Leucomycin A<SUB>3</SUB> (I) was oxidized with active manganese dioxide to dehydroleucomycin A<SUB>3</SUB> (II) which was identical with magnamycin B. Acid treatment of I yielded demycarosylleucomycin A<SUB>3</SUB> (VII), which was converted to a triacetate showing mass spectrum M<SUP>+</SUP> (m/e 725), and this observation confirmed the structure of I. The diene system of I was assumed to have a trans-trans configuration on the basis of its NMR spectrum, and a full structure of I was proposed with stereochemistry.

A series of new antibiotics, Kinamycin A, B, C, and D which are mainly effective against gram-positive bacteria, were extracted with chloroform from the broth filtrate of Streptomyces murayamaensis sp. nov. HATA et OHTANI. Kinamycin C (I), C<SUB>24</SUB>H<SUB>20</SUB>O<SUB>10</SUB>N<SUB>2</SUB>, m/e 496 (M<SUP>+</SUP>), was determined to have an 8-hydroxynaphthoquinone skeleton, nitrile, acetoxyl, and tertiary methyl groups from its ultraviolet (UV), infrared (IR), and nuclear magnetic resonance (NMR) spectra, and was further found to have a unique structure of N-C-N from some chemical reactions and X-ray diffraction. Structure relationship among I and other components A, B, and D was assumed to be due to the difference in the number and position of the acetoxyl group from analyses of IR, NMR, and mass spectra of their acetylated compounds. The antimicrobial activity of the four kinamycins increases with the decreasing number of acetoxy group, in the order of kinamycin C, A, D, and B. In addition, some derivatives obtained during structural studies on I were found to have nearly equal or increased antimicrobia1 activity compared with I and kinamycin D (XI).

We have been studied the relationship between the structures and the biological activities on 16-membered lactone ring macrolide antibiotics. The aglycone moiety from 16-membered macrolides has not been reported, but in the present series of work, we have chemically obtained the aglycone from leucomycin A_3 (LM A_3) (I). Treatment of (I) with m-chloroperbenzoic acid in CHCl_3 gave the N-oxide (II), which was refluxed with Ac_2O in CHCl_3 to obtaine aglycone, leuconolide-A_3 5,18-hemiacetal (III). In the above reaction, a neutral macrolide, 2'-acetyl 3'-desdimethylamino 3'-oxo LM A_3 (X) which 3'-dimethylamino group on mycaminose moiety was converted to ketone carbonyl was isolated from the same reaction product. Furthermore, (I) was reacted with Al-isopropoxide to give 9-dehydro 18-dihydro leucomycin A_3 (V). (V) was oxidized with m-chloroperbenzoic acid to N-oxide (VII), and(VII)was then treated with Ac_2O in CHCl_3 to obtain 9-dehydro 18-dihydro leuconolide-A_3 (VIII). In order to clarify the correlation between the structure and biological activity of mycaminose moiety, various derivatives were synthesized. The antimicrobial activities of the both glycone, (III) and (VIII) completly disappeared, and it was found that the decreasing of electro-density on dimethylamino group on mycaminose moiety resulted in the decrease of the activity.

In the course of various fermentations for the production of antibiotics by Streptomyces sp., glycerol, as well as glucose, has been found to be a preferred carbon source. In order to determine glycerol in the fermentation broth, a colorimetric method based on the procedures described by West was modified with respect to the condition of oxidation of glycerol by periodic acid. It has been found that, under a mild condition of periodic acid oxidation at 0 ℃ for 30 min, glycerol can be determined even when glucose is present. This method was applied in the practical production of three antibiotics : cycloserine, echinomycin, and streptomycin, and the following results were obtained : (1) The metabolism of glycerol was more active than that of glucose or starch; during each fermentation, the glycerol in the medium was nearly consumed after 45 hours.(2) The peak of antibiotic production in fermented broth appeared after the glycerol consumption was completed.
