For the structure determination of endohedral metallofullerenes (EMFs), X-ray crystallographic study is very powerful because it provides detailed information of the positions of encapsulated species as well as the cage structures. We have reported a number of X-ray crystal structures of EMFs and their derivatives, which are in good agreement with those suggested by theoretical calculations as well as NMR analyses. Herein we show the importance of interplay between experiments and theoretical calculations for the structure determination of EMFs by taking up La@C82, Gd@C82, and Sc2@C66 as the representative examples.

Hydrogen is known as an ultimate clean energy source and is thus discussed as a future sustainable energy carrier. Hydrogenases catalyze the reversible oxidation of the molecular hydrogen. We report the crystal structure analysis of [NiFe] hydrogenase from sulfate reducer at subatomic resolution. The structure reveals that the hydride bridge between nickel and iron at the active site and the possible proton bound site at the cysteine residue, resulting from the initial heterolytic splitting of dihydrogen by the enzyme. This finally clarifies the initial step in the mechanism of hydrogen conversion.

The atomic structures of liquid-phase bonded heterointerfaces between Al alloy and AlN single crystal was examined using aberration-corrected scanning transmission electron microscopy combined with high-sensitive energy-dispersive X-ray microanalysis. The formation of O-Mg-O monolayer structure was found at the interface, facilitating the bonding between the two dissimilar crystals during liquid-phase bonding processes. Understanding the spontaneous formation of such layered transition structures at the heterointerfaces will be a key for fabricating very stable liquid bonded Al alloy / AlN heterointerfaces.

We report enantiopure crystal growth and chiral helimagnetic ordering in chiral inorganic compounds CsCuCl3 with a chiral space group of right-handed P6122 or left-handed P6522. Our 2-step crystallization process makes it possible to grow centimeter sized enantiopure single crystals. Polarized neutron diffraction experiments probed that crystallographic chirality of CsCuCl3 strongly combined with helimagnetic chirality via Dzyalloshinskii-Moriya interaction.

We have prepared various ionic liquids containing cationic organometallic complexes, which exhibit intriguing physical properties and chemical reactivities. This review describes molecular design, preparation, and crystal structures of organometallic ionic liquids and related materials. Two topics are discussed, in which X-ray crystallography played an important role: magnetic-field effects on the crystallization of ferrocenium ionic liquids, and odd-even effects on the melting phenomenon of cobaltocenium ionic liquids.

Future of structural life science will continue to rely on crystallography using X-ray and electron beams. Multi-modal, multi spatio-temporal scale imaging will be the key to understanding complex biological phenomena at both fundamental biology and translational biomedical or industrial applications. This transformation is influencing the way structural and functional studies are carried out around the globe, and large insfrastructure facilities and institutions are developing new modus operandi to enable biology and medical communities accessing vast and diverse range of technologies, including X-ray, electron, photon, and neutron sources, and AI/ML.

I will describe the future direction of electron crystallography, by focusing on how electron beams contribute uniquely for atomic structure determinations. Both for electron diffraction and microscopy, the key lies on how we make the best use of dynamical scattering effects, which are significant for electrons compared with other probes including X-ray.

Current research in structural biology has advanced our insights much more than we expected 20 years ago. X-ray free-electron lasers and cryo-electron microscopy are technologies that have brought unprecedented achievements to researchers acquiring structural aspects of protein molecules at a higher level. This paper overviews current research in photosynthetic membrane proteins by utilizing these technologies and summarizes structural biology demands now encountering.

TransLesion Synthesis (TLS) is a DNA damage tolerance mechanism that allows continued DNA synthesis, even in the presence of damaged DNA templates. In response to DNA damage, TLS polymerases are recruited to replication forks via interactions with ubiquitinated Proliferating Cell Nuclear Antigen (PCNA) involving PCNA-interacting protein box (PIP-box) and ubiquitin-binding domains (UBDs). We now report the first crystal structures of human PCNA in complex with three TLS polymerase peptides containing the non-canonical PIP-box. TLS polymerases interact with PCNA in different ways, both from one another and from canonical PIP-box peptides. Furthermore, we discuss these TLS polymerases interact with ubiquitinated PCNA.

Crystal structure analysis using scanning transmission electron microscopy(STEM)is briefly reviewed. The various imaging techniques, such as bright field(BF), annular BF(ABF)and annular dark-field(ADF)are presented. Recent progress in STEM imaging is also described.
