With the sequencing of the genomes of various species, attention has turned to the structure, properties, and functional activities of proteins. However, rapid progress in the area of proteomics is premised on the availability of sufficient amounts of a large number of proteins. Here we described a novel cell-free system from wheat embryos for the high-throughput screening/ synthesis of gene products. Our system should open up many possibilities in the post-genome era.

Thermostable direct hemolysin (TDH) is a major virulence factor of Vibrio parahaemolyticus that causes pandemic food-borne enterocolitis mediated by seafood. TDH exists as a tetramer in solution, and it possesses extreme hemolytic activity. Here, we present the crystal structure of the TDH tetramer at 1.5 Å resolution. The TDH tetramer forms a central pore with dimensions of 23 Å in diameter and ∼50 Å in depth. π-cation interactions between protomers comprising the tetramer were indispensable for hemolytic activity of TDH. The N-terminal region was intrinsically disordered outside the pore. Molecular dynamics (MD) simulations suggested that water molecules permeate freely through the central and side channel pores. These findings imply a novel membrane attachment mechanism by a soluble tetrameric pore-forming toxin.

Glutamine : fructose 6-phosphate amidotransferase (GFAT) is a rate-limiting enzyme in the hexoamine biosythetic pathway and plays an important role in type 2 diabetes. We now report the first structures of the isomerase domain of the human GFAT in the presence of cyclic glucose 6-phosphate and linear glucosamine 6-phosphate. The C-terminal tail including the active site displays a rigid conformation, similar to the corresponding Escherichia coli enzyme. The diversity of the CF helix near the active site suggests the helix is a major target for drug design. Our study provides insights into the development of therapeutic drugs for type 2 diabetes.

Long-period structures of homologous phases Ga2O3(ZnO)m are closely related to the wurtzite structure of ZnO, and structural evolution is discussed based on the results from structure analyses. A unified structure model for Ga2O3(ZnO)m is established in(3+1)dimensional superspace. Zig-zag functions with large amplitudes are used as modulation functions in the model, which is a common feature in structures constructed by the so-called unit-cell twining. Advantages of the superspace description for homologous phases are explained in relation to the process of identification of materials.

X-ray crystallography is an important technique for structure-based drug discovery, mainly because it is the only technique that can reveal whether a ligand binds to the target protein as well as where and how it binds. However, ligand screening by X-ray crystallography involves a crystal soaking experiment, which is usually performed manual. Thus, the throughput is not satisfactory for screening large numbers of candidate ligands. In this study, we developed a technique to anchor protein crystals to mounting loops by using gel and inkjet technology; the method allows soaking of the mounted crystals in ligand-containing solution. This new technique may assist in designing a fully automated drug screening pipeline.

SPring-8 Angstrom Compact free electron LAser(SACLA)is the world second X-ray laser which started user operation in 2012. As an introduction of the special issue, here we show how the SACLA project got started. While originated as a tiny R&D program within RIKEN, the SACLA project has been grown to one of the five National Key Technology projects in Japan. SACLA is the first step of the Compact X-ray free electron laser. Further downsizing of the facility is envisaged.

Structural analysis including hydrogen position is important for estimating the reaction mechanism of hydrolases. We applied neutron and X-ray structural analysis to a mysterious β-1,4 glucanase from a fungi Phanerochaete chrysosporium (PcCel45A). The joint analysis of neutron and X-ray structures showed that PcCel45A utilizing an imidic acid form of asparagine residue as a general base instead of typical acidic residues.

X-ray fiber diffraction method was used to deduce the molecular structure of tobacco mosaic virus. Multi- dimensional multiple isomorphous replacement method was applied to overcome the information loss by the cylindrical average in the fiber diffraction pattern, and some other developments in the analysis made it possible to build the molecular model of the whole virus particle. Protein- protein interaction and protein- RNA interaction found in the molecular structure gave us a clear understanding on the mechanism of self assembly.

Locations of water molecules and exchangeable cations in a hydrated natural chabazite with a composition of Ca1.57Na0.49 [Al3.39Si8.55O24] ⋅12.47H [d2] O were successfully determined by single crystal X-ray diffraction. The structure analyses have revealed the presences of five partially occupied water sites (OW1-OW5) and four partially occupied exchangeable-cation sites (Ca1-Ca4), and have showed that fully coordinated exchangeable cations can adopt sixfold or sevenfold coordination. The distances between water sites and framework oxygen sites and between water sites suggest that the hydrogen bonds formed between water molecules and framework oxygen atoms are very weak, whereas strong hydrogen bonds exist between water molecules.

The Rad52 protein and its homologs play critical roles in eukaryotic homologous recombination. In the present study, we have determined the crystal structure of the N-terminal, homologouspairing domain of Rad52. The structure revealed an undecameric ring that has an exposed groove essential for DNA binding. Amino acid residues inside the groove are directly involved in DNA binding as evidenced by mutational analyses.
