Although much attention has been given to cell differentiation inducers as new types of anti-tumore agent, only a few studies has been reported on differentiation inducers from plant sorces. Therefore, we have searched for naturally occouring substances which induce differnciation of leukemia cells. From Condurango Cortex (Marsdenia condurango) (Asclepidaceae) and Periplocae Cortex (Periploca sepium B.) (Asclepidaceae), many kids of pregnan derivatives and some cardenolides. From withania (Withania somnifera) (Solanaceae) (using Indian market withania and withania cultivated at the medicinal plant garden of this university), more than thirty kinds of withanolides were isolated. From Physalis alkekengi (Solanaceae), physalin and neophysalin derivatives were isolated. Structural elucidation of these 60 kinds of steroid derivatives were carried out by means of spectral methods, especially using NMR spectroscopy including H-H COSY, H-C COSY, HMBC, NOE technics. These steroidal derivatives were tested on cell differentiatio inducing activity against mouse myeloid leukemia (M1) cells. Many kinds steroid derivatives showed the activity. Of these active compounds, some kinds of withanolids, 27, 33, 34 and 35 having 4β-hydroxy-5β,6β-epoxy-2-en-1-one structure of AB ring, showed potent differentiation inducing activity.

11-jodo-10-undecynoic acid (K-4172) was synthesised in our laboratory from 10-undecynoic acid reacting with iodine in presence of alkaline hydroxide. Fungistatic and bacteriostatic activities of K-4172 were examined. It showed significant antifungal activities, especially against Trichophytons and Crytococcus neoformans, but it had weak antibacterial activities.In in vivo test by experimental trichophytosis on guineapigs, it showed also significant therapeutic activities at the concentration of 0.5% in alcohol solution. It is very interest that in the in vivo test 0.5% tincture of K-4172 was more effective when it was applied in combination with 10% of 2, 4, 6-tribromophenyl caproate.Interperitoneal and oral LD50 of K-4172 dissolved in olive oil were 132 and 225mg/Kg respectively and intraperitoneal LD50 of its sodium salt was 134mg/Kg. Chronic toxicity test of K-4172 on mice and rat showed both almost no side effect when it was admidistrated intraperitoneally and orally. The authers persume from these data that K-4172 is safely available for treatment of human trichophytosis.

【背景・目的】血液透析中のトラブルの一つに透析回路の血液凝固がある.血液と空気の接触により凝固系が活性化され血栓を形成し凝固することが一因と考えられている.エアフリーチャンバとダイアフラム型圧力測定ポットを導入した透析回路「アーチループ」(AL)は従来型回路と比べ血液と空気の接触面積低減とプライミングボリューム低減という特徴を持つ.この回路の抗凝固特性を従来型回路と比較検討した.【対象・方法】当院通院中の維持透析患者12名を対象に,ALおよび従来型回路を用いて透析を同条件下で行い,透析開始3時間後の活性化凝固時間(activated clotting time:ACT)を測定し,両群で比較検討した.【結果】ACTは,従来型回路と比べALで有意に延長した(165.1±19.0 vs 153.2±15.2秒,p=0.002).止血不良などの出血傾向増強も認めなかった.【結論】アーチループ回路により回路内凝血の減少が期待できるがその効果はまだ限定的で汎用にはさらなる改良が望まれる.

Tensile tests were carried out on sintered Si<sub>3</sub>N<sub>4</sub> at room temperature under the ambient condition, and the influence of defects upon the fracture stress was discussed. As the surface of specimens was prepared carefully, fracture of 57 specimens out of 60 originated from the defects such as voids or inclusions which resulted from the sintering process. As a parameter to describe the size of the defect, &radic;<i>area</i> was used which is the square root of the area of a 3-dimensional defect projected to the direction of the maximum principal stress. The results obtained are as follows.<br>(1) &radic;<i>area</i> of defects which caused fracture nearly conforms to the first asymptotic distribution of the largest value and the fracture strength conforms to Weibull distribution.<br>(2) &radic;<i>area</i> is a good parameter to describe the size of the defect from the standpoint of fracture.<br>(3) Defects such as voids and inclusions act exactly as cracks of the same size.<br>(4) The fracture strength can be estimated by using the distribution of &radic;<i>area</i>, the fracture toughness and the mean grain size of Si<sub>3</sub>N<sub>4</sub>.
