Amphidromous gobies of Sicyopterus (Gobiidae: Sicydiinae) are distributed in tropical, subtropical, and temperate streams in the Indo-Pacific region. Two species, Sicyopterus japonicus (Tanaka, 1909) and Sicyopterus lagocephalus (Pallas, 1770), are known from Japan. In the present study, two specimens of an additional species were collected in Okinawa Island, in southern Japan. We compared morphologies of the type series of Sicyopterus longifilis de Beaufort, 1912 and Sicyopterus brevis de Beaufort, 1912 collected in Indonesia, with the specimens from Okinawa Island to revise the taxonomy of these species and to identify the Okinawan specimens. Syntypes of S. longifilis and S. brevis share many characters, including a unique mouth morphology. Although fin morphologies, tooth number, and the shape of the urogenital papilla differ between S. longifilis and S. brevis syntypes, these amount to normal sexual dimorphism of sicydiine gobies. De Beaufort collected his specimens at the same locality on the same date. We conclude that the syntypes of S. longifilis and S. brevis are actually males and females of same species. Therefore, they are subjective synonyms, and we give precedence to the name S. longifilis as the first reviser under Article 24.2 of the International Code of Zoological Nomenclature (International Commission on Zoological Nomenclature 1999). The two specimens from Okinawa Island were identified as S. longifilis and this is the first record of this species from Japan.

Twenty specimens of Acentrogobius ocyurus (Jordan and Seale, 1907) were collected in Manko, Okinawa-jima Island, Japan and Sulu Sea side of Puerto Princesa City, Palawan Island, Philippines. As the species had previously been reported from the South China Sea, Andaman Sea, and northern Australian waters, these specimens from Okinawa and Palawan represent the first records of the species from Japan and the Sulu Sea. Acentrogobius ocyurus was found on muddy substrata at the lowest tidal zone of the mudflat and was often found in burrows of mangrove pistol shrimp, Alpheus richardsoni Yaldwyn, 1971. Morphological traits suggested that A. ocyurus is closely related to A. globiceps (Hora, 1923), A. kranjiensis (Herre, 1940), and Acentrogobius sp. (“Suzume-haze” in Japanese name), with several homologous features including, the transverse pattern of the sensory papillae on the cheek, caudal fin pointed in middle, a black stripe behind the eye along the anterior oculoscapular canal, bright blue spots laterally on the body, and an oblique yellow stripe across the upper part of the caudal fin.

We evaluated the antiviral activity of a chlorine dioxide gas solution (CD) and sodium hypochlorite (SH) against feline calicivirus, human influenza virus, measles virus, canine distemper virus, human herpesvirus, human adenovirus, canine adenovirus and canine parvovirus. CD at concentrations ranging from 1 to 100 ppm produced potent antiviral activity, inactivating ≥ 99.9% of the viruses with a 15 sec treatment for sensitization. The antiviral activity of CD was approximately10 times higher than that of SH.

Seven specimens of Paloa villadolidi Roxas and Ablan, 1940, were collected from Okinawa and Ishigaki islands in the Ryukyu Archipelago, Japan, and their morphologies are described herein. Although this species has been considered as a synonym of Odonteleotris macrodon (Bleeker, 1853) which is redescribed in this study, the former is distinguished from the latter by having a steeper jaw (upper jaw tip not reaching orbit vs. ending below orbit center in O. macrodon), a contrasting number of canine teeth on the jaws (more upper-jaw canines than lower-jaw ones vs. more lower-jaw canines), the absence of distinct fin markings (vs. many small black spots on fin rays), no spots on the caudal-fin base (vs. with a red spot), and a higher number of cephalic sensory canal pores (17 vs. 13 or 14). This study also noted a unique canine teeth arrangement in P. villadolidi, which has canine teeth only in the anterior half of the lower jaw, unlike the canine teeth arrangement of four other butid genera (Incara Rao, 1971, Odonteleotris Gill, 1863, Ophiocara Gill, 1863, and Oxyeleotris Bleeker, 1874), which have an inner row of canine teeth in the posterior half of the lower jaw. Although further study is needed to determine the taxonomic status of P. polylepis Herre, 1927, the other nominal species of the genus, the present study tentatively considered it valid based on the original description, which described it as having a deeper body than P. villadolidi. The seven Japanese specimens were identified as P. villadolidi, as they have slenderer bodies than P. polylepis, and they represent the first Japanese records of the species.

A single specimen of Muraenichthys gymnopterus (Bleeker, 1853) was collected from a sandy intertidal flat having rocks and dead corals in the estuarine area of Nagura Amparu, Ishigaki-jima Island, southern Japan, in October 2020. This specimen collection constitutes the first record of M. gymnopterus from Japanese waters. In this study, the diagnostic characters between M. gymnopterus and M. hattae Jordan and Snyder, 1901 are provided, based on our morphological observations of 37 specimens, including Okinawan specimen, and previous studies, as follows: head length [M. gymnopterus 11.8–15.0% of total length (TL) vs. M. hattae 9.4–11.0%], trunk length (24–25.4% of TL vs. 28–31%), the horizontal distance from the dorsal-fin origin to a vertical line through the anus 73–87% of head length vs. 13–49%), the number of vertebrae (total 129–130 vs. 148–155; predorsal 30 vs. 47–53; preanal 41–44 vs. 51–55), the number of the lateral-line pores before the anus (43–45 vs. 51–55). Additionally, the body depth at the gill opening in TL and the trunk length in TL can also be used to distinguish between these two species (2.8–3.7% of TL vs. 1.4–3.0%). Although M. gymnopterus has previously been reported from tropical to temperate regions, we suspect that the records from temperate regions are based on misidentification of M. hattae.

A two-year-old male domestic cat showed lethargy, tonic-clonic convulsion, and mucosal jaundice. Upon admission, blood examination indicated severe neutropenia and thrombocytopenia, and ultrasonography revealed diffuse splenomegaly with a honeycomb appearance and abdominal lymph nodes enlargement in addition to a decrease in cardiac blood flow indicating a shock condition. Cytology of the spleen showed a cell population composed of immature large lymphoid cells with distinct nucleoli, suggesting lymphoma. The cat received symptomatic treatments but died four hours later. Reverse transcriptase polymerase chain reaction assay of the spleen sample indicated the presence of severe fever with thrombocytopenia syndrome (SFTS) virus S gene segment. Clinical features of this case that was diagnose as SFTS were similar to lymphoma. Therefore, pet owners and veterinary workers should be protected against infection of SFTS.

One female specimen of Stiphodon multisquamus Wu and Ni, 1986 was collected from Arida-gawa River, Wakayama Prefecture, Japan, in November 2018. As the previously known range of this species was from the South China Sea, including Malaysia, Vietnam, and China, to the Ryukyu Archipelago in Japan, this specimen was reported as the first record from the Japanese mainland and the northernmost record of this species. Stiphodon multisquamus is an amphidromous fish of which pelagic larvae develop in marine habitats. It was believed that this specimen was transported from the southern region by ocean currents during the pelagic larval phase and recruited into the river in Wakayama Prefecture.

Hepatitis E virus (HEV) is known to cause zoonotic infections from pigs, wild boars and deer. Domestic pigs have been used as an experimental animal model in medical research and training; however, the risks of HEV infection from pigs during animal experiments are largely unknown. Here, we retrospectively investigated the seroprevalence and detection rates of viral RNA in 73 domestic pigs (average 34.5 kg) introduced into an animal experimental facility in a medical school during 2012–2016. We detected anti-HEV immunoglobulin G antibodies in 24 of 73 plasma samples (32.9%), though none of the samples were positive for viral RNA. Plasma samples of 18 pigs were sequentially monitored and were classified into four patterns: sustained positive (5 pigs), sustained negative (5 pigs), conversion to positive (6 pigs) and conversion to negative (2 pigs). HEV genomes were detected in 2 of 4 liver samples from pigs that were transported from the same farm during 2016–2017. Two viral sequences of the overlapping open reading frame (ORF) 2/3 region (97 bp) were identical and phylogenetically fell into genotype 3. A 459-bp length of the ORF2 region of an amplified fragment from a pig transported in 2017 was clustered with the wbJYG1 isolate (subgenotype 3b) with 91.5% (420/459 bp) nucleotide identity. Based on our results, we suggest that domestic pigs introduced into animal facilities carry a potential risk of HEV infection to researchers, trainees and facility staff. Continuous surveillance and precautions are important to prevent HEV infection in animal facilities.

The present study investigated severe fever with thrombocytopenia syndrome virus (SFTSV) infection in raccoons in Wakayama Prefecture from 2007 to 2019. To perform surveillance, an enzyme-linked immunosorbent assay (ELISA) was established, and the sensitivity and specificity of the ELISA were 100% in comparison with a 50% focus-reduction neutralization assay. Using the established ELISA, we performed serosurveillance of SFTSV infection in 2,299 raccoons in Tanabe region, Wakayama Prefecture from 2007 to 2019. The first anti-SFTSV-positive raccoon was captured in October 2009. The seroprevalence of SFTSV infection was <10% between April 2009 and March 2013, 23.9% between April 2013 and March 2014, 37.5% between April, 2014 and March 2015, and over 50% from April 2015. Next, we performed detection of SFTSV genes in sera of raccoons captured in Wakayama Prefecture after April 2013. The results indicated that 2.4% of raccoons were positive for SFTSV genes and that the frequency of SFTSV infection among raccoons between January and March (0.7%) was lower than that between April and June (3.4%). In addition, virus genes were detected from many specimens, including sera and feces of two raccoons, and viral antigens were detected in lymphoid cells in lymphoid follicles in the colon by immunohistochemical staining. In conclusion, SFTSV had recently invaded the area and had rapidly spread among wild animals. The first patient in this area was reported in June 2014, indicating that raccoons are good sentinels for assessing the risk of SFTSV in humans.

Bats are the second diversity species of mammals and widely distributed in the world. They are thought to be reservoir and vectors of zoonotic pathogens. However, there is scarce report of the evidence of pathogenic bacteria kept in bats. The precise knowledge of the pathogenic bacteria in bat microbiota is important for zoonosis control. Thus, metagenomic analysis targeting the V3-V4 region of the 16S rRNA of the rectal microbiota in Rousettus amplexicaudatus was performed using high throughput sequencing. The results revealed that 103 genera of bacteria including Camplyobacter were detected. Campylobacter was second predominant genus, and Campylobacter coli and Campylobacter jejuni were identified in microbiome of R. amplexicaudatus. Campylobacteriosis is one of the serious bacterial diarrhea in human, and the most often implicated species as the causative agent of campylobacteriosis is C. jejuni. Therefore, we investigated the prevalence of C. jejuni in 91 wild bats with PCR. As a result of PCR assay targeted on 16S-23S intergenic spacer, partial genome of C. jejuni was detected only in five R. amplexicaudatus. This is the first report that C. jejuni was detected in bat rectal swab samples. C. jejuni is the most common cause of campylobacteriosis in humans, transmitted through water and contact with livestock animals. This result indicated that R. amplexicaudatus may be a carrier of C. jejuni.

In 2007–2008, a canine distemper virus (CDV) epidemic occurred among wild animals in Wakayama Prefecture, Japan, and many mammals, including the wild boar and deer, were infected. In this study, CDV prevalence among wild animals was surveyed before and after the epidemic. At first, an enzyme-linked immunosorbent assay (ELISA) with horseradish peroxidase-conjugated protein A/G was established to detect CDV antibodies in many mammalian species. This established ELISA was available for testing dogs, raccoons and raccoon dogs as well as virus-neutralization test. Next, a serological survey of wild mammalians was conducted, and it was indicated that many wild mammalians, particularly raccoons, were infected with CDV during the epidemic, but few were infected before and after the epidemic. On the other hand, many raccoon dogs died during the epidemic, but CDV remained prevalent in the remaining population, and a small epidemic occurred in raccoon dogs in 2012–2013. These results indicated that the epidemic of 2007–2008 may have been intensified by transmission to raccoons.

Post-import from the Republic of Indonesia to Japan in 2017, two juvenile, captive bred Asian small-clawed otters (Aonyx cinereus) exhibited gastrointestinal symptoms, including vomiting, diarrhea, and hematemesis, and died. One of them was examined postmortem. Microscopically, the small intestinal mucosa was necrotic with crypts lined by regenerating large epithelial cells. A gastric cardiac mucosal ulcerative lesion containing fungal yeasts and pseudohyphae morphologically indicated Candida spp. The lymph nodes exhibited marked lymphoid depletion. Canine parvovirus 2 (CPV-2) was isolated from an oral swab, and virus protein 2 (VP2) gene sequencing revealed new CPV-2a. To our knowledge, this is the first new CPV-2a infection report in Asian small-clawed otters. This infection should be considered in gastrointestinal symptom-related cases in this species.

Canine distemper virus (CDV) is one of the most serious pathogens found in many species of carnivores, including domestic dogs. In this study, hemagglutinin (H) genes were detected in five domestic Vietnamese dogs with diarrhea, and two CDVs were successfully isolated from dogs positive for H genes. The complete genome of one isolate, CDV/dog/HCM/33/140816, was determined. Phylogenetic analysis showed that all Vietnamese CDVs belonged to the Asia-1 genotype. In addition, the H proteins of Vietnamese CDV strains were the most homologous to those of Chinese CDVs (98.4% to 99.3% identity). These results indicated that the Asia-1 genotype of CDV was the predominant genotype circulating among the domestic dog population in Vietnam and that transboundary transmission of CDV has occurred between Vietnam and China.

Severe fever with thrombocytopenia syndrome virus (SFTSV) causes lethal hemorrhagic diseases in human, cats, and dogs. Several human cases involving direct transmission of SFTSV from diseased animals have been reported. Therefore, rapid diagnosis in veterinary clinics is important for preventing animal-to-human transmission. Previously, we developed a simplified reverse transcription-loop-mediated isothermal amplification (RT-LAMP) assay for human that does not require RNA extraction for detecting the SFTSV genome. In this study, we improved the simplified RT-LAMP assay for cats by introducing a dried reaction reagent and investigated the applicability of this method for diagnosing SFTS in cats. SFTSV RNA was detected in 11 of 12 cats naturally infected with SFTSV by RT-LAMP assay using both liquid and dried reagents. The RT-LAMP assay using liquid and dried reagents was also applicable to the detection of SFTSV genes 3–4 days after challenge in cats experimentally infected with SFTSV. The minimum copy number of SFTSV genes for 100% detection using the RT-LAMP assay with liquid and dried reagents was 4.3×104 and 9.6×104 copies/mL, respectively. Although the RT-LAMP assay using the dried reagent was less sensitive than that using the liquid reagent, it was sufficiently sensitive to detect SFTSV genes in cats with acute-phase SFTS. As the simplified RT-LAMP assay using a dried reagent enables detection of SFTSV genes more readily than the assay using a liquid reagent, it is applicable for use in veterinary clinics.

A previous serosurvey of Japanese encephalitis virus (JEV) among dogs suggested that dogs are well suited for use as sentinels for assessing the risk of JEV transmission to humans. To examine the clinical symptoms and duration of anti-JEV antibodies in dogs, three dogs were experimentally challenged with JEV. All JEV-infected dogs did not show any clinical signs or abnormal blood tests, except for C-reactive protein. Virus-neutralization titers rapidly increased and were maintained until 70 days postinfection, and neither the virus nor the viral genome was detected in blood. Thus, since dogs live in close proximity to humans as companion animals, they are well suited for use as sentinels for surveying the human risk of JEV infection.

Canine distemper virus (CDV) causes a lethal disease among members of the Carnivora. To clarify the distribution of CDV in wild animals, we examined 106 raccoon sera collected from two prefectures in Japan, Hyogo and Osaka, from 2005 to 2007. Among them, 34 raccoons (32.1%) possessed a virus-neutralizing (VN) antibody to KDK-1 strain (genotype Asia-1). There was no significant difference in seroprevalence of CDV regardless of places, gender, and body weights. In Hyogo, a geometric mean of VN titers to KDK-1 was significantly higher than that to Onderstepoort (vaccine strain), indicating that KDK-1-like CDV different from vaccine strain might have spread among raccoon population in Hyogo. In conclusion, CDV is epidemic among feral raccoons in Japan, suggesting that CDV might have been spreading among Japanese wild animals.

Canine coronavirus (CCoV) is an important pathogen that causes enteritis in dogs, but there is no information on CCoV infection in Vietnam. To examine the prevalence of CCoV infection among Vietnamese dogs, 201 serum samples were analyzed by virus-neutralization (VN) test. The results showed that antibody against CCoV-II was present in 87 dogs (43.3%). To detect genes of CCoV, fecal samples collected from 30 diarrheic and 50 healthy dogs were examinated by RT-PCR, confirming that 2 diarrheic dogs and 5 healthy dogs were positive for CCoV. Nucleotide sequences of N-terminal region of spike (S) gene indicated that CCoV strains were divided into two subgenotypes, CCoV-IIa and -IIb, respectively. Furthemore, we succeeded in isolating CCoV/dog/HCM47/2015, the isolate was plaque-purified three times, and 3′-terminal one-third of the genome was analyzed. Interestingly, the plaque-purified virus had a large deletion in ORF3abc and E genes (1,165 nt), and a short deletion in ORF7b gene (60 nt), suggesting that these regions are not necessary for in vitro replication of CCoV. Next, the antigenicity between the isolated CCoV-IIb and the other CCoV-IIa was compared by VN test, revealing that antigenicty of the isolated CCoV is equal or higher than that of the other CCoV. In summary, two subgenotypes of CCoV-II are spreading among Vietnamese dogs. The isolated virus with a large deletion after in vitro passage may be useful for the development of vaccine, owing to its antigenicity and efficient viral growth in vitro.

Reovirus infection is common in mammals. However, seroepidemiological data of reovirus neutralizing antibodies are limited in dogs. In this study, sera of 65 healthy dogs from six prefectures across Japan were tested for neutralizing antibodies against reovirus serotype 1 strain Lang (T1L), serotype 2 strain Amy (T2A) and serotype 3 strain Dearing (T3D) using plaque reduction neutralization test (PRNT). Seropositivity against reovirus T1L, T2A and T3D was 53.85%, 33.85% and 46.15%, respectively. Distribution of reovirus seropositive samples displayed no distinguishable geographical pattern. However, reovirus seropositivity increases with age and in dogs housed outdoor. Co-infection of multiple reovirus serotypes in dogs is also possible. These data will provide valuable insights towards the usage of reovirus in oncolytic virotherapy in canine cancers.
