Gladiolus (Gladiolus spp.) florets exhibit low ethylene sensitivity. Accordingly, the wilting of their tepals is an ethylene-independent process. Both trehalose and cycloheximide can extend the vase life of gladiolus florets. Floral senescence is probably regulated by programmed cell death. However, senescence-related genes have not been thoroughly investigated, except in ethylene-sensitive species. In this study, we analyzed the expression of senescence-associated genes by conducting transcriptome (RNA-seq) analysis. First, we examined the effects of 0.1 M trehalose (Tre), 300 μM cycloheximide (CHI), and 50 μM chloramphenicol (CAP) treatments on postharvest quality and senescence-related gene expression in gladiolus ‘Fujinoyuki’ cut florets. The Tre and CHI treatments extended the vase life of gladiolus florets by about 1 day, i.e., 30% of Cont. Tepals were sampled at 0 days (0d) and 2 days (2d) post-treatment. The RNA-seq analysis of floret tissues generated 81,136 unique sequences. Moreover, 2,892, 4,670, and 57 differentially expressed genes were identified from the 0d_Control (Cont) vs 2d_Cont, 2d_Cont vs 2d_CHI, and 2d_Cont vs 2d_Tre comparisons, respectively. Gene Ontology (GO) analysis suggested that cysteine-type endopeptidase activity was significantly higher for 2d_Cont than for 0d. Additionally, the 0d vs 2d_Cont comparison showed the cell wall-related GO terms were more enriched for 2d_Cont. The Kyoto Encyclopedia of Genes and Genomes analysis revealed an increase in the expression of sucrose synthesis-related genes in the 2d_Cont samples. Among the genes involved in starch and sucrose metabolism, the genes mediating cell wall degradation were more actively expressed in the 2d_Cont samples than in the 2d_CHI samples. The fragments per kilobase per million reads (FPKM) values were used to select candidate senescence-related gene families, including the cysteine protease, invertase, peroxidase, pectinesterase, and transcription factor (NAC [no apical meristem (NAM), Arabidopsis transcription activation factor (ATAF), and cup-shaped cotyledon (CUC)] and WRKY) families. The expression levels of transcription factor genes, including NAC 048, 68, 073 and WRKY 6, 11, 24, were validated by qPCR. The expression of these NAC and WRKY transcription factor genes was upregulated by CHI, suggesting their involvement in senescence or side reactions in gladiolus tepals. This study revealed several candidate genes and associated GO terms for senescence of cut florets, but further study is needed, especially on key genes, including transcription factors.

α-Ketoglutarate (α-KG) also known as 2-oxoglutarate (2-OG) is an intermediate metabolite in the tricarboxylic acid (TCA) cycle and is also produced by the deamination of glutamate. It is an indispensable cofactor for a series of 2-oxoglutarate-dependent oxygenases including epigenetic modifiers such as ten-eleven translocation DNA demethylases (TETs) and JmjC domain-containing histone demethylases (JMJDs). Since these epigenetic enzymes target genomic DNA and histone in the nucleus, the nuclear concentration of α-KG would affect the levels of transcription by modulating the activity of the epigenetic enzymes. Thus, it is of great interest to measure the nuclear concentration of α-KG to elucidate the regulatory mechanism of these enzymes. Here, we report a novel fluorescence resonance energy transfer (FRET)-based biosensor with multiple nuclear localization signals (NLSs) to measure the nuclear concentration of α-KG. The probe contains the α-KG-binding GAF domain of NifA protein from Azotobacter vinelandii fused with EYFP and ECFP. Treatment of 3T3-L1 preadipocytes expressing this probe with either dimethyl-2-oxoglutarate (dimethyl-2-OG), a cell-permeable 2-OG derivative, or citrate elicited time- and dose-dependent changes in the FRET ratio, proving that this probe functions as an α-KG sensor. Measurement of the nuclear α-KG levels in the 3T3-L1 cells stably expressing the probe during adipocyte differentiation revealed that the nuclear concentration of α-KG increased in the early stage of differentiation and remained high thereafter. Thus, this nuclear-localized α-KG probe is a powerful tool for real-time monitoring of α-KG concentrations with subcellular resolution in living cells and is useful for elucidating the regulatory mechanisms of epigenetic enzymes.

Tinea imbricata and tinea pseudoimbricata are variant types of tinea corporis characterized by annual-ring-shaped erythema. Although the skin lesions manifest similar symptoms, these two diseases are classified based on causative fungi. The former is caused by Trichophyton concentricum, an anthropophilic dermatophyte, and the latter is caused by dermatophytes other than T. concentricum, commonly zoophilic fungi such as Trichophyton mentagrophytes complex. Here, we report a 27-year-old Japanese male diagnosed with tinea pseudoimbricata attributed to Trichophyton tonsurans, an anthropophilic dermatophyte. We suspected that application of steroid ointment caused the annular pattern of his skin lesions. After three months use of topical luliconazole cream, treatment was finished. We also summarize the knowledge about tinea pseudoimbricata through previous reports with bibliographical consideration.

The present study aimed to investigate whether individuals with high social anxiety prefer robots over humans as their exercise/sports partner. This study included a questionnaire survey with 520 Japanese participants (mean age=44.39, SD=14.33). The results showed that for all exercise-/sports-related situations, participants who selected robots as exercise/sports partners scored significantly higher on the SIAS-6 assessing social anxiety than the participants who selected humans. It can be concluded that introducing robots as exercise/sport partners may help people with high social anxiety start and continue their daily physical activities.

Sick sinus syndrome (SSS) is a type of bradyarrhythmia that can lead to syncope. Cilostazol has been reported to be an effective treatment for human patients with SSS and other bradyarrhythmias. This report describes the successful long-term treatment with cilostazol in a dog with SSS. A nine-year old intact male Miniature Schnauzer presented with a history of syncopal episodes and unsteady gait. After cilostazol treatment, the total heart rate (HR), mean HR, and frequency of premature ventricular contractions (PVCs) increased, while the maximum HR and maximum pause time decreased. Additionally, the number of syncopal episodes decreased. The dog died suddenly, 1,418 days after the start of cilostazol treatment. Cilostazol may be a useful therapeutic agent in canines with SSS.
