The present paper is a brief review of our recent works on the crystal structures, structural disorders and ion-diffusion paths of various ionic and mixed conductors. Diffusion paths along the <100> directions are observed in the fluorite-type ionic conductors such as δ-Bi1.4Yb0.6O3, Ce0.93Y0.07O1.96, Y0.785Ta0.215O1.715 and α-CuI. In the prerovskite-type and double-prerovskite-type conductors, the mobile oxide ions move along the <100> directions near the stable positions and along <110> around the midway of diffusion pathway. The cubic prerovskite-type and double-prerovskite-type conductors exhibit three- and two-dimensional networks of diffusion pathways, respectively. K2NiF4-type conductor has a two-dimensional network of diffusion pathway through interstitial oxide ions. The oxide ions in apatite-type conductors diffuse along the hexagonal c-axis. The diffusion paths of two lithium-ion conductors are also presented.

Selenocysteine (Sec) is the 21st amino acid that is incorporated into proteins translationally. The selenocysteine tRNA (tRNASec) is first ligated with serine by seryl-tRNA synthetase, and the Ser moiety is converted to Sec in a tRNA-dependent manner. In Bacteria, the selenocysteine synthase SelA converts Ser to Sec directly, whereas in Eukaryotes and Archaea, phosphorylation of the Ser moiety is required prior to the Sec conversion. In this review, we describe the crystal structures of SelA, bacterial tRNASec, and the SelA•tRNASec complex. Based on the crystal structures and mutational analyses, the substrate discrimination and reaction mechanisms are discussed.

PLANET is a powder neutron diffractometer dedicated to high-pressure experiments. By combining the intense neutron source of J-PARC and the high-pressure devices designed for time-of-flight powder neutron diffraction, precise structure analysis of crystal, liquid, and amorphous solids is possible over wide pressure and temperature range of 0~20 GPa and 77~2,000 K. This beamline is effective for various studies in geophysics, planetary science, physics and chemistry. This paper overviews the beamline and introduces recent results obtained at PLANET.

There are many huge macromolecular complexes in living organisms. They are often hard to crystallize because of their size, complexity and heterogeneity. Cryo-electron microscopy (cryo-EM) is a suitable method to analyze the structures of such biological macromolecules, because it can be applied to various forms of samples, e.g. two-dimensional crystal, helical assembly, spherical virus, dispersed particle, cell organelle and cell, although attainable resolution depends on the system. In this review, I introduce these techniques and examples of the structure analysis, and briefly review the perspective of cryo-EM.

Fundamentals of powder diffractometry are presented, which are 1) the angle dispersive method, 2) problems in the measurements by the X-ray powder diffractometer, 3) errors from particle size and corrections for the preferred orientation, 4) energy dispersive method and 5) corrections for the anomalous dispersion.

In-situ synchrotron diffraction experiments were conducted in order to clarify the formation process of α'-martensite from the γ-phase induced by external strain, combined with Lorentz transmission electron microscopy (TEM) and high-resolution TEM observations. Lorentz TEM observation revealed that α'-martensite exist near the defect structures such as dislocations and stacking faults in the parent γ-phase. In addition, it is clearly demonstrated that ε-martensite with hexagonal symmetry appears as an intermediate phase during the plastic deformation of SUS304 stainless steel. Our experimental results suggested that the interfaces between twin structures of the γ-phase presumably play a crucial role in the formation of ε-martensite.

Protein crystallography has been leading the structural biology field by solving molecular structures at atomic resolution. However, it has been limited in the integration of structural information of macromolecules with a dynamic nature, which can adopt various conformations in time scales of motion even in large complexes. Therefore, it is critical to use multiple methodologies to understand biological phenomena, and connect the interfaces of molecules at both molecular and atomic resolution in macromolecular assembly. “Correlated structural analysis” is a concept in structural life science to fill the gaps between structural biology techniques. In Japan, the Platform for Drug Discovery, Informatics, and Structural Life Science has been launched with diverse expertise in bioinformatics, sample production, and structure determination. The platform supports not only conventional structural studies via X-ray crystallography, but also correlated structural analysis by integrating protein crystallography, small angle X-ray scattering, NMR, and electron microscopy data.
