Chili pepper is cultivated in the absence of fruit-promoting treatments or insect pollinators. The fertilization ability of both males and females, as well as natural self-pollination ability under high temperatures, are necessary for autonomous set fruit under high temperatures. The reproductive traits related to autonomous self-pollination ability and fertilization ability of both males and females after pollination under control (CK) and high temperature (HT) treatments were investigated in this study. ‘Takanotsume’ (TK) showed a significantly higher percentage of autonomous fruit set than ‘Peruvian Purple’ (PP) in both CK and HT treatments, suggesting that TK has a strong autonomous fruit set regardless of temperature. On the other hand, the percentage of autonomous fruit set of PP was 0% in the HT treatment, while autonomous fruit set was observed in the CK treatment. Therefore, TK had autonomous fruit set ability even at high temperatures when PP did not have any. TK had more pollen on the stigma than PP, suggesting that TK has a higher ability for autonomous self-pollination. Flower morphology was not considered a factor in the autonomous self-pollination of TK. In contrast, TK tended to release more pollen than PP. One factor that can support autonomous self-pollination is pollen dispersion. A significant difference was observed in the fruit set rate during artificial pollination between the treatments (CK and HT) of the pollen parent and the cultivars (TK and PP) of the seed parent. In fact, female fertility in TK was higher than that in PP, regardless of the temperature condition. Almost no fruit set of PP was observed via autonomous self-pollination in the HT treatment, but fruits were set by artificial pollination using both the pollen and pistil of PP in the HT treatment. This result is consistent with the finding that PP exhibits decreased autonomous self-pollination at high temperatures. The higher autonomous fruit-set ability of TK than PP under high temperatures may be attributed not only to superior female fertility, but also to its autonomous self-pollination ability under high temperatures. Therefore, pollen dispersal ability under high temperatures was considered a key factor for autonomous fruit-set.

The responses of three kiwifruit cultivars, Actinidia chinensis ‘Sanuki Gold’, A. chinensis ‘Rainbow Red’, and A. deliciosa ‘Hayward’ to various storage temperatures (0, 5, 10, 15 and 20°C) for 8 weeks were investigated. The rate of fruit which initiated ethylene production due to rot development increased with increases in storage temperature. Early-maturing cultivars, ‘Rainbow Red’ and ‘Sanuki Gold’ fruit stored at 5, 10, and 15°C showed drastic softening and a decrease in titratable acidity (TA) to an edible level within 4 weeks without detectable ethylene production, whereas fruit stored at 0 and 20°C maintained high firmness and TA even after 8 weeks unless they were infected with rot. A late-maturing cultivar, ‘Hayward’ fruit stored at 5 and 10°C softened more rapidly than when stored at 0, 15, or 20°C. Treatment with 1-Methylcyclopropene (1-MCP) did not suppress the low temperature modulated fruit ripening in any cultivars, indicating its independence from ethylene. These results suggest that ‘Sanuki Gold’ and ‘Rainbow Red’ are more sensitive to low temperatures compared to ‘Hayward’ and the sensitivity is involved in the determination of storage life and how early the fruit matures on the vine.

Paramylon is a β-1,3-D-glucan isolated from Euglena gracilis Z. This study was designed to evaluate the suppressive effects of the oral administration of paramylon on the development of atopic dermatitis (AD)-like skin lesions induced by repeated application of 2,4,6-trinitrochlorobenzene (TNCB) in sensitized NC/Nga mice. The effects of paramylon were assessed by measuring macroscopical and histopathological findings of skin, ear swelling, serum levels of total IgE, interleukin-4 (IL-4) and interferon-γ (IFN-γ) and IL-18 and IL-12 contents in the skin lesions. Oral administration of paramylon inhibited the development of AD-like skin lesions as exemplified by a significant decrease in dermatitis scores for the back, ear swelling and hypertrophy of the skin, infiltration of inflammatory cells in the skin, and serum IgE levels. Oral administration of paramylon reduced serum levels of both IL-4 and IFN-γ and IL-18 and IL-12 contents in the skin lesions. Oral administration of paramylon did not cause weight loss, as was observed with prednisolone. These results suggest that paramylon inhibits the development of AD-like skin lesions in NC/Nga mice by suppressing both the T-helper (Th) 1 and Th 2 cell responses. Our results indicate that paramylon treatment could provide an effective alternative therapy for the management of AD.

Floral induction by grafting without vernalization treatment (NV grafting method) has potential to shorten breeding times and to diversify the seed production of cabbage, an important leafy vegetable with a long and absolute low temperature exposure requirement for its floral induction. However, it is unknown whether the NV grafting method can be actually used for cabbage breeding and seed production. This is because the NV grafting method’s effect on the field performance of obtained progenies has not been investigated, as opposed to the conventional floral induction method by vernalization treatment. Therefore, in this study we compared the effects of two different floral induction methods on the agricultural traits of the obtained progenies. Two clonal lines of ‘Watanabe-seiko No.1’ cabbage were used for the experiment. In the two-year field experiment, we observed a consistent effect of clonal lines on vegetative growth; however, almost no effects of the floral induction methods on either vegetative or reproductive growth were observed. This was further supported by similar expression levels of FLOWERING LOCUS C homologs in the progenies at the young seedling stage. Pollen production and seed formation of the progenies were confirmed regardless of the floral induction method. In conclusion, cabbage seeds obtained by the NV grafting method are likely to show the same traits as those obtained by the conventional vernalization method. This indicates the direct applicability of this method to cabbage breeding and seed production.

In this study, two temperature-induced lipocalin genes SlTIL1 and SlTIL2, and a chloroplastic lipocalin gene SlCHL were isolated from ‘Micro-Tom’ tomato. The coding sequences of SlTIL1, SlTIL2 and SlCHL were 558, 558, and 1002 bp, respectively. By TargetP analysis, no characteristic transit peptides were predicted in the proteins of SlTIL1 and SlTIL2, while a chloroplastic transit peptide was predicted in the protein of SlCHL. The subcellular localization results indicated that SlTIL1 and SlTIL2 proteins were major localized in the plasma membrane, while SlCHL was localized in chloroplast. To understand the function of lipocalins, transgenic tomato over-expressed SlTIL1, SlTIL2 and SlCHL and their virus-induced gene silencing (VIGS) plants were generated. The phenotypes were significantly affected when the SlTIL1, SlTIL2 and SlCHL were over-expressed or silenced by VIGS, which suggested that the three lipocalins played important roles in regulating the growth and development of tomato. In addition, the level of ROS (O2− and H2O2) was low in SlTIL1, SlTIL2 and SlCHL over-expressed plants, while it was high in their silenced plants. The changes in the expression of SODs were consistent with the accumulations of ROS, which indicated that lipocalins might have an important role in abiotic oxidative stress tolerance in tomato plants. Especially SlTIL1 and SlTIL2 are localized around their membranes and protect them from ROS. The results will contribute to elucidating the functions of lipocalin in plants, and provide new strategies to improve the tolerance to abiotic stress in tomato plants.

Split pit in peach (Prunus persica (L.) Batsch) fruit is an internal disorder that can have adverse effects on fruit quality and shelf stability. As it is difficult to distinguish split-pit fruit from normal fruit by appearance, a nondestructive detection method is desirable to determine accurately the timing of split-pit occurrence and remove unwanted fruit from trees. In our recent study, we used an acoustic vibration method for the nondestructive detection of split pit in harvested peach fruit. Here, we demonstrate that this method can be used to detect split pit in unpicked peach fruit. Time-course monitoring of growing fruit revealed that the ratio of the third (f3) to the second (f2) resonant frequency (f3/f2), which is an indicator of split pit in harvested fruit, abruptly increased in unpicked split-pit fruit from early June to early July. In contrast, the f3/f2 values of normal fruit remained low until harvest in late July. The increase in f3/f2 values of split-pit fruit was observed in early June (stage II of fruit growth) and/or from late June to early July (stage III of fruit growth), suggesting that, at least under the experimental conditions in the current study, pit splitting occurred at these two different timings. Split-pit fruit detection rate at harvest increased towards the end of the second split-pit occurrence and split pit could be predicted with high accuracy thereafter. These results collectively suggested that the timing of split-pit occurrence in unpicked peach fruit could be predicted accurately by measuring f3/f2 values and unwanted fruit with split pit could be distinguished from normal fruit on trees. We discuss the possible applications of the nondestructive acoustic vibration method in combination with fruit thinning and the future use of this method in research aiming to develop effective prevention methods or resistant cultivars with reduced split pit.

‘Rainbow Red’ kiwifruit have been reported to gradually ripen during low temperature storage and on the vine in the absence of detectable ethylene. This study was conducted to compare the expression of ripening-related genes during storage at different temperatures and on the vine. Fruit at 5°C and 10°C ripened faster to eating quality within four weeks accompanied with increased expression of ripening-related genes: AcACO3, AcXET2, AcEXP1, AcPG, AcPMEi, AcSUS, AcβAMY1, AcβAMY2, AcGA2ox2, AcNAC3, AcNAC4, and AcMADS2. Fruit at 15°C required a longer period of eight weeks to attain eating quality in concurrence with delayed accumulation of the ripening-related genes. Fruit at 22°C ripened at the slowest rate and did not attain eating quality even after eight weeks, with very minimal accumulation of ripening-related genes. On-vine ripening occurred slowly at the early stages when the average field temperature was ~20°C, but the rate increased as the temperature dropped to ≤15°C accompanied by increased expression of ripening-related genes. These results indicate that both ripening on-vine and during low temperature storage are modulated by low temperature independent of ethylene.

Split-pit in peach fruit is a problematic disorder. Split-pit fruit cannot be detected based on external appearance, and contamination of fruit by split-pit reduces its reliability in the marketplace. Here, we demonstrate that split-pit fruit can be identified by a nondestructive acoustic vibration method and a unique approach based on the ratio of the third (f3) to the second (f2) resonant frequency. The response-resonant frequency spectra showed that the peaks of f2 frequencies in split-pit fruit were shifted to much lower values than those in normal fruit, whereas those of f3 frequencies showed only small shifts. The calculated f3/f2 ratios in most normal fruit were in the range of 1.35–1.4, whereas those in split-pit fruit were 1.45–2.0. Analysis of more than 300 fruit samples revealed that by setting the f3/f2 cut-off value at >1.45, 95% of split-pit fruit in the fruit samples were detected, whereas only 1.5% of normal fruit were missorted as split-pit fruit. A model for simulating the vibration properties of peach fruit was developed by using the finite element method. The simulated vibration patterns showed that f3/f2 values were increased by the insertion of split pit, indicating that, at least partially, the observed high f3/f2 values in split-pit fruit directly reflected split-pit occurrence. These results clearly demonstrate that the use of f3/f2 ratios obtained using an acoustic vibration method can effectively detect fruit with split-pit. The possibility of installing acoustic vibration devices in peach sorting lines and the application of portable devices to unpicked fruit on the tree are discussed.
