モエジマシダ(Pteris vittata)等のヒ素高蓄積植物はヒ素土壌汚染の浄化に適用可能と考えられている.本研究では, モエジマシダを用いたヒ素のファイトエクストラクション法による浄化技術に着目し, それによるヒ素吸収における微生物の関与の度合いを明らかにすることを目的として行った. 微生物植種の添加に伴うシダの栽培実験と土壌ヒ素の酸化試験を行い, ヒ素の土壌からシダへの移行特徴および形態変化を解析した. その結果, 土壌中の亜ヒ酸はヒ酸に酸化されたのち, その多くはシダにより吸収されることと, このヒ素酸化は微生物による作用であることが強く示された. さらに, シダ根圏試料に対して亜ヒ酸酸化酵素遺伝子aroAをターゲットとしたT-RFLP解析より, シダによるヒ素吸収に先立つヒ素形態の変化には, 土壌中のaroAを有する微生物が関与していると示唆された.

In resent Japanese elemental and lower secondary education, opportunity of experimental work for students has been limited, and it has made difficult to obtain fundamental knowledge in natural sciences. This recent situation causes needs to provide special and general science courses through laboratory work in the liberal arts education in science and engineering faculties of universities and colleges. In this paper, the author tries to edit new attempts of development of “Laboratory Study Course for Natural Science Fundamentals” . The concept of natural science fundamentals, and the reason for the need, the contents and the aims of the laboratory study course are discussed in this article.

Methane production in the environment has been recognized as an important factor for the global warming. It is also important as a process for self-purification in the polluted environment. In this study, the author isolated methanogenic bacteria which live in a lake sediment and investigated the methanogenic activity of the isolated bacteria. The author also studied the distribution of methanogenic activity in a lake sediment by using sediment samples which were obtained from each depth of the sediment, and temperature effects to the methanogenic activities. One coccus bacterial strain of methanogenic bacteria isolated from the lake sediment was a hydrogen utilizing methanogen. It was impossible to isolate acetate utilizing methanogen from the lake sediment because of the coexisting bacteria other than methanogens. There were many differences between hydrogen utilizing methanogenesis and acetate utilizing methanogenesis. Methanogenesis from hydrogen dominated in the lake sediment, especially in the deeper sediment. This methanogenic activity was also observed at low temperature. Methanogenesis from acetate was active at the upper sediment which contained much organic compounds, but not so high as methanogenesis from hydrogen.

Outbreaks of infectious diseases by new pathogenic microorganisms through water utilization have been reported in recent years. Especially, Legionnaires' disease has been recognized as a serious problem in modern societies since a high concentration of <i>Legionella pneumophila</i> often exist in water environment (such as cooling tower, bath water, recreational water) close to human activities. In this study, the quantitative detection method (colony-hybridization) for <i>L. pneumophila</i> was developed by combining the hybridization with the plate culture method. The developed detection method enabled <i>L. pneumophila</i> to be enumerated in various water samples. It was possible to quantify <i>L. pneumophila</i> without interferences of other bacteria by this method in case that the number of total coliforms on the plate was less than 10<sup>3</sup> CFU.
