mRNA vaccines have attracted considerable attention as a result of the 2019 coronavirus pandemic; however, challenges remain regarding use of mRNA vaccines, including insufficient delivery owing to the high molecular weights and high negative charges associated with mRNA. These characteristics of mRNA vaccines impair intracellular uptake and subsequent protein translation. In the current study, we prepared a minimal mRNA vaccine encoding a tumor associated antigen human gp10025–33 peptide (KVPRNQDWL), as a potential treatment for melanoma. Minimal mRNA vaccines have recently shown promise at improving the translational process, and can be prepared via a simple production method. Moreover, we previously reported the successful use of iontophoresis (IP) technology in the delivery of hydrophilic macromolecules into skin layers, as well as intracellular delivery of small interfering RNA (siRNA). We hypothesized that combining IP technology with a newly synthesized minimal mRNA vaccine can improve both transdermal and intracellular delivery of mRNA. Following IP-induced delivery of a mRNA vaccine, an immune response is elicited resulting in activation of skin resident immune cells. As expected, combining both technologies led to potent stimulation of the immune system, which was observed via potent tumor inhibition in mice bearing melanoma. Additionally, there was an elevation in mRNA expression levels of various cytokines, mainly interferon (IFN)-γ, as well as infiltration of cytotoxic CD8+ T cells in the tumor tissue, which are responsible for tumor clearance. This is the first report demonstrating the application of IP for delivery of a minimal mRNA vaccine as a potential melanoma therapeutic.

The efficacy of hybrid assistive limb (HAL) rehabilitation in the acute phase of stroke remains unclear. The purpose of this study was to evaluate the outcomes of patients with acute intracranial hemorrhage (ICH) who were treated with or without HAL rehabilitation. Among 270 patients with acute ICH from 2009 to 2014, 91 patients with supratentorial ICH were included in this retrospective study. Of these, 14 patients (HAL group) received HAL rehabilitation at approximately 1 week after ICH occurrence, while the remaining 77 patients received usual rehabilitation without HAL (N-HAL group). We obtained various patient data from the hospitals where the patients were moved to for further rehabilitation. Statistical comparisons were performed for the characteristics of the ICH patients, and outcomes between the HAL and N-HAL groups. There were no differences in outcomes between the HAL and N-HAL groups. However, patients with right ICH in the HAL group exhibited a significant association with a functional independence measure (FIM) score of ≥ 110 compared with patients in the N-HAL group (HAL group: 81.8%, N-HAL group: 43.9%, P = 0.04). In patients with right ICH, HAL rehabilitation was associated with improved outcomes as evaluated by the FIM score. Thus, HAL rehabilitation may improve outcomes of acute ICH in appropriately selected patients.

第30次南極地域観測隊(JARE-30)夏隊のセールロンダーネ山地地学・生物学調査は, 1988年12月29日から1989年2月1日にかけて山地西部で, 2月2日から9日にかけてあすか観測拠点をベースに付近の小山塊で実施された。2月になってからの調査活動は, ブリザード等の強風と地吹雪で効率的でなかった。JARE-26-29の地学調査に生物班が初めて加わったが, 調査計画の立案や行動形態に特に従来と変わったところはない。ここでは, 設営面を含む行動の概要と調査の概略, 調査期間の山地近辺の気象と雪氷状況を報告する。調査の成果については別途, 各分野で詳しく報告される。The summer party of the 30th Japanese Antarctic Research Expedition (JARE-30) carried out the geomorphological, paleomagnetic, geodetic, zoological and botanical field work in the western part of the Sφr Rondane Mountains for 35 days from December 29,1988 to February 1,1989,and around Asuka Station for 8 days from February 2 to 9,1989. The field work in February was largely hampered by bad weather including blizzard. In addition to earth scientists who have been in charge of the field operation of the Sφr Rondane Mountains since JARE-26 (1985), biologists joined the field party for the first time this season under the similar planning and operating scheme formerly adopted. This is the report describing the results of field operations including logistics, a summary of the field work, and some information on weather and surface conditions of snow and ice around the Mountains during this period.
