establish diagnostic and treatment methods.

Tick-borne encephalitis virus (TBEV) is a zoonotic agent causing severe encephalitis in humans. Rodent species which are potential hosts for TBEV are widely distributed in various regions in Japan. In this study, we carried out large-scale epizootiological surveys in rodents from various areas of Japan. A total of 931 rodent and insectivore sera were collected from field surveys. Seropositive rodents against TBEV were found in Shimane Prefecture in Honshu, and in several areas of Hokkaido Prefecture. These results emphasize the need for further epizootiological and epidemiological research of TBEV and preventive measures for emerging tick-borne encephalitis in Japan.

Rabies is a zoonotic disease caused by the Lyssavirus rabies virus (RABV) that can infect most mammals, including humans, where it has a case-fatality rate of almost 100%. Although preventable by vaccination, rabies causes c. 59,000 human fatalities every year worldwide. Thus, there exists an urgent need to establish an effective therapy and/or improve dissemination of vaccines for humans and animals. These outcomes require greater understanding of the mechanisms of RABV pathogenesis to identify new molecular targets for the development of therapeutics and/or live vaccines with high levels of safety. Importantly, a number of studies in recent years have indicated that RABV specifically suppresses host immunity through diverse mechanisms and that this is a key process in pathogenicity. Here, we review current understanding of immune modulation by RABV, with an emphasis on its significance to pathogenicity and the potential exploitation of this knowledge to develop new vaccines and antivirals.

Although rabies still kills many people, the global eradication of human rabies is considered to be feasible. Progress towards eradication may differ among regions with differing socio-economic statuses; therefore, states that successfully eradicate this disease must be vigilant for rabies re-emergence. Here, we discuss challenges that remain concerning current rabies prevention measures and risk assessment results concerning possible rabies introduction and spread in rabies-free Japan. We summarize the preventative measures undertaken by representative rabies-free countries and regions. Our risk assessment results show that the risk of rabies reintroduction under current circumstances is very low, and that subsequent spread of the disease would be minimal because of quite low value of basic reproduction number. Similar assessments conducted in other rabies-free areas also showed limited risks of introduction. The majority of rabies-free countries maintain their rabies-free status through strict import quarantine of carnivorous animals, efficient surveillance of animal rabies including wildlife, quick emergency responses, and raising public awareness of the disease. To maintain the current rabies-free status in Japan, we strongly recommend maintaining the current quarantine system and reinforcing stakeholder compliance for those involved in international movement of dogs. Moreover, sustainable surveillance systems targeting wildlife are indispensable.

The present report describes a newborn calf with spina bifida that presented with a giant mass of the lumbar region, as well as subsequent gross, histological, and immunohistochemical examinations. A malformed Japanese black calf (estimated weight = 20 kg) was euthanized immediately after birth. A gross evaluation revealed a giant mass (approximately 60 cm × 30 cm × 15 cm) covered by the hair coat in the lumbar region and connected with the hair coat of the trunk. The mass surface was divided by a deep polygonal groove and externally resembled a lobulated kidney. Histology and immunohistochemistry revealed that the giant mass comprised a vessel, bronchiolus lined with cuboidal epithelium, and small alveolus. Bone bleaching revealed various abnormalities, including spina bifida, vertebral fusion, vertebral deformity, vertebral malformation, vertebral scoliosis, and coxal bone malformation. Following a suggestion that the giant lumbar region mass was occupied by lung tissue, this case was considered to involve an asymmetric conjoined duplicitas that resulted in a very rare dichotomous spondylosis malformation.
