Bacillus thuringiensis has been utilized worldwide as an environmentally benign biopesticide. The quality control of formulations from B. thuringiensis are usually performed based on bioassays with insects, although the test methods and the species of test insects differ among countries. In Japan, a bioassay method with silkworm, Bombyx mori, was proposed by the Japan Plant Protection Association in 1973, which illustrated a protocol of procedure and a reference sample to calculate the relative potency of the test sample by Bombyx Unit. This bioassay method has been used without modification since that time for the quality control of all B. thuringiensis formulations commercially produced in this country. However, in order to simplify and speed up the bioassay method some modifications are called for. The authors review some problems with the current method and discuss possible modifications to resolve these issues and improve the assay.

The location and preference of adult Asian citrus psyllid, Diaphorina citri Kuwayama, a vector of the citrus huanglongbing (greening disease) Liberobacter asiaticum, on two host plants, Murraya exotica L. and Citrus depressa Hayata, were investigated in Okinawa Prefecture, Japan. While the adults of D. citri were found on all parts of the C. depressa tree and on the M. exotica tree, all of the D. citri eggs and most of the nymphs and adults were found on young shoots and young leaves. We suggest that it is possible to save labor in the investigation and control of the psyllid by attentively checking the young shoots of M. exotica. Investigation on the distribution of D. citri adults on M. exotica plantlets hung upside down in a container showed that although no difference was found between frequencies of males on young leaves and mature leaves, significantly more females were found on the apical buds, suggesting that females have a preference for this part of the tree.

The occurrence of Psylla evodiae Miyatake, which has been known to occur on Euodia meliifolia (Rutaceae) in the Yaeyama Islands of Japan and in Taiwan, has been newly recorded in Kyushu (Fukuoka, Nagasaki, and Kagoshima prefectures) and on Yakushima Island and Okinawa Island in southwestern Japan. In addition, it has been confirmed that P. evodiae feeds on Murraya paniculata (Rutaceae), which is a newly recorded host plant in southern Kyushu (Kagoshima Prefecture) and Yakushima Island. It has also been confirmed that P. evodiae feeds on Zanthoxylum beecheyanum var. alatum (Rutaceae) in Okinawa Island.

Triops longicaudatus, a small aquatic crustacean inhabiting the paddy fields of Japan, has eggs that can undergo diapause and survive under dry conditions for several decades. In this study, diapause eggs were embedded in resin and thin sections were observed under light microscopy. Observations revealed that there were abundant yolk granules, or many nuclei, in a diapause egg. Diapause eggs, post-diapause eggs and eggs just after oviposition were homogenized in their groups and the residual material was removed by centrifugation. The samples were analyzed for proteins using two-dimensional gel electrophoresis (2D-GE). The protein spots were visualized by silver staining, and revealed that there were more than 1,400 spots in a diapause egg, and two protein spots (26 kDa, 4.7 isoelectric point) appeared specifically in diapause eggs. It is suggested that these proteins have some relationship with long diapause. The proteins differ from a small heat-shock/α-crystallinprotein (p26) of Artemia franciscana.

The leaf beetle Ophraella communa is an oligophagous insect that infests several asteraceous plants, especially ragweed Ambrosia artemisiifolia, in fields of Japan. The feeding responses of O. communa to methanolic extracts of 11 genera and 17 species of asteraceous plants were examined to determine whether feeding preference depended on chemicals in the plants. The methanolic extracts of 8 asteraceous species stimulated feeding in O. communa, but the other 9 species showed no activity. Two triterpenoids (α-amyrin acetate and β-amyrin acetate) and two caffeic acid derivatives (chlorogenic acid and 3,5-dicaffeoylquinic acid) are known as feeding stimulants for O. communa. The distribution of these compounds was analyzed in 17 asteraceous species. These compounds were found in many species, including nonhost plants of this beetle, however amounts and compositions differed. Even when the methanolic extracts of A. artemisiifolia were added to extracts of some species which did not elicit feeding of O. communa, the feeding of O. communa was not induced by the inactive plants. Feeding deterrents as well as feeding stimulants in the leaves of asteraceous plants appear to determine the feeding preference of O. communa.
