Surface properties of cast films of poly(lactic acid) (PLA) containing 1 wt% of several glycolipid-type biosurfactants (BSs) were investigated. The wettability of PLA films containing a homologue of mannosylerythritol lipids (MEL-B), lactone-form sophorolipid (LSL), or cellobiose lipid (CL) was drastically higher than that of untreated PLA and several synthetic surfactants-containing PLA. Surface wettability was also dependent on the hydrophilicity of the substrate used during solvent casting of the PLA films. The wetting behavior of the opposing sides of MEL-B-containing films prepared on glass substrates differed significantly; the contact angle on the side of the film that had been in contact with the glass surface was significantly lower than that obtained on the side of the film that had been in contact with air. Time-of-flight secondary ion mass spectrometry (TOF-SIMS) analysis results showed that the MEL in MEL-B-containing thin PLA cast films was localized to a thin surface layer. These results suggest self-assembly of MEL-B and micro-phase separation between the PLA matrix and MEL-B domains. This resulted in the localization and orientation of MEL-B at the surface of the cast PLA film, which determined its specific wetting behavior.

We examined effects of a major lipotrope, myo-inositol, on the expression of primary glycolytic (glucokinase and phosphofructokinase) and fructolytic enzyme (ketohexokinase [KHK] and aldolase B) genes in the livers of rats fed a control diet, high-sucrose diet, or high-sucrose diet supplemented with 0.5% myo-inositol for 14 d. Supplementation with myo-inositol decreased the hepatic expression of fructolytic enzyme genes, but not that of glycolytic enzyme genes, and the levels of triglycerides, fatty acid synthase, and KHK proteins in high-sucrose diet-induced fatty liver. The study results suggest that myo-inositol represses primary fructlysis, but not glycolysis, in high-sucrose diet-induced fatty liver.

The objective of this study was to obtain optimization results from the biological hydrolysis of Oil Palm Empty Fruit Bunches (OPEFB) using Aspergillus niger (A. niger) BIOTROP 2173 isolated from grain. Optimized hydrolysis parameters include temperature, pH and time. The hydrolysis process was carried out by growing A. niger on OPEFB powder (± 30 mesh) through two schemes, namely hydrolysis on OPEFB pretreatment with 10% NaOH and hydrolysis on OPEFB non-pretreatment. The optimization results show that the best hydrolysis process of A. niger BIOTROP 2173 occurs in OPEFB pretreatment. The optimum conditions for temperature, pH and time obtained are 40°C, 6 and 24 hours, respectively. Although the amount of reducing sugar produced was lower than the OPEFB non-pretreatment, the performance of the cellulase enzyme during the hydrolysis process of OPEFB pretreatment was very good, with a fast hydrolysis rate. These results indicate that the performance of A. niger BIOTROP 2173 in the hydrolysis process is influenced by the pretreatment stage. The optimum conditions obtained then became a reference in the production of reducing sugar based on A. niger BIOTROP 2173. The amount of reducing sugar produced from OPEFB pretreatment was 0.94 mg.mL-1, while for OPEFB non-pretreatment was 15.83 mg.mL-1.

The oil recovery from Alyanak apricot kernel was 36.65% in control (unroasted) and increased to 43.77% in microwave-roasted kernels. The total phenolic contents in extracts from apricot kernel were between 0.06 (oven-roasted) and 0.20 mg GAE/100 g (microwave-roasted) while the antioxidant activity varied between 2.55 (oven-roasted) and 19.34% (microwave-roasted). Gallic acid, 3,4-dihydroxybenzoic acid, (+)-catechin and 1,2-dihydroxybenzene were detected as the key phenolic constituents in apricot kernels. Gallic acid contents varied between 0.53 (control) and 1.10 mg/100 g (microwave-roasted) and 3,4-dihydroxybenzoic acid contents were between 0.10 (control) and 0.35 mg/100 g (microwave-roasted). Among apricot oil fatty acids, palmitic acid contents ranged from 4.38 (oven-roasted) to 4.76% (microwave-roasted); oleic acid contents were between 65.73% (oven-roasted) and 66.15% (control) and linoleic acid contents varied between 26.55 (control) and 27.12% (oven-roasted).

The benefit of fish oil to health has been widely recognized because of the high contents of functional EPA (20:5n-3) and DHA (22:6n-3) in the oil; however, the application of fish oil has been limited by its high susceptibility to oxidation. This review reports the characteristics of EPA and DHA oxidation compared with those of other fatty acids such as linoleic acid (18:2n-6). In addition, effective approaches to protect against oxidation are discussed, focusing on the unique antioxidant potential of amine compounds. Finally, the exceptionally high oxidative stability of EPA and DHA in biological systems is discussed. Understanding the protective mechanism against EPA and DHA oxidation in such systems may be useful for the development of an effective antioxidant procedure for fish oil that is rich in EPA and DHA.

This study investigates the application of green technologies (microwave and ultrasound pretreatment) in the extraction of Moringa oleifera Lam. seed oil and its effects on oil expression, oil quality, and bioactive component. Moringa seeds were pretreated with microwave (90 W, 60 s) or ultrasound (50 W, 1 h) before mechanical expression. A separate group received no pretreatment before oil extraction. Oils from these groups were then compared. Results show that oil yield increased with ultrasound pretreatment (1.24%) and significantly increased with microwave pretreatment (3.11%). For oil flow rate, the microwave and ultrasound pretreatment resulted in faster extraction (7.67 and 6.93 kg/h respectively) as compared with the control (6.51 kg/h). For physicochemical parameters, the microwave and ultrasound group had significantly less free fatty acids and significantly greater unsaponifiable matter as compared with the control. For fatty acid composition, results show that moringa seeds procured from Davao Oriental had greater oleic acid content (~77%) as compared with those reported by other literature. For phytosterol content, the predominant phytosterols found were β-sitosterol, stigmasterol, and campesterol. Microwave and ultrasound pretreatment significantly increased total phytosterol (680.58 and 369.32 mg/kg respectively) as compared with the control (72.69 mg/kg) due to the mass transfer of the phytosterols. Microwave and ultrasound pretreatment also led to stigmastanol formation. For antioxidant activity, a comparison of both DPPH and FRAP assays depicts that the microwave group exhibited the best overall antioxidant activity. Lastly, for oil stability, a lower peroxide value was found in the microwave and ultrasound groups across time intervals, which may be attributed to their antioxidant activity. In summary, ultrasound and microwave pretreatment can improve oil expression, oil quality, and bioactive content of the mechanically expressed moringa oils.

The present study provides valuable data that the herbal water byproduct of Japanese peppermint, produced during the steam distillation extraction of an essential oil, can be utilized as an antibacterial agent. The major ingredient in the herbal water from Japanese peppermint ‘Hokuto’ was menthol, with a concentration close to its water solubility. The herbal water produced showed excellent antibacterial efficacy against typical gram-negative and gram-positive bacterial strains of Escherichia coli and Staphylococcus aureus, respectively, and the antibacterial efficacy was maintained even when the herbal water was diluted up to an appropriate concentration of 50%. The efficacy of the herbal water against E. coli was higher than that against S. aureus, which is likely because of the difference in the efficacy of menthol against these two different bacterial strains. The excellent antibacterial efficacy of the herbal water is mainly attributed to the function of menthol, while other trace ingredients also contributed to the antibacterial efficacy. The Japanese peppermint herbal water byproduct, generally treated as industrial waste and disposed, can be easily commercialized as an antibacterial agent if efforts are made to maintain a constant menthol concentration throughout the steam distillation essential oil extraction process.

The marine eukaryotic microheterotroph thraustochytrid genus Aurantiochytrium is a known producer of polyunsaturated fatty acids, carotenoids, and squalene. We previously constructed a lipid fermentation system for Aurantiochytrium sp. strains using underutilized biomass, such as canned syrup and brown macroalgae. To improve the productivity, in this study, Aurantiochytrium sp. RH-7A and RH-7A-7 that produced high levels of carotenoids, such as astaxanthin and canthaxanthin, were isolated through chemical mutagenesis. Moreover, metabolomic analysis of the strain RH-7A revealed that oxidative stress impacts carotenoid accumulation. Accordingly, the addition of ferrous ion (Fe2+), as an oxidative stress compound, to the culture medium significantly enhanced the production of astaxanthin by the mutants. These approaches improved the productivity of astaxanthin up to 9.5 mg/L/day at the flask scale using not only glucose but also fructose which is the main carbon source in fermentation systems with syrup and brown algae as the raw materials.

The skin of the walnut kernels can get dark during the pre- and post-harvest stages of the production. Dark kernels are less palatable for most consumers but are still edible and maybe preferable, especially in the ground form, for industrial use. In this study, we investigated the differences between oil oxidation indexes, fatty acid and tocopherol compositions of the oils, total polyphenol contents and antioxidant capacities of the extracts of light and dark walnuts. In addition, we evaluated the effects of packaging under nitrogen and vacuum-packaging techniques and storage temperature on these characteristics of both light and dark walnuts during storage for 6 months. Peroxide values and free fatty acid contents of all samples were higher at the end of storage compared to initial values, being more noticeable at 20°C than at 4°C. Increases in the free fatty acid contents were quite higher in dark walnuts than the light ones (6.1 and 3.1 fold, respectively) and the highest values of conjugated diene and peroxide were determined in the samples packaged under air and stored at 20℃. Dark walnuts had lower total phenolic, α- and γ-tocopherol contents and antioxidant activities compared to the light ones. Total phenolic and tocopherol contents decreased over time. We conclude that due to the antioxidant compounds in the fruit, oxidation parameters of dark walnuts are still acceptable at the end of 6-month storage even in the packages with air and at non-refrigerated conditions.

Cancer is the world’s biggest health problem and cancer-induced mortality happened all over the planet after the heart disease. The present study was to scrutinize the anti-leukemia effect of diosmin against Dalton Ascitic Lymphoma (DAL) induced leukemia in mice. DAL cell was used for induction the solid tumor. Body weight, life spans, tumor volume and mean survival time was estimated. Antioxidant, biochemical and pro-inflammatory cytokines were estimated. Diosmin showed the cell viability effect at dose dependent manner against the both cell lines. DAL induced solid tumor mice showed the decreased body weight, mean survival days, non viable cell count and increased the tumor volume, viable cell count and diosmin significantly (p < 0.001) reverse the effect of DAL. Diosmin significantly (p < 0.001) altered the hematological, differential leukocytes, antioxidant, biochemical, pro-inflammatory cytokines at dose dependently. Collectively, we can say that diosmin might alter the DAL induced abnormality via antioxidant and anti-inflammatory effects.

The aim of the survey was to study the effect of heat stress (HS) on health lipid indices in milk of Holstein-Friesian cows. The study was conducted in a cattle farm with Holstein-Friesian cows in the region of Karnobat (Southeastern Bulgaria) in 2018. Cows were housed in semi-open free stall dairy barn, fed year-round ad libitum with a total mixed ration. The study included 22 cows on different parities studied in two periods - at thermo-neutral environment conditions and at heat stress, respectively, May and August. Extraction of milk fat was performed by the Rose-Gottlieb method. Conditions of HS lead to changes in the values of health lipid indices associated with a decrease in the values of Atherogenic index (AI), Thrombogenic index (TI), Lipid Preventive Score (LPS) and Desaturase (18) index (DI 18) and an increase in Health promoting Index (HPI), polyunsaturated fatty acids/saturated fatty acids (PUFA/SFA), unsaturated fatty acids/saturated fatty acids (UFA/SFA), mono unsaturated fatty acids (MUFA), Desaturase (16) index (DI 16) and hypocholesterolaemic/hypercholesterolaemic ratio (h/H). Increasing the Temperature-humidity index (THI) above 72, results in a decrease in the AI values and an increase in those of the PUFA/SFA. The values of health lipid indices showed a moderate positive correlation with those of THI (PUFA/SFA - 0.36) with rectal temperature (h/H, MUFA/SFA, UFA/SFA) rp from 0.36 to 0.37, and with respiratory rate (h/H, PUFA/SFA), rp of 0.33 and 0.31, respectively. Under the influence of heat stress, changes in the metabolic processes occur in the body of dairy cows leading to changes in the fatty acid content of milk related to the improvement of health lipid indices in terms of human health due to an increase in UFA and reduction in SFA.

We discovered that Candida floricola ZM1502 is capable of selectively producing the promising hydrophilic biosurfactants, acid-form sophorolipids (SLs), from glycerol. However, productivity was very low (approximately 3.5 g L–1) under the initial culture conditions. Here, we describe the design of culture medium for abundant production of acid-form SLs by C. floricola ZM1502 using waste glycerol and hydrophobic substrates in order to develop a method for SL production and disposal of waste glycerol produced by oleo-chemical industries. Urea provided the best nitrogen source for acid-form SL production from glycerol among four nitrogen sources tested [urea, NaNO3, NH4NO3, and (NH4)2SO4]. Among carbon sources we compared, hydrophobic substrates (soybean oil and oleic acid) led to productivities of approximately 20 g L–1, indicating that hydrophobic substrates provided fatty acid moieties for SL production. Addition of olive oil and oleic acid to waste glycerol enhanced acid-form SL production to 42.1 ± 0.9 and 37.5 ± 3.4 g L–1, respectively. To develop a potential industrial process, we explored other suitable hydrophobic substrates for SL production, which were obtained on site from oleo-chemical industries. Alkyl C18 esters (Pastell M-182), along with waste glycerol, increased acid-form SL production to 48.0 ± 3.4 g L–1 over a 7-d period. Furthermore, we demonstrated abundant production of acidic SLs at the mini-jar fermenter scale, obtaining 169 g L–1 over 180 h using a fed-batch cultivation technique. Efficient acid-form SL production by C. floricola could have a great impact on the development of bio-industrial processes using waste glycerol as a substrate.

Palmitoleic acid shows a variety of beneficial properties to human health. In this study, enrichment of palmitoleic acid from sea buckthorn pulp oil by two-step solvent crystallization and molecular distillation was investigated. Sea buckthorn pulp oil was first converted to its corresponding mixed fatty acids (SPOMFs) containing 27.17% palmitoleic acid. Subsequently, the effects of various factors on crystallization (i.e., crystallization temperature, type of solvent, ratio of SPOMFs to solvent (w/v), crystallization time) and molecular distillation (distillation temperature) were assessed on a 5-g scale. It was found that optimal primary crystallization conditions were a 1:15 ratio of SPOMFs to methanol (w/v), -20°C and 12 h. Secondary crystallization conditions were set to a 1:4 ratio of methanol to palmitoleic acid product obtained from the first step crystallization to methanol (w/v), -40°C and 6 h. For further purification of palmitoleic acid by molecular distillation, the optimal distillation temperature was determined to be 100°C. After purification by crystallization and molecular distillation under the optimal conditions, the final product consisted of 54.18% palmitoleic acid with an overall yield of 56.31%. This method has great potential for adoption by the food and medical industries for the preparation of palmitoleic acid concentrate for nutritional studies.

The ruling that partially hydrogenated oils (PHO) are no longer “generally recognized as safe (GRAS),” has accelerated the replacement of PHO ingredients with fat alternatives having increasingly lower or no trans fat content. In the present study, we developed a Fourier-transform infrared (FTIR) spectroscopic procedure in conjunction with multivariate partial least squares regression (PLSR) and found it suitable for the accurate prediction of low (0.5%) total trans fat content, as percentage of total fat, measured as fatty acid methyl esters (FAME), in the lipids extracted from 24 representative fast foods. This multivariate data analysis approach is relevant because the precision of the current univariate FTIR official method (AOCS Cd 14-09) is reportedly poor below 2% of total fat, while PLSR has allowed us to accurately predict the concentration of low trans fat in fast foods. The performance of a portable FTIR device was also evaluated and compared to that of a benchtop FTIR spectrometer. For both infrared data sets, PLSR-predicted concentrations of total trans FAME, ranging from approximately 0.47% to 11.40% of total FAME, were in good agreement with those determined by a primary reference gas chromatography (GC) method (R2>0.99); high prediction accuracy was also evidenced by low root mean square error of cross-validation (RMSECV) values. The lowest RMSECV error of 0.12% was obtained with the portable device. The lowest total trans FAME concentration, determined by GC to be 0.42%, was accurately predicted by the portable FTIR/PLSR procedure as 0.47% of total FAME.

Curcumin is a bioactive compound with proven antioxidant and anti-inflammatory activities, but has low water solubility and dermal absorption. The inflammatory process is considered as the biological response to damage induced by various stimuli. If this process fails to self-regulate, it becomes a potential risk of cancer. The objective of this work was to evaluate the anti-inflammatory activity of curcumin administered to mice with induced atrial edema using two topical vehicles: organogels and O/W-type nanogels at pH 7, Organogels and O/W-type nanogels at pH 7 were prepared, characterized and the anti-inflammatory activity was assessed. A histopathological analysis of mouse ears was performed and two gel formulations were selected. Thermograms of organogels indicated that increasing the gelling agent improved the stability of the system. Deformation sweeps confirmed a viscoelastic behavior characteristic of gels in both systems. During the anti-inflammatory activity evaluations, the nanogels demonstrated greater activity (61.8 %) than organogels; Diclofenac® (2-(2,6-dichloranilino) phenylacetic acid), used as a control medication achieved the highest inhibition (85.4%); however, the drug produced the death of 2 (40%) of the study subjects caused by secondary adverse events. Histopathological analysis confirmed the data.

Although extracellular carbonylated proteins (CPs) are found at higher levels in sun-exposed skin, their impact on the cellular functions of fibroblasts and their involvement in the progression of photoaging skin are not fully clarified. In our previous study, we reported that extracellular CPs increase levels of intracellular oxidative stress and result in the accumulation of newly synthesized CPs in normal human dermal fibroblasts (NHDF). Furthermore, fibroblasts exposed to CP-BSA, which is a model of extracellular CPs, had upregulated expression levels of mRNAs encoding matrix metalloproteinase-1 (MMP-1) and interleukin-8/CXCL8 (IL-8/CXCL8). These facts suggested the possibility that extracellular CPs induce a fragile structure in the dermis through the degradation of collagen and elastin. The purpose of this study was to characterize the efficacy of natural carotenoids, such as astaxanthin analogs, produced by Hematococus pluvialis (CHPs) to improve the impaired functions of fibroblasts exposed to CPs. CHPs suppressed the intracellular CP levels elevated by CP-BSA, restored mRNA expression levels of factors involved in the formation and assembly of collagen and elastin fibers and improved the formation of those fibers impaired by CP-BSA. We conclude that CHPs function as antiaging substances due to their restoration of the impaired formation of collagen and elastin fibers caused by extracellular soluble CPs.

The lipid metabolism disorder is the key role of Nonalcoholic fatty liver disease (NAFLD). Selenoprotein P plays an important role in the pathological process of lipid accumulation. Coix lacryma-jboi seed oil (CLSO) is an active component extracted from Coix lacryma-jobi seed (CLS) which has been found to be effective of reducing blood fat and antioxidative. But the effect and mechanism of CLSO on NAFLD are not clear. The aim of this study was to explore the therapeutic effect and mechanism of CLSO in the treatment of NAFLD. Our result showed that CLSO decreased the liver/body weight ratio, lowered the total cholesterol (TC) and triacylglycerol (TG), and elevated the high density lipoprotein (HDL) in serum. CLSO reduced the lipid deposition in the liver of NAFLD rats. In addition, CLSO could bring down the abnormal expression of superoxide dismutase (SOD) and malondialdehyde (MDA). Moreover, CLSO significantly declined the liver apolipoprotein E (apoE), apolipoprotein E receptor (apoER) and selenoprotein P 1 (SePP1) expression. In vivo, CLSO decreased the lipid droplets and TG level, reduced the protein expression of SePP1, apoER, phosphor-adenosine 5′-monophosphate (AMP)-activated protein kinase (p-AMPK) in the cytoplasm of HepG2 cells induced by oleic acid and palmitic acid (OP). At the same time, lipid accumulation was observed in the Sepp1 high expression cells induced by endoplasmic reticulum (ER) activator tunicamycin (Tm). CLSO could identically reduce the protein expression of SePP1, apoER, p-AMPK in the cytoplasm of HepG2 cells induced by Tm. This result not only proved the CLSO had therapeutic effect on NAFLD, but also confirmed its mechanism associated with degrading the phosphorylation of adenosine 5′-monophosphate (AMP)-activated protein kinase (AMPK) which led to the decrease of the expression SePP1/apoER2 in order to reduce lipid accumulation. The study suggests CLSO has great medicinal value in treating NAFLD besides its edibility.

This report focuses on acceleration of the recombination of lophyl radicals with a lophine dimer derivative by forming molecular assemblies. A newly synthesized cationic amphiphilic lophine dimer formed molecular assemblies with a diameter of ~220 nm in an aqueous medium. When the molecular assemblies were formed, the rate of recombination of lophyl radicals, produced by ultraviolet light irradiation, was accelerated 50,000-fold compared to that in an organic solvent. The rate enhancement is likely derived from the short diffusion distance of the lophyl radicals in the molecular assemblies. These results revealed that accumulation of the lophine dimers via self-assembly remarkably accelerated the recombination of lophyl radicals. This novel photoisomerization system could rapidly control the interfacial properties or morphologies of molecular assemblies when used for applications, such as efficient delivery of drugs or active components.

Colletotrichum nymphaeae is the causal agent of strawberry anthracnose, which is one of the most important disease affecting strawberry plant in Iran. This research aimed to apply the selected plant essential oils (EOs) such as Achillea millefolium, Mentha longifolia, and Ferula kuma to the management of strawberry anthracnose disease under in vitro, in vivo, and greenhouse conditions. In vitro tests indicated that all the EOs and fungicide were able to inhibit mycelial growth and conidial germination of the pathogen. Scanning electron microscopy (SEM) revealed that EOs significantly suppressed the mycelia growth and caused a change in morphology of fungal mycelia. The severity of strawberry anthracnose disease was significantly (p ≤ 0.05) reduced by all EOs under in vivo and greenhouse conditions. Results of all experiments showed that M. longifolia EO was the best EO to control C. nymphaeae. Also, EOs almost reduced weight loss and preserved firmness, ascorbic acid, total phenol, antioxidant activity (DPPH), and enzyme peroxidase activity in treated fruit. Moreover, EOs preserved the sensory quality of strawberry fruit during the storage period so that there were no significant differences between treatments (EOs) in their appearance, flavor, odor attributes, and overall evaluation compared to the control. Our results indicate that EOs are excellent bio-fungicides for the management of strawberry anthracnose.

Fatty acids and their derivatives are interesting cosmetic ingredients because they show the selective antibacterial activity against Staphylococcus aureus (S. aureus). However, the antibacterial activity in mixed systems containing several active ingredients is unclear because previous studies focused antibacterial systems containing one kind of fatty acid. In the present study, the minimal inhibitory concentration (MIC) and the fractional inhibitory concentration (FIC) were evaluated for myristic acid/lauric acid, myristic acid/palmitoleic acid, and myristic acid/lactic acid mixed systems to show the effect of the coexisting components on the selective antibacterial activity of myristic acid. In the myristic acid/palmitoleic acid mixed system, the antibacterial activity against S. aureus was enhanced by additive effect, whereas the antibacterial activity was not observed against S. epidermidis. On the other hand, the myristic acid/lauric acid mixed system showed antibacterial activity against S. epidermidis: Lauric acid impaired the selectivity of antibacterial activity of myristic acid. These results suggest that the selective activity of myristic acid varies with the additives. The present findings are useful for designing formulations of cosmetics and body cleansers containing myristic acid.
