Gut microbiota imbalance plays an important role in the pathogenesis of various diseases. Here, we determined microbe–microbe interactions and gut microbiome stability in a Japanese population with varying body mass indices (BMIs) and enterotypes. Using 16S ribosomal RNA gene sequencing, we analyzed gut microbial data from fecal samples obtained from 3,365 older Japanese individuals. The individuals were divided into lean, normal, and obese groups based on their BMIs. They were further categorized according to their gut microbiota enterotypes: Bacteroides (enterotype B), Prevotella (enterotype P), and Ruminococcus (enterotype R). We obtained data on different host factors, such as age, BMI, and disease status, using a survey questionnaire evaluated by the Mykinso gut microbiome testing service. Subsequently, we evaluated the co-occurrence network. Individual differences in BMI were associated with differences in co-occurrence networks. By exploring the network topology based on BMI status , we observed that the network density was lower in the lean group than that in the normal group. Furthermore, a simulation-based stability analysis revealed a lower resistance index in the lean group than those in the other two groups. Our results provide insights into various microbe–microbe interactions and gut microbial stability and could aid in developing appropriate therapeutic strategies targeting gut microbiota modulation to manage frailty.

Purpose: We have previously reported the physiological and biochemical effects of bath salts. In this study, we used bath additive containing artificial carbon dioxide and sodium chloride (S bath additive), and the acute effects of bathing once with S bath additive and chronic effects of daily bathing with S bath additive for 15 consecutive days were compared with the effects of bathing once in plain water. Improvement in 10 symptoms was investigated using a 5-point verbal rating scale (VRS).Methods: Nine patients aged 51-82 years (mean, 66.6 years) with cold intolerance and lower leg pain were treated with balneotherapy. Whole-body bathing at 40°C was performed for 10 min daily (day 1, bathing in plain water; days 2-16, bathing with S bath additive). On days 1, 2, and 16, changes in patient’s subjective symptoms were evaluated using a VRS ranging from -1 to +3.Results: Compared with bathing once in plain water, bathing once with S bath additive was improvement of coldness, body warmth, blood circulation promotion, limb warmth, lightness of foot, and coldness in limbs. Daily bathing with S bath additive for 15 days significantly improved all symptoms compared with bathing in plain water. A comparison between single and daily bathing with S bath additive showed that relieving fatigue and lumbago were significantly improved after consecutive bathing, demonstrating the benefits of long-term usage.Discussion: Bathing once with S bath additive had acute effects on symptoms associated with peripheral circulation, such as coldness in limbs, body warmth, and limb warmth. Daily bathing with S bath additive improved bone- and joint-related symptoms, such as lumbago and shoulder stiffness, demonstrating that the chronic effects of S bath additive differ from the acute ones. These results are consistent with previous studies on natural hot springs which found that the efficacy of short-term treatment was distinct from that obtained after >2 weeks of treatment.

Imbalance of the gut microbiota plays an important role in the pathogenesis of various diseases. Although many clinical studies have analyzed the gut microbiota, the definition of normal gut microbiota remains unclear. In this study, we aim to establish the average gut microbiota in the healthy Japanese population. Using 16S ribosomal RNA gene sequencing, we analyzed gut microbial data from fecal samples obtained from 6,101 healthy Japanese individuals. Based on their ages, the individuals were divided into three groups: young, middle-age, and old. Individuals were further categorized according to body mass index (BMI) into lean, normal, and obese groups. The α and β diversities in the old group were significantly higher than those in the young and middle-age groups. The Firmicutes/Bacteroidetes ratio of subjects in the obese category was significantly lower compared with those of subjects in the lean and normal categories in the young and middle-age groups. Genus Bacteroides was the dominant gut microbiota across all the BMI categories in all the age groups. Among the top ten genera, the abundances of Bacteroides, Bifidobacterium, Anaerostipes, Blautia, Dorea, Fusicatenibacter, Lachnoclostridium, and Parabacteroides were significantly lower in the old group than in the young and middle-age groups. The correlation network at the genus level revealed different microbe-microbe interactions associated with age and BMI. We determined the average Japanese gut microbiota, and this information could be used as a reference. The gut microbiota greatly differs based on the life stage and metabolic status of the host, and this gives rise to a variety of host–gut microbe interactions that can lead to an increased susceptibility to disease.

An antioxidant Was purified from human placenta extract (HPE) by using gel filtration, liquid-liquid extraction, silicagel column chromatography, and HPLC. The purified antioxidant was identified to be L-tryptophan (L-Trp). L-Trp showed higher inhibitory activity than mannitol and DMSO on the Fenton reaction-induced degradation of 2-deoxy-D-ribose. L-Trp also had much higher inhibitory activity on the cytochrome P-450-dependent lipid peroxidation than the previously identified antioxidants of HPE, L-phenylalanine, L-tyrosine and uracil. On the other hand, the inhibitory effect of L-Trp on the Fenton reactioninduced protein oxidation was smaller than that of uracil. These results suggest that L-Trp is a main antioxidant of HPE of which the effect is based on the suppression of lipid peroxidation in the oxidative stress status.

We report a time series (1948–2100) of global-scale meltwater from mountain glaciers and ice caps (MGI) estimated by the global glacier model HYOGA2. HYOGA2 calculates the temporal fluctuation of the mass balance for 24,234 individual glaciers worldwide. It covers 90% of the total glacier area, except for glaciers in Greenland and Antarctica. HYOGA2 also accounts for regionally distributed changes in glacier area and altitude associated with glacier retreat and advance. By computation of individual glacier changes, future dissipation and glacier mass and area changes can be simulated in the model. The cumulative volume loss of water between 1948 and 2005 was estimated to be 25.9 ± 1.4 mm sea level equivalent (SLE). A future projection under a high-emission scenario demonstrated significant losses of water from MGI equivalent to 60.3 ± 7.9 mm SLE between 1948 and 2060 and 99.0 ± 14.9 mm SLE between 1948 and 2099.
