Parthenocarpy is a trait where fruit set and growth are triggered without pollination and fertilization. In the tomato (Solanum lycopersicum L.), this trait is considered attractive as it reduces the cost and labor requirements for fruit setting. In this study, we investigated the inheritance of parthenocarpy in ‘MPK-1’—a parthenocarpic tomato cultivar derived from a cross between a variant from a self-fertilization posterity of ‘Severianin’, which exhibited strong parthenocarpy and a non-parthenocarpic cultivar. It was reported that ‘MPK-1’ contains a pat-2 gene because ‘Severianin’ which has a pat-2 gene is its only parthenocarpic ancestor. However, we found that parthenocarpy in ‘MPK-1’ is controlled by a novel parthenocarpic gene, not pat-2. This novel gene, which was designated as Pat-k, is semi-dominant and located on chromosome 1. We also showed that the size of the parthenocarpic fruit of ‘MPK-1’ is similar to that of the pollinated fruit at maturity. Thus, ‘MPK-1’ may be used as a new parthenocarpic resource for breeding.

Pepper yellow leaf curl disease (PepYLCD) caused by begomoviruses is one of the most devastating diseases affecting pepper (Capsicum spp.) production worldwide. In our previous study, a loss-of-function allele, pepy-1, encoding messenger RNA surveillance factor Pelota was identified as a begomovirus resistance gene from a C. annuum cultivar BaPep-5. In this study, to investigate the effectiveness of pepy-1 conferred resistance against begomovirus in the field, we conducted a three-year evaluation under natural field conditions in Indonesia. The lowest PepYLCD incidence and significantly higher fruit productivity were observed in BaPep-5 when compared to six other commonly cultivated pepper cultivars. The subsequent comparison between BaPep-5 and the susceptible BaPep-4 showed that pepy-1 slowed down the disease onset and progression, resulting in a higher fruit productivity trait in the field. Multiple comparison analyses using an F2 population obtained by crossing BaPep-5 with BaPep-4 showed that the pepy-1 homozygous individuals had significantly higher fruit productivity, twice than those of the Pepy-1 homozygous or heterozygous individuals. In conclusion, the introgression of pepy-1 is effective to reduce the economic loss of pepper fruit production under natural field infection of begomoviruses.

Indonesia is one of the world’s largest fresh pepper (Capsicum spp.) fruit-producing countries, and hot peppers are essential spices in Indonesian cuisine. During the last two decades, begomovirus, which is transmitted by the whitefly, Bemisia tabaci (Gennadius), and causes pepper yellow leaf curl disease, began to cause a huge economic loss by damaging pepper plants in Indonesia. In the present study, a highly efficient inoculation method was established for Pepper yellow leaf curl Indonesia virus (PepYLCIV), the most infectious bipartite begomovirus in pepper plants cultivated in North Sumatra, by combining agroinoculation and subsequent grafting. Partial tandem repeats of PepYLCIV DNA A and B were constructed and cloned into a binary pGreenII vector, and their infectivity was tested. Co-inoculation of Nicotiana benthamiana L. and Solanum lycopersicum L. ‘Momotaro’ with PepYLCIV DNA A and DNA B resulted in the production of typical begomoviral symptoms. Both the injection of the cotyledons with cultured agrobacteria and the inoculation of the hypocotyl with agrobacterial colonies induced viral symptoms in pepper No. 218 (C. annuum L.) seedlings in approximately 55–75%. When agroinoculated symptomatic No. 218 was grafted onto an uninfected ‘Takanotsume’ (C. annuum), all newly elongated shoots from the rootstock of ‘Takanotsume’ produced typical begomoviral symptoms. Agroinoculation combined with subsequent grafting provides a highly efficient method for introducing PepYLCIV into pepper plants.

Capsinoids are low-pungent capsaicinoid analogues in chili pepper fruits. They exhibit various bioactivities in humans similar to capsaicinoids, but do not produce a nasty burning sensation, encouraging their application in foods and supplements. Previous reports demonstrated that loss-of-function of putative aminotransferase (pAMT) leads to low-pungency and capsinoid accumulation. Therefore, the pamt allele is a useful gene in chili pepper breeding programs to enhance health-promoting properties. Eight loss-of-function alleles have been identified in the Capsicum genus, but the variation in pamt alleles remains to be fully elucidated. In this study, we identified one novel loss-of-function allele from the analysis of low-pungent chili pepper ‘No. 4034’ (C. chinense). ‘No. 4034’ contained mainly capsinoid with an undetectable level of capsaicinoid. A genetic complementation test was conducted by crossing ‘No. 4034’ with other accessions. The results indicated that ‘No. 4034’ possessed a loss-of-function pamt allele. Sequence analysis showed that the novel mutant allele contained a 7-bp insertion (TCGGTAC) in the 16th exon region, which we designated as pamt9. The insertion caused a frameshift mutation and resulted in a truncated protein. Gene expression analysis showed that the expression level of pAMT specifically decreased among biosynthetic genes tested here in ‘No. 4034’, compared with that of pungent accession. pamt9 will be useful for low-pungency and capsinoid breeding, and will provide additional information for variations in pAMT mutants.

Tomato yellow leaf curl disease caused by begomoviruses is a serious threat to tomato (Solanum lycopersicum L.) production. If begomoviruses, transmitted by whitefly (Bemisia tabaci), infect tomato plants during early growth, production can be almost entirely lost. Tomato yellow leaf curl Kanchanaburi virus (TYLCKaV), a bipartite Begomovirus, is emerging as an important threat to solanaceous crop production in Southeast Asia. The lack of mechanical transmission of some begomoviruses is a major experimental constraint. In this study, an agroinoculation method using TYLCKaV in tomato plants was established. Partial tandem repeats of TYLCKaV DNA A and DNA B were constructed and cloned to a binary pGreenII vector, and their infectivity was tested. Co-inoculation of TYLCKaV DNA A and DNA B to Nicotiana benthamiana L. produced typical begomoviral symptoms, and both of the viral DNA components accumulated in the upper uninoculated leaves, suggesting systemic infection of TYLCKaV. Two agroinoculation methods were conducted on tomatoes. First, excised sections of tomato shoots were agroinoculated with a soaking procedure. Although two Agrobacterium tumefaciens strains were tested, approximately 40% of inoculated plants only showed viral symptoms for EHA105. Second, agrobacterium from a cultured petri dish was directly inoculated with a colony inoculation procedure. When EHA105 was used, approximately 92% of inoculated plants showed viral symptoms. Sequencing the recovered viral DNA from the upper uninoculated leaf clarified that TYLCKaV had successfully infected the tomato plants. The colony inoculation procedure is labor-saving, and viral symptoms develop in susceptible tomato plants within approximately a month from sowing the seeds. This method could contribute to simple and speedy evaluation of TYLCKaV resistance of tomato plants.

‘No.80’ (Capsicum chinense) from the Caribbean is a valuable genetic source from the aspect of its non-pungent and highly aromatic traits. In the present study, the non-pungency, volatile components, and phylogenetic origin of ‘No.80’ were analyzed with another C. chinense cultivar, ‘No.2’ from Brazil, which is also non-pungent but less aromatic. Expressions and deduced amino acid sequences of acyltransferase (Pun1) of ‘No.80’ and ‘No.2’ were normal compared with a pungent cultivar, ‘Habanero’. Insertions of 7-bp and 8-bp resulting in frameshift mutations were found in the coding regions of putative aminotransferase (p-AMT) of ‘No.80’ and ‘No.2’, respectively. Co-segregation of these insertions with the non-pungent phenotypes in F1 and F2 populations obtained from crossing ‘No.80’ or ‘No.2’ with ‘Habanero’ suggested that non-pungency in these cultivars arose from genetic mutations of p-AMT that occurred independently. Moreover, molecular phylogenetic analysis suggested that ‘No.80’, a close relative of ‘No.2’, originates from capsicums migrated from the South American mainland. In addition to pungency, we assessed the volatile components of the highly aromatic ‘No.80’, the less aromatic ‘No.2’, and their F1 hybrid using gas chromatography. ‘No.80’ contained higher levels of aroma-contributing volatiles than ‘No.2’, which correlated with the stronger and weaker aromas of two cultivars. Further, the fruit of F1 progenies emitted a number of volatile compounds between or higher than their corresponding parents. Based on these results, the approaches for breeding highly aromatic non-pungent cultivars are discussed.
