Prevalence of antibodies to Toxoplasm gondii was studied using the latex agglutination (LA) method, followed by sucrose density gradient centrifugation (SDGC) method on the small Indian mongoose (Herpestes auropunctatus), which inhabits Amami-Oshima Island. Of the 362 samples, 38 (10.5%) revealed positive. Single or double peaks in the 7–8 and/or 12–14 fraction to LA titer by SDGC indicated the early stage of T. gondii infection. It is suggested that domestic/feral cats play an important role for spreading this zoonotic pathogen to the mongoose as well as other species that are endemic to this island. Future studies are warranted to prevent the transmission of T. gondii among cats and wild animals in order to maintain the ecosystem health.

Here, we investigated the prevalence of Salmonellaenterica, with and without resistance to 17 common antimicrobial agents, in 706 green anoles (Anolis carolinensis) that were collected in Naha and Tomishiro Cities, Okinawa Main Island, Japan, between 2009 and 2014. Salmonella strains, including S. enterica Weltevreden and Enteritidis serovars, were identified in the large intestinal content samples extracted from 15 (2.1%) of the analyzed green anoles. No antimicrobial resistance was detected. Thus, the present study demonstrates that although the prevalence of Salmonella and the risk of its transmission from the green anoles to humans or other animals on Okinawa Main Island are relatively low, the green anole population nevertheless represents a potential source of Salmonella infection that could affect human health in this region.

Endangered penguins are in need of the genetic management to estimate the risk for loss of genetic diversity from populations resulting from habitat fragmentation or inbreeding. However, as for genome diversity in penguin species, there are limited reports and it is insufficient to give useful and appropriate information. In the present study, we obtained the full length sequence of the MHC class II gene, which is expected to be a useful genetic marker for biodiversity in conservation genetics. The 4.4 kb genome region containing two novel genes was determined for nucleotide sequences using genomic DNA extracted from Humboldt penguin, by using the inverse PCR method. Homology analysis of MHC class II genes with those from other birds suggested that the novel two genes were alpha and beta genes. In addition, phylogenetic analysis suggested that the beta gene of penguins was clustered with the beta genes from waterfowl. These observations provide basic information on the structure of MHC class II locus to relieve the genetic diversity of penguin species.

The transgenerational maintenance of symbiotic microbes that benefit host nutrition and health is evolutionarily advantageous. In some vertebrate lineages, coprophagy is used as a strategy for effectively transmitting microbes across generations. However, this strategy has still not been studied in birds. Accordingly, the aim of the present study was to evaluate the role of maternal cecal feces consumption by Japanese rock ptarmigan (Lagopus muta japonica) chicks as a strategy for acquiring essential gut microbes. Both the duration of coprophagy behavior by the chicks and the development process of the chick cecal microbiome (n=20 one- to three-week-old chicks, from three broods) were investigated. In all three broods, coprophagy behavior was only observed from 3 to 18 days of age. Furthermore, there was no significant difference in the number of bacterial operational taxonomic units (OTUs) in 1-week-old chicks (n=651) and adults (n=609), and most of the main OTUs observed in the adults were already present in the 1-week-old chicks. These results indicate that, in this precocial bird species, coprophagy may contribute to the early establishment of cecal bacteria that are essential for food digestion and, thus, chick survival. In fact, Japanese rock ptarmigan chicks consume the same food as their hens from the time of hatching. This behavior may have applications to ex-situ conservation.

We investigated the prevalence of Salmonella enterica and its antimicrobial resistance from 79 green anoles, the invasive alien species inhabits Haha-jima of the Ogasawara archipelago. Samples were collected during the period between 2009 and 2010. The resistance of S. enterica of these samples against 12 common antimicrobial agents was also determined. Salmonella strains, including serovar Oranienburg and Aberdeen, were detected from the large intestines of 30.4% of 79 green anole samples. And 37.5% of which were resistant to Oxytetracycline. This study suggests that green anoles may play an important role of the infection of S. enterica on this island. Attention is needed from the aspect of public and ecological health.

Ex situ conservation of Japanese rock ptarmigans began in 2015 with the aim of reintroducing artificially raised birds into their original habitat. However, the current raising method in captivity seems insufficient in terms of the survivability of artificially raised birds in natural conditions. Feeding management is one potential reason for such insufficiency. In this study, we performed a comprehensive analysis of the hydrophilic metabolites by LC-MS/MS for the cecal feces of Japanese rock ptarmigans under in situ and ex situ conservation to reveal their gut chemical environment. We also analyzed the developmental processes of cecal microbiomes both in situ semi-wild and ex situ captive individuals. Metabolites of nucleic acid were rich in the in situ individuals, and free amino acids were rich in the ex situ individuals. The differences in the microbiome composition between in situ and ex situ individuals were also pronounced; major genera of in situ individuals were not detected or few in ex situ individuals. The alpha diversity of the cecal microbiome of semi-wild chicks at 1 week of age was almost the same as that of their hens, while it was very low in captive individuals. Sub-therapeutic use of oxytetracycline, a diet rich in protein and energy, and isolation from adult birds are considered to be causes for these great differences in gut chemical and microbiological environment between in situ and ex situ individuals.

We investigated the effect of treatment with hot water extracts from the spent mushroom substrates (SMSs) of Lentinula edodes and Hypsizygus marmoreus on the resistance of rice leaves to Pyricularia oryzae infection. The spraying of the SMS extracts clearly suppressed the development of lesions caused by Py. oryzae infection. The accumulation of phytoalexins momilactones A and B, oryzalexin A, and sakuranetin was markedly induced by the spraying of extracts. The enhanced expression of defense related genes PR1b and PBZ was also found in leaves sprayed with the extracts. Treatments with the extracts also affected phytohormone levels. The levels of N6-(Δ2-isopentenyl)adenine and trans-zeatin markedly increased in response to treatment, whereas the levels of salicylic and jasmonic acids were largely unchanged.

An outbreak of encephalitozoonosis occurred in a rabbit colony at a zoo in Japan. Throughout the two years after the onset, all 42 rabbits were investigated clinically, pathologically and serologically for prevention and control of the disease. Eleven rabbits (11/42, 26.2%) showed clinical symptoms. Of 38 rabbits examined to detect specific antibodies against Encephalitozoon cuniculi, 71.1% (n=27) were found seropositive; 20 out of 30 clinically healthy rabbits (except for 8 clinical cases) were seropositive. The infection rate was 76.2% (32/42), including five pathologically diagnosed cases. The results of serological survey revealed that asymptomatic infection was widespread, even among clinically healthy rabbits. However, encephalitozoonosis was not found by pathological examination in any other species of animals kept in the same area within the zoo. Isolation and elimination of the rabbits with suspected infection based on the results of serological examination were carried out immediately; however, encephalitozoonosis continued to occur sporadically. Therefore, all the remaining rabbits were finally slaughtered. Then, the facility was closed, and all the equipment was disinfected. After a two-month interval, founder rabbits were introduced from encephalitozoonosis-free rabbitries for new colony formation. Since then, encephalitozoonosis has not been seen in any animals at the zoo. In this study, biosecurity countermeasures including staff education, epidemiological surveillance and application of an “all-out and all-in” system for rabbit colony establishment based on serological examination were successfully accomplished with regard to animal hygiene and public health for the eradication of E. cuniculi.

We investigated for the first time the prevalence of avian haemosporidia of genera Plasmodium, Haemoproteus, and Leucocytozoon among birds and mosquitoes on Tsushima Island of Japan, which is located between Japan and the Korean Peninsula. Of 55 wild birds belonging to 33 species, 16 (29.1%) tested positive for haemosporidia as follows: Plasmodium spp. (11/55; 20.0%); Haemoproteus spp. (2/55; 3.6%); and Leucocytozoon spp. (3/55; 5.5%). A genetic lineage isolated from the Eurasian Sparrowhawk (Accipiter nisus) was identical to that of the known avian malaria parasite P. circumflexum. Several genetic lineages were identical or closely related to the parasite lineages that were previously detected in birds and mosquitoes in Japan and Korea. Another single identical genetic lineage was also detected in both migratory and resident birds. A total of 753 mosquitoes from 12 species were collected; and one fully fed Aedes albopictus was positive for avian Plasmodium (1/753; 0.13%) which is identical to a genetic lineage detected in both mosquitoes in Japan and birds in Korea. Blood-meal identifications of blood-fed mosquitoes showed direct contact between the mosquitoes and 4 species of mammals including humans, cattle, rodents, and the endangered Tsushima leopard cat (Prionailurus bengalensis euptilura). Migratory birds use Tsushima Island as a site for wintering, breeding, and resting, and our results suggest the transmission of avian haematozoa between resident and migratory birds during their stay on Tsushima Island.
