リピッドマイクロスフェア (リピッドエマルション) 製剤は植物油 (大豆油) 中に付加価値の高い薬物を溶解あるいは分散させたo/w型エマルション製剤である。この製剤では油溶性あるいは疎水性の薬物を油相である分散相に効率良く溶解あるいは分散することができる。リピッドマイクロスフェアは医薬領域におけるドラッグデリバリーシステム, つまりドラッグキャリヤー製剤として, ある特別な (病巣) 部位のみに薬物を標的化, 薬物の徐放化, 持続化, 不安定な薬物の保護, 薬理作用の増強などのような利用が期待できる。そこで本総説では, 最近の注目すべき研究例を紹介しながら, リピッドマイクロスフェアの調製から応用までを詳述する。

A questionnaire survey of children and their parents visiting pharmacies dispensing medical insurance-covered drugs was conducted to gain information on medication methods.It was found that mothers played a very important role in children's drug taking and their skill in getting children to take drugs was important in achieving a high compliance rate.The dosage forms that seemed to be the most difficult to take were powders and granules,partly because of their frequent prescription,followed by liquids,solutions,and syrups.The reasons for difficulty in taking drugs were“bitterness",“smell",“coarseness",“sweetness",and“large volume" in decreasing order of frequency.To have their children take drugs,parents rarely used a commercial jelly for facilitating medication but often gave them ice cream,yogurt,in decreasing order of frequency.The infrequent use of commercial jellies for facilitating medication was despite the fact that the flavor had been changed to one liked better by children.The results of this survey suggest that jellies for facilitating medication for children may contribute more to improving compliance if the various needs of parents for improvements can be met.

The degree of lytic action due to interactions between nonionic surfactants and liposomes was determined in terms of affinity for lipid membranes. The solubilization potential of some surfactants toward liposome membranes was estimated based on physicochemical measurements such as fluorometry to detect the release of fluorescent markers (calcein) from liposomes, turbidimetry in suspensions and surface tensiometry. The lytic action of the alkyl poly (oxyethylene) ether (POE) toward lipid membranes was greatest when the alkyl and ethylene oxide chain lengths were 12 and 8, respectively. The lytic action of nonionic surfactants with various hydrophobic portions (hydrogenated caster oil POE ethers, cholesteryl POE ethers, nonylphenyl POE ethers and POE stearate) was found to depend on structure. Maximum activity in each homologous series was 1.2 I.O.B. (Inorganic Organic Balance).The critical micelle concentrations (cmc) of dodecyl POE (5) and POE (10) ether in the absence and presence of liposomes were determined using a surface tension meter. Addition of liposomes caused cmc of the surfactants to increase. Dodecyl POE (5) ether causes marker release at lower concentration than cmc without change in turbidity. Dodecyl POE (5) ether concentration increase further, marker release from liposomes greatly increase minor change in turbidity. For dodecyl POE (10) ether, marker release started from cmc with decrease in turbidity. The lytic action of nonionic surfactants on liposomes would thus appear related to their capacity formixed micelle formation with phospholipids.

Due to an emergency warning from a pharmaceutical company, we evaluated the procedure of providing drug information to medical staffs. The warning was made regarding DEHP, which is a plasticizer of PVC eluted in etoposide injections, and intravenous transfusion sets made from PVC are used in such injections. We tried to determine whether or not any leaching of DEHP occurred during etoposide injection. Moreover, we investigated the properties of DEHP, the mechanism of DEHP leaching from PVC, and also inspected other medicines that may also have leaching of DEHP in PVC administration sets. As a result, no clinical evidence was found to support this urgent notice. However, passing through several years, related information became gradually clear. Therefore, in the information asked for quick offer, it was suggested that gradual and continuous information service becomes indispensable especially.

本総説では, 緑茶カテキンの皮膚適用時の有益効果に焦点を当て, その秘めたるパワーについて紹介する。緑茶カテキンは, 皮膚癌の発ガン過程抑制効果, 抗炎症効果, 紫外線ダメージから皮膚を保護する効果, しみやそばかす生成予防, 美白効果, 光老化に対するアンチエイジング効果, ホルモンバランス異常に対する改善の効果を有することが報告されている。また, 緑茶カテキンの欠点である不安定性を改善するためにさまざまな技術が開発されており, その1例として, 緑茶カテキンを油中に溶解させることにより, 安定化させるとともに, 皮膚への浸透性を高めるわれわれの技術も紹介する。全世界で増加している皮膚癌患者を救う, あるいは自己治癒力, 自己免疫力を高めることによって若く健康な肌を保つ等, 緑茶カテキンの秘めたる可能性は, 近い将来の医薬品や化粧品の発展に必ずや大きな貢献をするものと期待される。
