The purpose of this research was to compare the accuracy of newly described P wave-related parameters (P wave area, Macrux index and mean electrical axis) with classical P wave-related parameters (voltage and duration of P wave) for the assessment of left atrial (LA) size in dogs with degenerative mitral valve disease. One hundred forty-six dogs (37 healthy control dogs and 109 dogs with degenerative mitral valve disease) were prospectively studied. Two-dimensional echocardiography examinations and a 6-lead ECG were performed prospectively in all dogs. Echocardiography parameters, including determination of the ratios LA diameter/aortic root diameter and LA area/aortic root area, were compared to P wave-related parameters: P wave area, Macrux index, mean electrical axis voltage and duration of P wave. The results showed that P wave-related parameters (classical and newly described) had low sensitivity (range=52.3% to 77%; median=60%) and low to moderate specificity (range=47.2% to 82.5%; median 56.3%) for the prediction of left atrial enlargement. The areas under the curve of P wave-related parameters were moderate to low due to poor sensitivity. In conclusion, newly P wave-related parameters do not increase the diagnostic capacity of ECG as a predictor of left atrial enlargement in dogs with degenerative mitral valve disease.

The objective of this study was to collect epidemiological data on neoplasms in pet ferrets in Japan. A questionnaire to collect information was made available to Japanese veterinary practitioners through the web site of the Japanese Society of Exotic Pet Medicine. Completed questionnaires were returned from 29 practices, and 945 neoplasms met the criteria for inclusion in the study. Neoplasms were found in every organ system except the respiratory system; the endocrine (418; 44.2%), integumentary (196; 20.7%) and hemolymphatic (184; 19.5%) systems were most commonly affected. The most common tumor types were pancreatic islet cell tumor (211: 22.3%), adrenal gland tumor (207; 21.9%) and lymphoma (152; 16.1%). The age of the affected ferrets ranged from less than 3 months to more than 7 years of age. Tumor incidence was highest in ferrets between 4 and 6 years of age. No sex predilection was found. These results were similar to those recently published in North America. Most Japanese pet ferrets are imported from North America, and their husbandry including diets is similar to that in North America, which may explain the similar tendencies in the incidence of neoplasms in this study and those of findings in North America.

A 7-year-old castrated male ferret developed unilateral cervical lymphadenomegaly over a 1-month period. Histological examination revealed proliferation of tumor cells in a diffuse and partially nodular pattern. The tumor cells were predominantly Hodgkin cells and binucleated Reed-Sternberg cells, characterized by abundant, clear, vacuolated cytoplasm, pleomorphic, ovoid nuclei with thick nuclear membranes and distinct nucleoli. Multinucleated cells, resembling lymphocytic and histiocytic (L&H) cells, were also observed. Immunohistochemically, the tumor cells expressed Pax-5, BLA-36 and vimentin. A small population of the tumor cells expressed CD20. This case showed proliferation of Hodgkin/Reed-Sternberg cells in conjunction with L&H cells that were histologically analogous to feline Hodgkin’s-like lymphoma. However, Pax-5 and BLA-36 expression along with rare CD20 expression were consistent with classical Hodgkin’s lymphoma in humans.

Canine anaplasmosis is regarded as an infection by Anaplasma platys rather than zoonotic Anaplasma phagocytophilum in subtropical areas based on the assumption that the common dog tick species is Rhipicephalus sanguineus, which transmits E. canis and presumably A. platys. We investigated asymptomatic dogs and dog ticks from 16 communities in Nantou County, Taiwan to identify common dog tick species and to determine the prevalence of Anaplasma and Ehrlichia spp. Of total 175 canine blood samples and 315 ticks, including 306 R. sanguineus and 9 Haemaphysalis hystricis, 15 dogs and 3 R. sanguineus ticks were positive for E. canis, while 47 dogs and 71 R. sanguineus ticks were positive for A. platys, via nested PCR for 16S rDNA and DNA sequencing of selected positive amplicons. However, among the dogs and ticks that were positive to A. platys 16S rDNA, only 20 dogs and 11 ticks were positive to nested PCR for A. platys groEL gene. These results revealed the importance of searching for novel Anaplasma spp. closely related to A. platys in dogs and ticks. Seropositivity to a commercial immunochromatographic test SNAP 4Dx Anaplasma sp. was not significantly associated with PCR positivity for A. platys but with infestation by ticks carrying A. platys (P<0.05). Accordingly, R. sanguineus may be involved in transmission of A. platys but may not act as a reservoir of E. canis and PCR results for 16S rDNA could be a problematic diagnostic index for A. platys infection.

This study evaluated the accuracy of a newly developed veterinary portable blood glucose meter (PBGM) with hematocrit correction in dogs and cats. Sixty-one dogs and 31 cats were used for the current study. Blood samples were obtained from each dog and cat one to six times. Acceptable results were obtained in error grid analysis between PBGM and reference method values (glucose oxidation methods) in both dogs and cats. Bland–Altman plot analysis revealed a mean difference between the PBGM value and reference method value of −1.975 mg/dl (bias) in dogs and 1.339 mg/dl (bias) in cats. Hematocrit values did not affect the results of the veterinary PBGM. Therefore, this veterinary PBGM is clinically useful in dogs and cats.

<p>A 4-year-old female Siamese crocodile (<i>Crocodylus siamensis</i>) housed at a zoo died without any prior clinical signs. During necropsy, numerous scattered, well-demarcated, yellowish-white, firm nodules were observed throughout the liver and lungs. Microscopic examination with periodic acid-Schiff staining revealed granulomatous inflammation in the liver and lungs. Liver granulomas were characterized by the presence of a connective tissue barrier and hyphae, and the centers of the granulomas showed signs of necrosis. Lung samples showed characteristics similar to those observed in the liver samples. The fungus was identified as <i>Aspergillus fumigatus</i> based on its appearance on Sabouraud dextrose agar, microscopic examination with lactophenol cotton blue staining and genetic sequencing. Therefore, zoo veterinarians should pay close attention to fungal infections in captive animals.</p>

In this study, we attempted to clarify whether puppy socialization and command training class, which was limited to puppies approximately 4 months of age, prevented behavior problems in dogs. We evaluated the behaviors of 142 dogs with 4 types of training experience by using a behavior test and the Canine Behavior Assessment and Research Questionnaire (C-BARQ). Dogs in the puppy class (PC) group (n=44) attended the class for 1 hr each week for 6 weeks, dogs in the puppy party (PP) group (n=39) attended a 1-hr “puppy party,” dogs in the adult class (AC) group (n=27) undertook basic obedience lessons for adolescent/adult dogs for 1 hr each week for 6 weeks, and dogs in the no class (NC) group (n=32) underwent no formal training. The behavior test evaluated each dog’s response to commands, owner’s recall, separation, a novel stimulus and strangers. The C-BARQ evaluated 15 canine behavioral factors. The behavior test results indicated that the PC and AC groups showed significantly higher response to commands than the PP or NC group. Thus, participation in puppy and adult classes improved the obedience behavior of dogs, regardless of age. Positive response to strangers in the PC group was significantly higher than that in the AC and NC groups and tended to be higher than that in the PP group. Therefore, PC may help prevent canine behavioral problems such as disobedience or fear of strangers.

<i>Escherichia coli</i>(<i>E. coli</i>) isolated from the uterus of a Thoroughbred mare with bacterial endometritis was used to evaluate the effect of progesterone (P<sub>4</sub>) on the immune response of mares. Peripheral blood mononuclear cells (PBMCs) were collected from 10 nonpregnant clinically healthy adult mares (range, 4-12 years) during diestrus, four Thoroughbreds and six Hokkaido native horses. Cell proliferation and expression of cytokine mRNA, including interferon (IFN)-γ, tumor necrosis factor (TNF)-α and interleukin (IL)-10, of PBMCs stimulated with <i>E. coli</i> and P<sub>4</sub> were examined <i>in vitro</i>. P<sub>4</sub> was shown to have significantly inhibited <i>E. coli</i> induced proliferation and expression of IFN-γ in PBMCs. These results indicate that P<sub>4</sub> inhibits the immune response to <i>E. coli</i> in mares.

A four-year-old dachshund presented with a two-week history of pyrexia, depression, and cough. Four months earlier, the owner observed the dog swallow a whole popsicle stick, but the animal showed no clinical signs at that time. Radiography, ultrasonography, and computed tomography confirmed an intrathoracic linear foreign body and pleural effusion in the right thorax. The pleural fluid was bloody and purulent, and contained inflammatory cells and Escherichia coli. The dog was diagnosed with pyothorax induced by a foreign body, and was treated successfully by surgical removal of the foreign body, partial lung lobectomy, thoracic lavage, and antibiotics. The foreign body was identified as a popsicle stick that the dog had eaten.

シマハイエナ (Hyena hyena) の膵臓の葉区分を肉眼解剖学的に観察し, 膵管と副腎管の走行を検討した. また, 組織学的に外分泌部と膵島の配置を確認し, 免疫組織化学的手法により, 膵島におけるA, B, D, およびPP細胞の分布状態を検討した. 膵臓は胃の大弯付近から十二指腸近傍にかけての間膜に発達していた. 幽門部を境界に鋭く折れるため, 前半部を左葉, 後半部を右葉と判断することができた. 右葉よりさらに後方に, 特徴的な独立した小さな葉が確認され, これを後葉 (caudal lobe) と名付けた. 導管は合計3本確認され, 大十二指腸乳頭近傍に到達するものと, そこからさらに前方に分岐するものを膵管と推定し, 後葉から十二指腸に至る最後部の管を副膵管と定めた. 組織学的には多数の膵島が外分泌部の間に観察された. A細胞およびPP細胞は膵島の辺縁部に限局し, B細胞とD細胞は, 膵島内に偏りなく分布していた. また, 外分泌部に単独で散在するB細胞が観察された. ハイエナ類の膵臓の形態はこれまでに記載されたことがない. 肉眼的には後葉の存在と膵管の分岐が特記された. 組織学的には, B細胞の膵島での均等な分布と外分泌部での散在が, シマハイエナの特徴であるといえる. これらの結果は, 食肉類の中で独特の進化を遂げたハイエナ科における膵臓の形態学的データとして, 今後の比較検討にも用いることができよう.

Wilms’ tumor 1 (WT1) expression has been investigated in various human cancers as a target molecule for cancer immunotherapy. However, few studies have focused on WT1 expression in dogs. Firstly, cDNA of canine WT1 (cWT1) was molecularly cloned from normal canine kidney. The cross-reactivity of the anti-human WT1 monoclonal antibody (6F-H2) with cWT1 was confirmed via Western blotting using cells overexpressing cWT1. Immunohistochemical staining revealed that cWT1 expression was detected in all canine lymphoma tissues and in some normal canine tissues, including the kidney and lymph node. cWT1 is a potential immunotherapy target against canine cancers.

Renal Fanconi syndrome has recently been associated with the ingestion of pet jerky treats from China in mostly small breed dogs in North America, Australia and Europe. We report here about two dogs with Fanconi syndrome following pet jerky treats exposure in Japan. A mixed-breed dog and a French bulldog showed weight loss, polyuria and polydipsia. For years, the owners had been feeding large quantities of pet jerky treats containing chicken prepared in China. Diagnostics revealed glycosuria without hyperglycemia, severe aminoaciduria, and in one case also ketonuria, hypokalemia and metabolic acidosis. A diagnosis of Fanconi syndrome associated with long-term consumption of Chinese pet jerky treats was made. Both dogs recovered fully following withdrawal of the pet jerky treats and supportive care. Fanconi syndrome of dogs in association with the consumption of pet jerky treats of Chinese origin can cause a broad proximal tubular defect with glycosuria and generalized amino aciduria, and should be also considered in Asia. Jerky treats associated Fanconi syndrome can be completely reversible following withdrawal of the treats and supportive care to correct the metabolic abnormalities.

We examined whether multidrug resistant (MDR) canine lymphoma increases gene expression for platelet-derived growth factor receptor α (PDGFRα), vascular endothelial growth factor receptor 2 (VEGFR2), and c-KIT, and whether toceranib phosphate (TOC) has potential as a treatment for MDR canine lymphoma. The clinical data showed that PDGFRα expression was higher in canine subjects with MDR T-cell lymphoma than in those with untreated T-cell lymphoma, and that c-KIT expression was greater in subjects with T-cell lymphoma than in those with B-cell lymphoma. TOC monotherapy was well tolerated without serious adverse effects, and two of the five subjects that received TOC exhibited partial responses. The data presented here could contribute to the assessment of TOC-based therapy for dogs with MDR or T-cell lymphoma.

Cancer stem cells or tumor-initiating cells (TICs) are a small subpopulation of cells that have the capacity to self-renew, differentiate and initiate tumors. These cells may function in tumor initiation, aggression and recurrence. Whether spheres derived from canine rhabdomyosarcoma cells have stem cell-like properties is unclear. We induced sphere formation in the canine rhabdomyosarcoma cell lines, CMS-C and CMS-J, and characterized the spheres in vitro and in vivo. Sphere-forming cells were more resistant to vincristine, mitoxantrone and doxorubicin than adherent cells. Xenograft transplantation demonstrated that 1 × 103 sphere-forming cells derived from CMS-C were sufficient for tumor formation. The sphere assay showed that the sphere-forming cells were present in these tumors. These results suggest that the spheres derived from canine rhabdomyosarcoma cells may possess characteristics of TICs. This study provides the foundation for elucidating the contribution of TICs to rhabdomyosarcoma tumorigenesis.

Cytotoxic T lymphocyte associated gene-4 (CTLA-4) is a costimulatory molecule, expressed on the surface of activated T cells that negatively regulates T cell activation. In humans, alternative splicing of the CTLA-4 gene generates two major isoforms of mRNA, and a soluble form of CTLA-4 (sCTLA-4) was detected in normal human serum. We describe alternatively spliced mRNA expressed in peripheral blood mononuclear cells obtained from a healthy dog lacking the transmembrane domain coded by exon 3 of the CTLA-4 gene. Immunoprecipitation and western blotting of dog serum revealed a band of approximately 23-kDa, which is consistent with the predicted size, based on the amino acid sequence of the canine sCTLA-4 obtained in this study.

The feline F12 gene was examined to identify a mutation associated with coagulation factor XII (FXII) deficiency in a litter of 6 cats, including 2 cats with severely reduced FXII activity (7.1 and 9.3%, respectively) and 4 cats with moderately reduced FXII activity (range 36.0 to 46.3%). Cats with severely reduced FXII activity were homozygous for a G to C missense mutation in exon 13 of the F12 gene, resulting in an amino acid change (p.G544A). Cats with moderately reduced FXII activity were heterozygous for this mutation. Expression studies revealed reduced secretion of p.G544A mutant FXII protein from transfected HEK293 cells compared with wild type FXII. These results reveal a novel F12 mutation in FXII deficient cats and define the underlying mechanism for low FXII activity in homozygotes.

The goals of this retrospective clinical case series study were to describe the management of anesthesia, and to report perioperative complications in cats undergoing subcutaneous ureteral bypass (SUB) placement due to ureteral obstruction. Medical records of client-owned cats with ureteral obstruction and anesthetized for SUB placement between 2012 and 2015 in a veterinary teaching hospital were reviewed. Twenty-seven cases were identified. Duration of anesthesia and surgery (mean ± standard deviation) were 215 ± 42 min and 148 ± 36 min, respectively. Hypothermia was the most common intraoperative complication. Hypotension, hypocapnia, hypertension and bradycardia were also frequently observed. Out of 22 cats who experienced intraoperative hypotension, 17 received inotropes and vasopressors. There was a significant decrease in creatinine (P=0.008) and total solids (P=0.007) after SUB placement when compared with baseline values. Postoperative complications included pain, anorexia, nausea, hypertension, and urinary tract-related problems. No death occurred in the postoperative period. Risk factors could not be identified. Successful management of anesthesia for SUB placement involves rigorous anesthetic monitoring and immediate treatment of complications. Perioperative complications appear to be common. This study could not identify risk factors associated with this procedure.

In this study, we attempted to isolate Escherichia coli from healthy adults in Ho Chi Minh City, Vietnam, and characterized its antimicrobial resistance profile, extended-spectrum β-lactamase (ESBL) genotype, phylogenetic grouping and virulence gene profile. A total of 103 E. coli isolates were obtained, and most of them were antimicrobial resistant such to streptomycin (80.6%), tetracycline (67.0%), ampicillin (65.0%), sulfamethoxsazole/trimethoprim (48.5%), nalidixic acid (43.7%), chloramphenicol (34.0%), cefotaxime (15.5%), ciprofloxacin (15.5%), kanamycin (12.6%), ceftazidime (10.7%), fosfomycin (4.9%) and gentamicin (2.9%). However, all these E. coli strains were susceptible to imipenem. Surprisingly, of 103 strains, 74 (71.8%) and 43 (41.7%) strains showed resistance to more than 3 and 5 classes of antimicrobials, respectively. Furthermore, 10 E. coli strains were ESBL-producers and positive for blaCTX-M genes (7 for blaCTX-M-9 and 3 for blaCTX-M-1), while five were additionally positive for blaTEM genes. S1-nuclease pulsed-field gel electrophoresis analysis revealed that 7 and 3 strains of E. coli carry blaCTX-M genes on their large plasmid and chromosome, respectively. Phylogenetic analysis exhibited that majority of the E. coli strains was grouped into A (44.7%), followed by B1 (23.3%), B2 (18.4%) and D (13.6%). Virulence genes associated with diarrheagenic E. coli, such as astA, EAF, eaeA, elt and eagg were also detected in ESBL-producing E. coli as well as antimicrobial resistant strains. These data suggest that commensal E. coli of healthy human could be a reservoir for antimicrobial resistance determinants and some of them might be harmful to human.

7歳雄のヘラジカ(Cervus elaphus nelsoni)が体重減少,削痩,過度な流涎,歯軋り,発熱,食欲不振および呼吸困難の症状を3週間呈した後に,安楽殺され,剖検された.このヘラジカは1997年3月9日にカナダから韓国に輸入された.肉眼病変は瀰漫性の線維素性肺炎であった.組織学的には軽度な神経細胞の空胞変性と限局した脳幹部神経核の神経網の海面状変性と広範な星状膠細胞の増生が見られた.プロテアーゼ抵抗性プリオン蛋白質(PrP^<res>)に対する免疫組織化学では全ての脳組織切片で陽性であったが,延髄の閂の切片で最も顕著であった.PrP^<res>は脳および脊髄のウエスタンブロット法でも検出された.本ヘラジカと接触のあった残りのヘラジカおよびシカは淘汰され,慢性消耗性疾患は陰性であった.著者らの知る限りでは,本症例は米国およびカナダ以外の国での最初の症例であった.
